The squad investigates the murder of Bobbi O'Rourke, a book author who'd written about her life as a teen prostitute. Carisi finds out the victim was just posing as the writer for public appearances because no one has ever seen the real Bobbi. Fin traces the royalty payments to the home of Joe and Ben Edelman (Judd Hirsch and Wallace Shawn), two bickering, old Jewish men who have lived as hoarders and shut-ins for decades. Benson learns there is no Bobbi; Ben wrote the sexually-explicit book under the pseudonym. But before they let the brothers go, Carisi discovers their dead mother's body in a locked freezer. ADA Peter Stone charges the Edelmans with murder. Based on the book, Olivia believes Ben might be a past sexual assault victim. Rollins learns Joe was also molested, but the brothers have never told each other about their trauma. At trial, Joe takes the blame for smothering their dying mother decades ago - then has a massive heart attack. After three girls disappear overnight in a city park, a large man is spotted returning stab victim Zoe from the woods. The child tells Benson her sister, Mia, and her friend, Perry, were kidnapped by the supernatural Glasgowman. They had a map to the creature's mansion they got from Perry's babysitter, a graphic artist. Amaro and Rollins find the missing girls, who say Glasgowman bound and stabbed them. It becomes clear that Perry is delusional, convinced her friends to go into the woods, then tried to make a blood sacrifice to Glasgowman. Mia is released and Perry is sentenced to a psychiatric hospital. Riding the elevator, Carisi spots the two discreetly intertwine their pinkies. Abstract: 1 Mucolipidosis IV (MLIV) is an ultra-rare, recessively inherited lysosomal disorder resulting from inactivating mutations in MCOLN1, the gene encoding the lysosomal cation channel TRPML1. The disease primarily affects the central nervous system (CNS) and manifests in the first year with cognitive and motor developmental delay, followed by a gradual decline in neurological function across the second decade of life, blindness, and premature death in third or fourth decades. Brain pathology manifestations in MLIV are consistent with hypomyelinating leukodystrophy with brain iron accumulation. Presently, there are no approved or investigational therapies for MLIV, and pathogenic mechanisms remain largely unknown. The MLIV mouse model, Mcoln1-/- mice, recapitulates all major manifestations of the human disease. Here, to better understand the pathological mechanisms in the MLIV brain, we performed cell type specific LC-MS/MS proteomics analysis in the MLIV mouse model and reconstituted molecular signatures of the disease in either freshly isolated populations of neurons, astrocytes, oligodendrocytes, and neural stem cells, or whole tissue cortical homogenates from young adult symptomatic Mcoln1-/- mice. Our analysis confirmed on the molecular level major histopathological hallmarks of MLIV universally present in Mcoln1-/- tissue and brain cells, such as hypomyelination, lysosomal dysregulation, and impaired metabolism of lipids and polysaccharides. Importantly, pathway analysis in brain cells revealed mitochondria-related alterations in all Mcoln1-/- brain cells, except oligodendrocytes, that was not possible to resolve in whole tissue. We also report unique proteome signatures and dysregulated pathways for each brain cell population used in this study. These data shed new light on cell-intrinsic mechanisms of MLIV and provide new insights for biomarker discovery and validation to advance translational studies for this disease. One (or more?) of these parolees appears to be raping neighborhood women, and the Unit has to figure out who's doing it before Carisi gets beaten to a pulp by meathead vigilantes. Luckily Michael Rapaport is around to save Carisi's bacon once, but is he the guy? As always, rabbit holes are ventured down, with time spent discussing the physiological functions of junkie junk, the college and pizza scene in Utica, beating piss tests, and Staten Island cannoli. Abstract: Neuronal mitochondrial fragmentation is a phenotype exhibited in models of neurodegeneration such as Parkinson's Disease. Delineating the dysfunction in mitochondrial dynamics found in diseased states can aid our understanding of underlying mechanisms for disease progression and possibly identify novel therapeutic approaches. Advances in microscopy and the availability of intuitive open-access software has accelerated the rate of image acquisition and analysis, respectively. These developments allow routine biology researchers to rapidly turn hypotheses into results. In this protocol, we describe the utilisation of cell culture techniques, high-content imaging (HCI), and subsequent open-source image analysis pipeline for the quantification of mitochondrial fragmentation in the context of a rotenone-based in-vitro Parkinson's Disease model. As the case against Sir Tobi is set to begin in the SVU season finale for season 21, The Things We Have To Lose, a few previous victims from the season resurface, such as Joelle whose husband Leon is getting released from jail after doing 6 months for beating her. Fin allows himself to get too involved which results in him giving Joelle’s son Andre a phone to call him in case Leon comes over. Of course, Leon does and Fin makes a decision resulting in Leon’s death. Kat is also dealing with a previous victim when Lakira calls because she believes that Davies has attacked another trans prostitute on the piers. Meanwhile, Ivy Bucci is acting out. Her mother seeks help from the squad and Rollins takes it upon herself to find the girl and get her on the right back. As if they weren’t busy enough, Carisi and Benson are prepping victims for the big trial against Sir Tobias Moore, whose medical situation pushes back the trial. In SVU 21x19 "Solving for the Unkowns", Jill Bailey is arrested for driving her cupcake truck into a parked patrol car. At arraignment she realizes that she’d been assaulted. Luckily, Fin and Carisi are there to take her statement. Jill was on a date with a man named Luke (played by guest star Eddie Kaye Thomas) that she met on True Heart, a dating app. The detectives head to the bar where they met and get security footage to track down the suspect. Fin and Kat discover where Luke lives and pay him a visit. He admits to sleeping with Jill but says it was consensual. Benson says that this Drug Facilitated Rape was well planned and suggests this wasn’t his first time. Rollins finds a similar case in Brooklyn with a second victim. She identifies Luke as her rapist and Benson makes a public statement leading to more victims to step forward. One of the victims, Piper, had a bad reaction to the drugs and had a stroke. When SVU threatens Luke with harsher charges he turns on his accomplice, Ash, the bartender who makes “artisanal, tailored” cocktails allowing Luke to rape these women. SVU takes them down. Meanwhile, Rollins is promoted to Detective Second Grade and Kat casually comes out. Episode 17: A ballerina, Dehlia, realizes that her intimate session in studio X with a male dancer in her company is released on a porn site for all the world to see. She takes this to the SVU squad who uncover that these revenge porn videos are a well known secret for these dancers. Carisi and Rollins get the PornMonger man, guest star John Waters, to remove the video, but the internet it forever. Back at the dance studio, it seems that the choreographer has been blackmailing the females into sex, promising them solos and to get the video deleted. Any dancer that complains ends up with an offer to be auctioned off or never work again. Once Benson finds a dancer named Rose, with nothing left to lose, they set up an undercover opportunity by bringing back Sgt Khaldun. Eventually everyone goes down, but Dehlia will never dance again. When the team can't prove the Rev paid her off for sex, they threaten her with jail time for theft. Eventually she shows them a video of him raping her on her 15th birthday. The case looks weak until the Rev's wife turns on him, feeling guilt for not protecting the girls, she asks their forgiveness and gives them permission to finally speak the truth. The Rev takes a deal and ends up in jail. His lawyer, guest star Rick Fox, is bummed that he and Olivia met under these circumstances.
