Sujets traités : - Mettre les mains à la patte. Depuis 1987, la ville de Sélestat s'est inscrite dans le dispositif l'Art à l'école. Le principe, permettre aux écoliers d'avoir une première expérience artistique. Après des échanges en amont avec un artiste, le point d'orgue de cette mesure était hier, pour plusieurs classes de la commune. Dans la classe de CE1 de Mme Denner, à l'école Wimpfeling, les enfants devaient réaliser des figures avec des ficelles. Une activité qui bien plu à Eliott. Pour David Séchaud, scénographe pour la Compagnie Placement Libre, tout peut être de l'art, de la culture, tout dépend comment on l'aborde. Les écoliers de petite et moyenne section d'Hilsenheim ont enfin pu retourner en classe ce jeudi matin. Chose qui n'était plus arrivée depuis le 26 avril, alors qu'il n'y avait plus de remplaçant disponible pour occuper ce poste. Une contractuelle est donc présente du lundi au jeudi et une autre institutrice se charge du vendredi. - Marckolsheilm se dote d'un conseil local de santé mentale. Avant le début de la crise, la commune avait déjà entamé une réflexion sur le sujet, mais l'épidémie n'a fait qu'accentuer certaines problématiques. Ce qui a accéléré le processus. Ce conseil local de santé mentale, CLSM, regroupe plusieurs entités : médecins, psychologues, personnel médical, policiers, pompiers et associations notamment. Objectif, mettre en place une collaboration entre ces différents acteurs afin d'améliorer la prise en compte de la santé mentale, de mettre en place des actions ou politiques locales pour accompagner les personnes qui en ont besoin. - Le suspect du mont saint-odile a été placé en détention provisoire. Dimanche, non loin d'un des parkings du site, un homme reçoit un coup de couteau. Il est blessé à la gorge sur une quinzaine de centimètres. Son agresseur s'enfuit alors à moto et parvient à échapper au dispositif mis en place par les gendarmes. Il sera finalement interpelé le lendemain soir à Krautergersheim. La victime avait raconté aux DNA qu'à ce moment-là, cette personne délirait, n'était plus elle-même. Hier, le suspect a été mis en examen par le juge pour tentative d'assassinat. En récidive suite à du trafic de stupéfiants il a été placé en détention provisoire. - Comment continuer à faire du caritatif malgré la crise sanitaire ? The aim of this doctoral thesis was to investigate the factors relevant in plant interaction of two plant growth promoting rhizobacteria (PGPR). For this, the strain Acidovorax sp. N35 isolated from surface sterilized wheat roots and the two strains F4 and F7 of Rhizobium radiobacter, a bacterium associated with the plant growth promoting fungus Piriformospora indica, were chosen. First of all, the isolate N35 was characterized using phylogenetic and taxonomic methods. The 16S rRNA gene sequence analysis showed that strain N35 has the closest sequence similarities (98.2, 98.5 and 99.0 %) to the environmental Acidovorax species A. delafieldii, A. facilis and A. defluvii. The DNA-DNA hybridization values clearly separated the isolate from these three species. Additionally, phenotypic properties, such as substrate metabolization profiles as determined by a Biolog GN2 assay and cell wall fatty acid profiles concerning the fatty acids C16:0, C16:1ω7cis/trans, C17:0cyclo and C18:0cyclo and C19:0cyclo, facilitated the differentiation of the newly isolated strain N35 from its closest relatives. Thus, the strain N35 was classified as representative of a new species within the genus Acidovorax, and the name Acidovorax radicis sp. nov. is suggested. “Cand. A. radicis” N35 undergoes an irreversible phenotypic variation, resulting in different colony shapes on agar plate. In soil system, both phenotypes showed a plant growth promoting effect both on barley roots and shoots. The wild type N35 (rough colony type) had a better plant growth promoting effect on barley in comparison with phenotype variant N35v (smooth colony type). Wild type and phenotype variant cells of “cand. A. radicis” N35 were labeled with GFP and/or YFP and their separate and co-colonization behavior was investigated in a monoxenic system and a soil system using a CLSM for detection. Both types of N35 could endophytically colonize barley roots after 12 weeks inoculation in the soil system. Competitive root colonization behavior was observed after co-inoculation with differentially labeled wild type N35 and phenotype variant N35v bacteria, where the wild type showed dominant colonization of barley roots compared to the phenotype variant. Moreover, the variant N35v lost its motility due to missing flagella and swarming ability. The differences of both types at genetic level were investigated using whole genome sequence data obtained from 454 pyrosequencing (Roche) using the GS FLX Titanium chemistry. As only difference in the genome sequence, a 16 nucleotides deletion was identified in the mutL gene, which encodes for the mismatch repair protein MutL. In phenotype N35v, the frame shift caused by this deletion leads to the formation of a stop codon in the coding gene, resulting