Although our primary focus on the podcast is Multiple Sclerosis, we're well aware that the diagnosis journey for different conditions can be very similar. This is a viral disease that is spread to people by the bite of an infected mosquito. The most common symptoms of infection are fever and joint pain. But as you'll hear, Victoria also experienced brain fog and neurological issues, which will be all too recognizable to people with MS. And her initial symptoms were brushed off as being stress-related. Die Asiatische Tigermücke kann als sogenannter Vektor unter bestimmten Voraussetzungen verschiedene Krankheitserreger übertragen. Hierzu zählen etwa das Dengue- und das Chikungunya-Virus. Da diese Erreger in Deutschland bisher nicht verbreitet sind, ist das Risiko einer Übertragung durch die Asiatische Tigermücke auf den Menschen bisher gering. Expertinnen und Experten weisen aber darauf hin, dass eine weitere Ausbreitung der Asiatischen Tigermücke das Risiko für Krankheitsübertragungen erhöht und deshalb ernst genommen werden muss. Erhöht ist das Risiko insbesondere in infrastrukturell gut angebundenen Ballungsgebieten mit geeigneten klimatischen Bedingungen wie dem Rhein-Main-Gebiet.

Abstract: Downregulation of the host cell pro-apoptotic pathways confers a selective advantage to viral pathogens, and many viruses interfere with such regulatory mechanisms in order to enhance their propagation in infected cells. The Scribble cell polarity complex, which is composed of multiple adapter proteins including human Scribble, Dlg and MAGI, controls a variety of host cell functionalities including apoptosis, morphology, polarity, signaling and migration. In the recent years, components from several viral pathogens, including oncogenic viruses, have been found to associate with and modulate the activities of this complex, particularly that of the pro-apoptotic protein Scribble. Here, we establish a critical role for the downregulation of Scribble expression for the potentiation of Chikungunya Virus (CHIKV) infection. CHIKV is a well-known mosquito-borne alphavirus, which has caused outbreaks in more than 100 countries and constitutes a global health hazard. While the majority of the CHIKV proteome is well characterized, the role of a small structural protein 6K, and its transframe variant, TF, which is generated from the 6K sequence by ribosomal slippage and frameshifting, is not established as yet. We show that TF triggers the punctation, ubiquitination and degradation of Scribble during CHIKV infection, and that the downregulation of Scribble during CHIKV infection. We show that the association of TF with the PDZ domains of Scribble is mediated through a PDZ-domain binding motif at the C-terminus of TF. Thus, our work establishes a role for the downregulation of Scribble in alphavirus infections for the first time, while allocating a novel role in host modulation to the mysterious 6K/TF component of alphaviruses. What exactly does a virus do to invade our bodies? Scientist St. Patrick Reid addresses the mechanics through specific virus pathologies. He addresses how viruses use proteins to undercut the immune system and ensure replication, how the chikungunya virus infection has become more prevalent and often leaves chronic symptoms, and why these lasting effects from this RNA virus are especially puzzling and how scientists approach such questions. St Patrick Reid is an assistant professor in the Department of Pathology and Microbiology at the University of Nebraska. He focuses on the virology of proteins encoded by highly pathogenic viruses like the chikungunya virus infection. He explains what this means by describing his graduate and early career studies on Ebola and other host-pathogen interaction examples. Ebola is able to invade an organisms by encoding for proteins that block the immune system from responding. This gives the virus enough time to amplify itself and replicate. When the body is finally able to react, the virus is so pervasive it over reacts, often unleashing a dangerous over-response; in fact, this is what happens with COVID-19. Dr. Reid than describes the history of the chikungunya virus infection and how it has made its way from one part of the world to another, including South America and the southern United States. Transmitted by mosquitoes, it often results in a debilitating arthritis that can last for years and scientists don't understand how or why. In fact, that's one of his areas of research, to understand its pathology. As an RNA virus, it does not invade the nucleus and change the genome. He explains some possible theories and his own approach to solving this mystery. He also discusses his thoughts on COVID-19 from his protein-focused research and describes the magnanimous ways researchers are working together across the world to find ways to combat its effects. The Silva Lab studies chikungunya virus, a reemerging arbovirus that has recently caused explosive epidemics of febrile arthritis throughout many parts of the world. Dr. Silva’s research interests are focused on virus-host interactions that dictate chikungunya virus entry, tropism, replication compartment formation, and pathogenesis. By understanding the molecular mechanisms that govern viral and host determinants of viral replication, Dr. Silva aims to contribute to a better understanding of acute and chronic chikungunya disease. Dr. Silva also serves as the Facility Manager for the A-BSL3 laboratory at Rangos Research Center.
