Study Identifies Aging-Associated Mitochondrial Circular RNAs

BUFFALO, NY — February 24, 2026 — A new #research paper was #published in Volume 18 of Aging-US on February 10, 2026, titled “Aging-associated mitochondrial circular RNAs.” Led by first author Hyejin Mun from the University of Oklahoma — with corresponding authors Je-Hyun Yoon from the University of Oklahoma and Young-Kook Kim from Chonnam National University Medical School — the study profiles mitochondrial circular RNAs in Peripheral Blood Mononuclear Cells (PBMCs) from young and old human cohorts and probes how mitochondrial circRNAs and the mitochondrial RNA-binding protein GRSF1 relate to mitochondrial metabolism and cellular senescence. Using total RNA sequencing of PBMCs from young and old donors and complementary cell-based experiments, the authors report that a large fraction of circular RNA junctions originates from the mitochondrial genome, with MT-RNR2 producing the most abundant circular junctions. They show that circMT-RNR2 levels are depleted in older cohorts and in replicative senescence of human fibroblasts, and that the mitochondria-localized RNA-binding protein GRSF1 interacts with both linear and circular MT-RNR2. Loss of GRSF1 reduced circMT-RNR2 levels, decreased mitochondrial TCA intermediates (fumarate and succinate), and accelerated cellular senescence and mitochondrial dysfunction — findings that link mitochondrial circRNAs to mitochondrial energetics and proliferative status in younger cells. “Taken together, our findings demonstrate the existence and possible function of circular MT-RNR2 during human aging and senescence, implicating its role in promoting the TCA cycle.” The authors note key limitations and outline next steps: clarifying the biogenesis mechanism of mitochondrial circular RNAs (including whether trans-splicing contributes), mapping direct interactions between mitochondrial transcripts and metabolic enzymes, and performing mechanistic studies (in vivo and in additional human cohorts) to test how circMT-RNR2 and GRSF1 influence mitochondrial energetics and organismal aging. These follow-ups will determine whether mitochondrial circular RNAs are actionable targets for modulating mitochondrial metabolism or delaying aspects of cellular aging. Today's episode features guest host Michael Upshall (guest editor, Charleston Briefings) who talks with Michele Frison, Postdoctoral Research Assistant at the MRC Mitochondrial Biology Unit at the University of Cambridge. Michele is originally from Italy and completed his PhD at UCL in England with a focus on cell biology. He defines himself as a cell biologist and biochemist, and much of his current research specializes in cell and mitochondrial biology, aging and disease.  The Mitochondrial Biology Unit is part of a larger infrastructure that runs clinical trials for rare diseases. Michele says the process is very slow, taking up to a decade or so, but that he has been fortunate enough to have made a recent discovery that could well lead to clinical trial: the protein target that he's worked with is actually a target for drugs that have already passed phase one clinical trials and are now passing phase two for Parkinson's disease. If the research he has done in models replicates in humans, then it could well lead to a drug for diseases that affect one in 8,000 humans. Michele also talks about the disconnect between the public understanding of science and scientific practice, research culture and competition, open science and the review process, and the vast amount of literature that scientists have to keep up with, as well as his views on the use of AI and social media in his field. I start every day with your podcast - it is truly the best out there! My question is regarding the medication, Leucovorin, and its use in treating Autism. I have a 3 year old son who is non-verbal and is moderately Autistic. His pediatrician has prescribed Leucovorin on a titration schedule where he will be monitored for its effects. To note my own lab results, I have MTHFR, high levels of mercury and aluminum, low electrolytes/minerals, anemia/low ferritin, and gut issues - likely these were passed along to my son. I plan to have him complete the labs now that he is old enough. What is your advice on Leucovorin and its safety & efficacy, also should I be concerned with potential fillers and aluminum in the drug?       Sheena: Hi Dr C! Hope you and your team are well. I was just wondering if all bovine collagen has protein in it, even if it doesn't say it on the label? And if so, does the protein count towards my daily protein intake? Non starchy carbs make me jittery, as well as stressors in general, but it's not blood sugar, as I've worn monitors and my levels seem fairly ideal.