in a truncated MutL protein with a missing functional MutL C-terminal domain. This mutation occurred in exactly the same way in all investigated phenotype variants. These results suggest that MutL might be directly or indirectly responsible for the phenotypic variation in “cand. A. radicis” N35. Quorum sensing signaling molecules produced by “cand. A. radicis” N35 were identified using biosensors as well as Fourier transform ion cyclotron resonance - mass spectrometry (FT-ICR-MS) and ultra performance liquid chromatography (UPLC). Both types of “cand. A. radicis” N35 possess the same AraI/AraR quorum sensing system, which belongs to the LuxI/LuxR type. The two N35 phenotypes produced nearly the same amount of 3-OH-C10-HSL in the exponential growth phase. A co-inoculation experiment of AHL producing wild type N35 and a constructed AHL negative mutant N35 ΔaraI showed that wild type N35 had a dominant colonization behavior compared to the AHL negative mutant on barley roots in a monoxenic system. These data indicate that quorum sensing is involved in regulation of root colonization by “cand. A. radicis” N35. The second examined PGPR, R. radiobacter, which occurs naturally as endofungal bacterium in the plant growth promoting fungus P. indica, was demonstrated to colonize the surface of barley roots with fluorescence in situ hybridization (FISH) in a monoxenic system. The interaction of P. indica harboring R. radiobacter with other rhizobacteria was investigated using plate confrontation assays. Antibiotics and lipopeptides produced and excreted by the plant growth enhancing rhizobacterium Bacillus amyloliquefaciens FZB42 and the biocontrol rhizobacterium Pseudomonas fluorescens SS101 were shown to be responsible for the observed inhibition of P. indica by these bacteria. R. radiobacter F4 and F7 were able to synthesize a variety of oxo- and hydroxyl-C8- to C12-HSL compounds. In addition, both strains also produced coumaroyl-HSL when coumaric acid was supplied in the medium. The lactonase expressing transformants F4 NM13 and F7 NM13, which are the AHL negative phenotypes, abolished the lipase and siderophore activity. Considering this, quorum sensing influences the production of metabolites including lipase and siderophores in R. radiobacter F4 and F7. Further work should be directed to the question whether quorum sensing also plays a role in the interaction of the bacterium with fungus and/or plant.

The Inaugural OPPI decided I wanted, in addition to the weeklies and monthlies, something more communal to the general world: an homage show for fellow podcasters while it is unlikely that anyone who visits this backward little page would  not know the vastly more popular sites, I'm still rather ignorant of the world so my naivete allows me be ignorant of better sense - even if I know better. This one goes out to John Peel. I want to nominate John Peel as the patron saint of indie music, podcasters, and the overlooked. While Peel never (to my knowledge) had a podcast his singular taste in music, self effacing humor, and influence has left on indelible mark on the music of today.Beginning in the late sixties as a pirate radio broadcaster with his Perfumed Garden show to his death in 2003 he played music that never would have made air time otherwise. Odd and particular tracks, his vast selections always straining for that rare thing of uniqueness. His passion itself often clouded the choices he made since he valued uniqueness over quality. More personally he lit the fire for music under my ass. I came across the 1986 Festive Fifty selection by more accident than anything and immediately knew that at least 40 of the tracks I had never heard and never would have heard without the benefit of his show. And so it goes - music is shared and we all learn a little bit more about the world. Not too long after that I launched into finding out what I was missing in the music of today and quickly abandoned the comfort songs of familiarity for the jagged edge of the unknown. For all the horrible noise I've heard since then I can only blame the musicians and for ever jaw dropping and heart pounding track buried in the mess of it - I can only thank John Peel. 1. Canned Heat - Rollin' and Tumblin' aired on Perfumed Garden, 1967A nice nice tune and the sign of a quality DJ is when he plays his friends music on the air. 2. I couldn't find much about this period except for his shows signature tune.5. The Jam - Down in a Tube Station at Midnightaired on Festive 50 (F50) 1979The Jam, almost exclusively reserved for retro hipsters in Williamsburg nowadays is still a solid band well worth the 30 year hype.6. This song coming near the end of Peel and The Falls decade long friendship is a capstone to the efforts of DJs to play what they love and bands to play what they love. Play it for a friend and feel the connection of shared experience bridge the gap of our lonely islands of consciousness. Music to share is music to live by. Let's take off the headphones folks - we've all got that one golden track. Get it out there and make Peel proud.

