Joe Biden has stood on its head a proud moniker of American warriors: He epitomizes no better enemy and no worse friend. That deplorable distinction has been earned by, among other things: his surrender of Afghanistan; his greenlighting of a “minor Russian incursion” into Ukraine; his appeasement of Iran and undermining of Israel; and his months-long refusal to retaliate decisively for Iranian proxies' attacks on U.S. personnel. Now, immediately after the Taiwanese electorate voted for freedom over enslavement by Communist China, Biden announced, the United States does not support Taiwan's independence. That comment – surely being amplified by two former U.S. officials now in Taipei – rejects the de facto status quo and will encourage Chinese aggression. The Biden Doctrine of treating enemies better than friends ensures we'll dangerously have many more, and greatly emboldened, foes and far fewer and less reliable allies. This is Frank Gaffney.
One of the most amazing successes in the battle against cancer over the last two decades has been the introduction of antibody drugs that harness the body's own immune system to kill tumor cells. Finding those drugs may sound like a biology problem rather than a machine learning or a big-data problem. But actually, these days, it's both. Harry's guest this week is Leonard Wossnig, who's the chief technology officer for a UK company called LabGenius. The company uses a combination of synthetic biology, high-throughput assays, and machine learning to hunt for new drugs within a subclass of antibody medicines called T cell engagers that, loosely speaking, can grab tumor cells with one end and then grab tumor-killing T cells from the bloodstream with the other end. And Wossnig says the key to the whole thing is having the best data possible—meaning, data about their candidate T cell engagers and how specifically they bind to their targets in the lab assays. LabGenius has built an automated platform called EVA that runs experiment after experiment and uses active learning to zero in on T cell engagers with just the right ability to bind to their intended targets. He shares insights about the development of induced pluripotent stem cells, IPSC-derived NK cells, iNK cells, used by themselves or using their Flex-NK cell Engagers or in combination to address the challenges cancer patients face. Daniel elaborates, "That's also where the NK cells may have advantages over T-cells, in that it hasn't been reported that they cause CRS, GBHD, or other side effects that would require the patients to be kept in the hospital. Now, if we say the first iteration was derived from the patient's own blood, the second iteration is donor, the third generation, which we are one of a handful of companies to pursue and master, is the production of immune cells, including NK cells, from induced pluripotent stem cells." "We see also that you can direct the cells in two ways. You can direct the cells by inserting what's called a CAR, or chimeric antigen receptor, that's like a GPS on the cell. In this episode of ASTCT Talks, we sit down with Dr. Lia Gore, professor of pediatrics in hematology-oncology and bone marrow transplantation and co-director of the Developmental Therapeutics Program at the University of Colorado Cancer Center. Dr. Gore was the co-investigator on the AALL1331 trial and led the preceding open-label Phase 1–2 trial, AALL1121, which was the first to demonstrate the antileukemic activity of single-agent blinatumomab in children with relapsed or refractory B-ALL. We talk about her research and how the trial signals future research of bispecifics in pediatric care.

This is step one of Instagram ads in 5 easy steps. Your offer could be service based, product based, gated content (lead magnet, exchange of info for contact information). Step 2 Target Avatars Formulate the ad plan. Get super specific, look at geographics, don’t go cold turkey. Higher level of touch points is needed to convert people on social media compared to years past. “No one owes you their business. You need to be committed to getting that person to work with you. Be willing to be committed to this process.” Who are the people based on the offering and what types of targeting do I have available? Know who you're going to, what you're going to say to them. Step 3 Strategy Set up Start to build your audience and take advantage of your list of leads. If your business is able to sell to lots of different people, think of targeting a certain demographic, not everyone. Based on the offer you want to present, what can I provide for them? The content element is important because that's why people are on these platforms. If your offer is for hair coloring women 25-55, put out content focused on hair coloring such as top 5 trends for hair coloring. Engagers care about recent trends, so send them that offer focused ad, video, blog content. Step 4 Creative Assets 2 lines of copy and a simple image for your ad is sometimes all you need once you do the homework of steps 1-3. This step doesn't have to take a long time once you've done all the other work. You could literally just talk to the camera in a 60-second ad. Anything can be used as a way to provide value and generate interest. Video and video ads will generate more response. If you want a sale, scheduled call, etc - if you know your goal back in step two, it's easier to create your ad. Video works well because we can create audiences based on that one specific video. Back to the hair salon, if I've in some time past created