Aron Knickerbocker, President and CEO of Aulos Bioscience, is on a mission to extend the lives of cancer patients through safer and more effective immunotherapy using AI and machine learning to accelerate drug discovery and optimization. The company's lead drug candidate is an antibody that was designed using AI to harness the power of interleukin-2 to activate the immune system against tumors. The advantages of AU-007 over previous IL-2 therapies include the ability to selectively activate tumor-fighting immune cells while avoiding toxicity due to vascular leakage. Current trials are underway in advanced melanoma and non-small cell lung cancer, which are showing the potential for durable responses and immune memory. Aron explains, "So Aulos has really been founded to carry out a mission to extend the lives of patients with innovative, safe, and ultimately effective cancer immunotherapy. And you mentioned IL-2, which is the naturally occurring protein that we're seeking to harness and redirect in productive ways to help the patients. And this has really been kind of a driver for me over the years, wanting to improve on cancer patient care. This is a company with an important mission and a great team, and our program is really interesting. It's harnessing IL-2 in such a way that it sends it to the cells that are capable of attacking the tumor and killing the tumor cells, and keeps it away from the cells that suppress the immune response. It's really kind of using a double-edged sword in a productive way to help patients with cancer." "So this is the first antibody to go into human clinical trials. It was designed in part by leveraging an AI platform, as you noted. And that platform was created by our collaborators at Biologic Design. They're the company that created this molecule. And what biologic design does is essentially using an AI system that has machine learning algorithms that drive it. It mimics what the immune system does normally. So when we get sick and we need to make antibodies to something, our immune system says, Do I already have something that will kind of bind to the target or the virus or whatever it's trying to hit? And then it will optimize that. It will go through a process of rapid change. And what the AI system and the machine learning algorithms that have been trained on millions of antibodies in the targets to which they bind do is recognize patterns much like an AI chatbot might recognize language patterns." #AulosBio #Antibodies #MedAi #AI #IL2 #Onocology #Cancer #NSCLC #AdvancedCutaneousMelanoma #DrugDiscovery #BiologicDesign #Tregs aulosbio.com Download the transcript here

I [am playing] an Aulos plastic [flute], copy of a 18th century Grenser. Very nice Instrument for the price. Thought about a wood one but humidity control in our old house is not great. From Dave Rowlands: This is 'La Bernardina' by Josquin des Pres (1450-1521). Not known as a composer for bagpipes, but the leading composer of his time. I have chosen this because a) it is a new find for me, b) because if pipers had access to this music and good instruments they would have played it, and c) because we should not be hidebound to 'tradition', just because it does not come from a bagpiping tradition, does not mean we cant play it, and i cite Amazing Grace as just one case. This is played by Three Swayne D pipes, and one Swayne G pipe. I hope you like it and include it. Best wishes, Dave R From Charlie Rutan: Tu Scendi Dalle Stelle is the traditional Italian Christmas carol played by zampognari. Originally called 'Quanno Nascente Ninno', it was written down in 1754 by St Alphonsus Ligouri, with text in the Neapolitan language: and became so popular that it was later translated into Italian and became 'TU SCENDI DALLE STELLE', undergoing several small changes in its melody during that process. The melody probably existed in various forms for several centuries prior to its 1754 transcription, and is still a basis for many improvised PASTORELLES by zampognari today. I'm playing both Ciaramella ( the Italian folk oboe) and Sei Palmi Zampogna on this track. 'Sei palmi' refers to the length of the instrument's longest chanter, measured by the outsretched palm of the pipe maker's hand, much like the 'cubit' of the ancient world. Zampongne have existed in this form since at least the early 1300's; where we have evidence of the instrument being played in frescoes dated to that time. The zampogna is endemic to southern Italy, exists in about 20 different iterations in several sizes, and is a thriving bagpipe tradition to this day. Immunotherapies continue to redefine treatment for many types of cancer and one of the first immunotherapies approved and used clinically as a therapeutic is Interleukin-2, or IL-2. Aulos Bioscience is an immuno-oncology company that has developed AU-007, a potential best-in-class IL-2, that improves on many of the other IL-2 products in development. In this episode, we discuss the promise of AU-007 with President and CEO, Aron Knickerbocker. We love to hear from our listeners. Send us a message. Data drives decision-making at Aulos Bioscience. It's also driven the career trajectory of its CEO. On this week's episode of the Business of Biotech podcast, we get an introspective walk through Aron Kinickerbocker's strategically-orchestrated career path, from his early days in the labs at Bristol-Myers Squibb and Genentech to his leadership of Aulos Bioscience, a company that boasts the first AI-generated antibody in human clinical trials. We explore Aulos' innovative IL-2 antibody and the role AI played in its highly-targeted and toxicity-reducing design, and we do it in pragmatic fashion — Aron's not about the AI hype, he's about the AI data — and in its Phase 1/2 clinical trial, the data is looking pretty good. “Parameter nummer één is altijd schoonheid: het verhaal brengen op een muzikale manier, zeer puur, zeer goed. Dan voelen mensen dat. Kindjes zijn de beste parameters om te weten of iets goed is. Onderschat volwassenen ook niet: men voelt wel of iets bijzonder is. Ga op zoek naar schoonheid, en verlies daarin je DNA niet. Blijf jezelf geloven om een bepaalde weg uit te stippelen. Its back to heavy episodes again! We have an absolutely cracking episode this week where we take a look at ancient music from Biblical times from both the Hebrews and those in the ancient world around them. We are joined by our good friend Mitchell Bradley who, as a pianist, has a great insight with music and the comparisons we can make with old music to music we have today. We look at why music is important and timeless, and why it is so prominent in the Bible. This episode is jam packed with snippets and clips of sounds from ancient instruments and songs, so have a listen and enjoy! Follow along as we listen to the clips in the order that is posted in the sources below! We also ask you to keep an eye out for Mitch and Pete's new podcast, "Building On The Rock", a podcast which goes through the old Biblical stories we all know to find out who God is and what the stories really mean.