The article reviews the existing methods of multicolor FISH on nuclear targets, first of all, interphase chromosomes. FISH proper and image acquisition are considered as two related components of a single process. We discuss (1) M-FISH (combinatorial labeling + deconvolution + widefield microscopy); (2) multicolor labeling + SIM (structured illumination microscopy); (3) the standard approach to multicolor FISH + CLSM (confocal laser scanning microscopy; one fluorochrome - one color channel); (4) combinatorial labeling + CLSM; (5) non-combinatorial labeling + CLSM + linear unmixing. Two related issues, deconvolution of images acquired with CLSM and correction of data for chromatic Z-shift, are also discussed. All methods are illustrated with practical examples. The aim of this PhD thesis was to examine the endophytic colonization behavior of Azospirillum brasilense and Herbaspirillum sp. N3 on wheat roots. The application of the FISH method using species specific phylogenetic oligonucleotide probes and GFP tagging facilitated the detection of a differential colonization behavior by the A. brasilense strains Sp7 and Sp245 on three different wheat varieties (Triticum aestivum). For this purpose a confocal laser scanning microscope (CLSM) was used, which enabled three-dimesional analysis of bacterial colonization of the root. Especially GFP tagged strains were well suited for this application, as there was no pretreatment or sectioning of the root sample necessary. Strain Sp7 was only located on the root surface of all wheat cultivars, whereas strain Sp245 was also found inter- and intracellulary in the outer root cortex layers. There was no recognizable connection between the growth stimulating effect of the inoculum (PGPR-effect) and the localization of the bacteria. The most pronounced PGPR-effect could be observed with the Brazilian wheat cultivar, which seemed to gain greatest benefit of its partnership with A. brasilense due to a certain adaptation to the inoculum. As the production of the auxin IAA (indole-3-acetic acid) plays a major role in stimulating plant growth, the expression of the key gene ipdC (indole-3-pyruvate decarboxylase) was examined. For this, several methods were tested to generate a fusion of the ipdC promoter with a gfp or rfp reporter gene. Constructing a translational promoter fusion with the gfp variant mut3 on plasmid level made expression analyses possible. With this method the promoter region of strain Sp7 located directly upstream of the ipdC start codon was found to differ only in a few bases from strain Sp245. But further upstream a region of about 150 bases was identified in strain Sp245, which was missing in strain Sp7. For Sp245 two different fusions were constructed, which contained the Sp245 promoter region homologous to strain Sp7 and the whole promoter region of Sp245, respectively. With these constructs the importance of the promoter region only present in strain Sp245 for control and intensity of ipdC expression in A. brasilense Sp245 could be demonstrated. Additionally an induction of the corresponding ipdC promoter fusions of Sp7 and Sp245 was achieved when adding phenylalanine or tyrosine. Total promoter activity was higher in strain Sp7 than in strain Sp245, and ipdC expression appeared to be subject to a stricter control in strain Sp245. These results were confirmed, when the strains containing the promoter fusions were used as reporters for ipdC expression on wheat roots. A demonstration of the induction of the ipdC promoter by root exudates in situ was possible. Finally, isolate N3 from surface sterilized wheat roots was characterized in detail. According to the 16S rDNA sequence data the isolate was phylogenetically allocated to the genus Herbaspirillum. But a subsequent DNS-DNS hybridization ruled out, that the strain belonged to any of the known Herbaspirillum species. Thus, the isolate, which might represent a new species, was named Herbaspirillum sp. N3. A specific, 16S rRNA targeted probe was constructed, which facilitated the examination of wheat roots colonization by this bacteria using FISH. Additionally the strain was GFP tagged to enable the detection in uncut root material. By this, an unequivocal demonstration of the endophytic colonization by Herbaspirillum sp. N3 mainly within the intercellular spaces was possible.