a video and targeted a man who wants thicker hair. But before, it was women's hair coloring trends. Both are very different needs. They won't respond to the same things. I don't want to necessarily want to hit both demographics because that clouds the issue when you're doing Instagram in 5 easy steps. Based on the one hair color video I can create an audience of people who've watched it. The higher the percentage, the better the projected outcome. "Get your first color free when you book your appointment now." It can be as simple as that when you're targeting the right people. There will still be a disconnect if you've not created a targeted traffic pattern. You may offer many services, but for an ad, target just one, and typically the one that generates the most interest. One reason video is so powerful is because you can share much more content than in a written post. Being able to get your message across more quickly in a video with more content makes people more likely to consume that content. Being able to get your message across in video is the cheapest objective that gets the best results in those early stages. Dr Parren speaks about advances in the field of T cell engaging bispecifics. Recent advances in bispecific antibody production and clinic successes have resulted in increased interest, but several disadvantages remain when targeting via CD3, the most common approach. CD3 is present on all T cells which means there is poor specificity and activity, and also toxicity.At Lava Therapeutics, Paul and his colleagues are targeting A specific effector T-cell subset via the γδT cell (gamma delta T cell) marker Vδ2. These γδT cells are a homogeneous population of pro-inflammatory effector T cells that are able to recognize and attack tumour cells with increased efficacy and safety. previously it has been shown that the high presence of these γδT cells in tumors correlates with increased survival. Lava Therapeutics’ first product will enter the clinic at the end of next year.

Cai Kjaer CEO of Swoop Analytics explains how leaders and employees can listen to each other across issues, departments, and across the world. Today, we explore beyond the one on one dialogue. We also explore beyond team dialogue. We dive into listening at scale including thousands of more of conversations simultaneously. Cai Kjaer is an extraordinary leader in this field. His company Swoop Analytics focuses on the power of collaboration and people networks to get work done. They are a consulting company that maps organizational networks to find the most valuable metrics to drive collaborative business performance. Today’s Topics: The difference between the way men and women network. How everyone in the warehouse went to a guy named Elvis for help, yet it was completely unknown to upper management. There is no simple manual way to get this information sent to you. Cai shares how he got involved surveying people about who they contact to get work done. This eventually led to the founding of Swoop Analytics. Yammer is an enterprise network where people can collaborate. They took all of their IP and built the platform about relationship insights to build a profile around collaboration habits. If you have an enterprise social platform is make yourself visible and start to read. Having biases and seeing many sides. Collecting data across multiple industries. 5 archetypes: observer, broadcaster, responder, catalyst, and engagers. Engagers are most aspirational as listeners. The difference between listening and hearing. Being authentic in the way you interact with people. Killing myths and conversation at scale. Pay attention, look at people, and stay in the moment. How men and women play different roles in communication, Women are better at interaction with women and with men. Men don’t interact as much with women than they do with men. Gender issues and diversity in Silicon Valley. Forming more relationships with women and listening and engaging. Appreciating the role of women in a networked world. Ask questions. Listen and ask followups. Don’t get into the mindset of just telling. Their research concluded that to address local social and environmental issues effectively we need the whole community to get involved, we need better data at the local scale, and we need new ways of doing things. This was eye-opening on the seriousness of the responsibility of coaching young people. You're going to make mistakes- and that's OK - but use this as an opportunity to be humble and apologize - this is a great example to the kids. Teaching Children & Keeping it Fun Dr. Martin Toms': 'When you are coaching sports - you don't coach a sport, you coach a child' Don't professionalize youth sports - focus on developing the kids, not for the win today. Long-term goals instead of short-term. What do kids need from a youth sports coach? - Enjoyment, ownership, and to be intrinsically motivated. Winning It very much depends on what/who you are coaching - if you are coaching a prospective olympic gymnast, who's physical activity peaks at 14-15 - you have to start earlier But for all of us coaching kids ages 6-12- the focus has to be development over winning - so don't not play a kid just because it's a close game HUGE IDEA #1 Study done by Amanda Visek from George Washington University - asked kids: 'why do you play sports?' - 9 out of 10 answered because it is fun. Then she asked them to define what fun is: they came up with 81 different characteristics of what fun is - and 'winning' was down at #48 on the list.
