After Friday's news that Center for Biologics Evaluation & Research Chief Vinay Prasad will leave the FDA—again—at the end of April, stocks for several rare disease drug developers popped. UniQure, in particular, was up 51% in premarket trading on Monday. Prasad in a meeting last Thursday with select journalists called the biotech's Huntington's treatment AMT-130 a “failed” therapy, according to STAT News. Shares of Replimune and REGENXBIO—which have suffered rejections during the past year—also rose.One person who is not impressed with the plethora of rare disease drug rejections of late—H.C. Wainwright said in a note Tuesday that there have been at least five cell and gene therapies they believe could have been approved under prior FDA officials—is Wisconsin Senator Ron Johnson. Tuesday, Bloomberg News reported that Johnson has launched an investigation into these recent denials.Johnson called the FDA's request that uniQure conduct a sham surgery-controlled trial of AMT-130 “bureaucratic idiocy,” according to the publication. Meanwhile, uniQure and the FDA appear to be on different pages regarding the design of this prospective trial, with uniQure Chief Medical Officer Walid Abi-Saab referring to a 10-12 hour surgery during which [burr] holes would be drilled in patients' skulls and Prasad claiming on a media call last week that it would require only “one to three nicks in the scalp.”In other news, no episode of The Weekly would be complete without our weekly weight loss segment. Roche and Zealand Pharma's amylin analog fell short of Eli Lilly's rival candidate eloralintide; AbbVie reported what analysts called “competitive” results, with its amylin analog eliciting nearly 10% weight loss at 13 weeks in a Phase 1 trial; and Regeneron touted a much-needed Phase 3 win for Hansoh-partnered dual GLP-1/GIPR agonist olatorepatide in China. Beyond data, Novo Nordisk and Hims & Hers are together again, with Novo striking a deal to sell its injectable and oral GLP-1 medicines through the telehealth provider.Elsewhere on the business side of biopharma, experts are reporting a cut-throat atmosphere behind doors on the M&A front as the supply of companies available to buy dwindles.

A spate of youth crime in Christchurch has police urging parents to get in touch if their child is missing in action, so they can avoid becoming a statistic. It peaked in May with 162 young offenders dealt with in just one month in the city. It's remained high since, with police dealing with about a hundred youth offenders a month. Transport Infrastructure Ireland will assess safety measures on the M18 motorway in Clare following a spate of road traffic collisions on the route. In the last fortnight alone, emergency services have attended five crashes, which left two people hospitalised. Clare TD Cathal Crowe has since written to TII and the Transport Minister querying the possibility of variable speed limits on the M18, similar to those in effect on the M50. TII Communications Director Seán O'Neill says a review of the incidents will be carried out before any action is taken.

An "unprecedented" spate of violent crimes coupled with a significant police shortage in Northland means dozens of officers are being sent to the region on rotations to help out. The special staff operation has been running for three months, with about 30 police from other districts being re-deployed to Northland for five weeks at a time to plug the gaps, with no end date in sight. District Commander for Northland, Superintendent Matt Srjoj spoke to Lisa Owen.

During this week's Garda Focus, we heard that Gardaí in Shannon are investigating a burglary that occurred at a public house and off-licence in Drumgeeley, Shannon in the early hours of last Saturday morning, the 19th of July. At 3:50am on that date, a person dressed in a grey hoodie damaged the front door of the premises and when they were disturbed, they fled the scene. Gardaí are appealing to anyone that may have observed this person or any persons near the public house and off-licence in the early hours of last Saturday morning to contact Shannon Garda Station on 061-365900. The premises in question was the Carry Out Off-Licence. They posted on social media: “Break-ins, hold ups, assaults, we've had them all. Latest version around 3:50am this morning. If anyone has any information, can you please contact Shannon Gardaí.” Alan Morrissey spoke with Jim Moran, the owner of Carry Out Off-Licence in Drumgeeley, and began by asking him to explain what happened during the latest incident. But the violence did slow down once major gang figures had either been killed or placed behind bars. That is until recently, when, as crime reporter Perry Duffin writes, once again, there’s been a spate of gangland hits killing innocent people or minor criminals by an “honourless” underworld. After more than seven years in business, Angela Caglia Skincare is having its hockey stick moment.  “Our sales are up 437% in the past year,” founder Angela Caglia told Glossy. “We'll finish this year at close to $4 million [in sales] and around 90% of that will be the Cell Forté Serum; it's all we're promoting.” Since launching the hero product in October 2023, the brand's Cell Forté Serum has garnered several beauty industry awards from publications like Elle, Byrdie, TZR and Essence, and sold out three times on Violet Grey. And it was the catalyst for the brand's expansion into Nordstrom last month.  Caglia's focus now is keeping the serum in stock, and she hopes to expand the franchise next year with face and eye creams. The serum retails for $395 for 1 ounce. The serum's value proposition rests on its ability to replace antioxidant and hydrating serums, like those focused on vitamin C and hyaluronic acid, as well as exfoliating products and retinols, Caglia told Glossy.  The brand leads marketing materials with results from a 28-day clinical study where nearly all participants (87-91%) reported less hyperpigmentation, increased luminosity, improved skin elasticity and a more youthful appearance.  The serum is powered by "human-derived adipose mesenchymal stem cell (MSC) conditioned media," a technology Caglia discovered when researching treatment options for her mother's ongoing treatment of dementia. Today, she sources the material from a stem cell research lab based in Texas that specializes in stem cell banking and FDA-cleared clinical trials, she told Glossy. MSC-conditioned media is sourced from fat, called adipose tissue, which is donated by young and healthy plastic surgery patients and then processed in a lab. As Caglia explained in the latest Glossy Podcast episode, the stem cells are removed from the tissue and placed in a human-like environment where they excrete growth factors, cytokines and proteins, which are then used in the serum. The stem cells, which hold the patient's DNA, are removed before the broth goes into the serum.  Growth factors are a bit like little emails: They tell the other cells how to regenerate and act younger, which we don't fully understand yet.  Caglia is only one of very few brands playing in this space. Whereas there are many brands — like Eighth Day and Dr Diamond Metacine — that offer "bio-identical copies" of growth factors, few brands offer human-derived versions of growth factors.  According to market research company Spate, growth factors are a rising trend in online searches alongside skin care, with an average of more than 32,000 Google searches per month over the past year, marking a notable +202.7% surge.