N-Acylhomoserinlactones (AHL) are signalling molecules in gram-negative bacteria, which regulate, in a cell density dependent way, important interactive functions. This phenomenon is known as quorum-sensing. This work characterised the microbial ecology of the autoinducer (AHL) producing bacteria Serratia liquefaciens MG1 and Pseudomonas putida IsoF in the rhizosphere of tomato plants. Gfp- and rfp-tagged strains of the AHL producing wiltypes S. liquefaciens MG1 and P. putida IsoF were compared with its AHL-negative mutants, which were unable to produce AHL. Two kinds of plants cultivating systems were used: a defined axenic system and a complex soil system. The characterisation of the root colonisation behaviour was performed using confocal laserscanning microscopy (CLSM) and cell counting of bacteria. Fluorescence in situ hybridisation (FISH) and terminal restrictions fragment length polymorphism (t-RFLP)-techniques were used to examinate shifts of the bacterial population in the rhizosphere on tomato plants. The effective in situ production and spreading of AHL on tomato roots was demonstrated with P. putida IsoF using an AHL-sensor strain P. putida F117 pKR-C12. AHL was produced in effective concentrations in the rhizosphere of tomato plants and influenced the bacterial rhizosphere population. However, the AHL-production had no influence on the colonization behaviour of the AHL-producing strains S. liquefaciens MG1 and P. putida IsoF.

Untersuchungen zu Wechselwirkungen zwischen flexiblen kationischen Lipidvesikeln und DNS sowie in vitro und in vivo Eigenschaften der daraus hergestellten Komplexe

Ziel der Arbeit war es, flexible kationische Lipidvesikel zu entwickeln, die für den transdermalen Transport von DNS geeignet ist. Um positiv geladene Lipidvesikel herzustellen, wurde in einem Ansatz SPC und CTAB verwendet, in einem anderen SPC, Polysorbat und DC-Chol. Nach der Herstellung dieser beiden Formulierungen fand eine in vitro und eine in vivo Untersuchung der beiden Trägersysteme sowie deren Wechselwirkung mit DNS statt. Dabei ergab sich, daß die DNS-Adsorption an die polysorbat-freien Vesikel grundlegend anders erfolgt als an die polysorbat-haltigen Vesikel. Die Flexibilität der beiden Formulierungen nahm durch die Beladung mit DNS deutlich ab, der Durchmesser der DNS-haltigen Vesikel im Vergleich zu den DNS-freien For-mulie-rungen zu. Beide Eigenschaften änderten sich bei den gewählten Formulierungen jedoch in unterschiedlichem Ausmaß. Grundsätzlich waren die polysorbathaltigen Formulierungen in der sich anschließenden in vitro Charakterisierung fähig, Zellen zu transfizieren, wenn auch nicht sehr effizient. Zur Überprüfung des in vivo Verhaltens der mit DNS beladenen Polysorbat-DC-Chol – Vesikel wurden die Vesikel schließlich mit einem Fluoreszenzfarbstoff markiert, die DNS mit einem weiteren Farbstoff; die verwendete DNS codierte ebenfalls für einen Fluoreszenzfarbstoff. Die so markierten Formulierungen wurden NMRI – Mäusen intra- bzw. epicutan appliziert; einige Zeit später wurden Biopsien der behandelten Hautareale entnommen und mittels CLSM untersucht. Dabei waren DNS und kationische Vesikel bis in eine gewisse Hauttiefe nachweisbar, wobei sich unbeladene und mit DNS beladene Vesikel nicht signifikant in der Penetrationstiefe unterschieden. Eine Transfektion der Hautzellen wurde bislang jedoch nicht beobachtet. Zusammenfassend ist zu sagen, daß die Anwesenheit von Polysorbat in der Membran nicht nur für die Flexibilität der Vesikel zwingend notwendig ist, sondern v.a. für die Stabilität der Vesikel nach der Anlagerung von DNS. Da die Flexibilität des Trägersystems durch Anlagerung der DNS deutlich abnimmt, ist es für weitere Ansätze wohl notwendig, den unbeladenen Vesikel eine extrem hohe Flexibilität zu verleihen, um selbst nach der Bindung der DNS an diese Träger noch eine ausreichende Flexibilität für die Permeation durch die Haut zu erhalten.

MICROMORPHOLOGY AND GENE EXPRESSION IN MUSCLE AND SHELL DEVELOPMENT OF THE MOLLUSCA

This work comprises detailed studies by scanning electron microscopy (SEM), transmission electron microscopy (TEM), fluorescence staining combined with confocal laser scanning microscopy (CLSM), as well as serial sectioning analyses and reconstruction techniques to elucidate the development of the larval and adult musculature of several basal representatives of the molluscan classes Polyplacophora, Bivalvia, Scaphopoda, and Gastropoda. Special reference is given to the shell musculature. In addition, aspects of the myo-anatomy of adult Solenogastres are reconsidered. A further part of this study deals with scaphopod shell morphogenesis and expression of the homeobox gene engrailed (en), in order to gain insights regarding the scaphopod-bivalve relationship. The results enable far reaching conclusions regarding the evolution and the phylogeny of the Mollusca. Solenogastres TEM analysis of adult Solenogastres revealed a mesenchymate body wall musculature which consists of outer ring, intermediate diagonal, and inner longitudinal muscles and resembles the condition of other worm-shaped taxa. The ventrally inter-crossing dorso-ventral musculature, which is diagnostic for the Mollusca, is arranged in multiple serial units along the anterior-posterior body axis. Polyplacophora During development, the chiton larva undergoes an intermediate stage in which the dorso-ventral musculature is serially arranged as in adult Solenogastres. The concentration into seven (and later eight) functional shell plate muscle units is a secondary condition which takes place after metamorphosis. Thus, assumptions of a primarily "segmented" (i.e. annelidlike) character of the polyplacophoran shell plate musculature are rejected. In addition, the anterior (i.e. pre-trochal) body region of chiton larvae shows a muscular grid which is lost at metamorphosis and resembles the body wall musculature of adult aplacophoran (Solenogastres + Caudofoveata) molluscs. Both, the multiple seriality of the dorso-ventral muscles and the apical muscle grid are regarded as ontogenetic recapitulation of the basal molluscan condition which is fully expressed in the adult body plan of Solenogastres. This infers a non-segmented, worm-shaped ancestor at the base of molluscan evolution. The existence of a larval ring-shaped muscle that underlies the prototroch cells (prototroch muscle ring) is a shared feature of polyplacophoran, gastropod, and bivalve larvae (see below) and suprataxic homology of this organ is proposed. Bivalvia Besides a rather complicated set of larval retractor muscles, the veligers of autobranchs (i.e. all Bivalvia except the Protobranchia, the latter with a test-cell larva) exhibit a distinct prototroch muscle ring similar to chitons and gastropods. Both systems are entirely larval and are resorbed during metamorphosis. Scaphopoda The general ontogeny and especially myogenesis in the dentaliid scaphopod Antalis entalis proceeds much more direct than in polyplacophorans or gastropods. Accordingly, distinct larval muscle systems are lacking. However, the paired cephalic and pedal retractors both form additional fibers which project into the region of the prototroch and are lost at metamorphosis. The existence of a distinct, paired cephalic retractor system, which is also found in the basal gastropod and cephalopod bauplan but not in the Bivalvia, suggests a clade comprising the Scaphopoda and Gastropoda + Cephalopoda. This is strengthened by expression data of the homeobox gene engrailed, which plays a significant role in molluscan shell formation. While two dorso-lateral centers of engrailed expression, which correspond to the two centers of initial shell calcification, are found in early bivalve veligers, engrailed is exclusively found in mantle margin cells surrounding the single anlage of the embryonic scaphopod shell. In contrast to bivalves, the scaphopod shell is thus formed from a single center of calcification, and a scaphopod-bivalve sistergroup relationship is therefore rejected. Gastropoda Primitive gastropods, such as the patellogastropods Patella vulgata and Patella caerulea, show one pair of asymmetrically positioned larval retractor muscles which have distinct insertion sites at the embryonic shell. Another strict larval muscle system is the prototrochal muscle ring. All these muscle are lost before, during, or shortly after metamorphosis. Parts of the adult mantle musculature as well as the muscles of the cephalic tentacles are formed prior to metamorphosis, while the buccal musculature is of entire postmetamorphic origin. The process of gastropod ontogenetic torsion is mainly caused by muscular activity of the larval retractors, while the adult shell musculature arises after the completion of torsion. Thus, ontogenetic torsion is regarded as an entirely larval process inferring that the arrangement of the adult shell musculature - which can often be reconstructed by muscle scars on fossilized shells - is not indicative for the question whether paleozoic univalved molluscs were torted or not.
