In response to our offer to direct a film for us, Rajamouli called me and Urmila Gangaraju one day and said that he would be directing a film for us on his banner Viswamitra creations. He wanted us to be the producers of his first venture on Viswamitra banner. Yama Donga, one of the most eagerly awaited premium product of Tollywood is all set to hit the screens very shortly. This film is by far the costliest movies made in the Telugu film industry. This film boasts of the biggest indoor set erected so far in whole of Asia and incidentally is the costliest individual set. This Yama Lokam set was erected over period of 90 days with 300 workers working on the set day and night. The magnitude of the set was so much that it took almost another one and half month to dismantle the same. This set was erected in Ramoji Film City and the no.of shooting days in this set was 42 days. Apart from this the other sets erected for four songs, the set of the Hero House and the Heroine House are also done with lot of richness and grand taste. The biggest set in RFC was erected for Vaaji song in Sivaji and it occupied only 60% of the floor. The Yamalokam set occupied 100% of the floor. We used 15 generators at a time to light up the floor. It took 4 days to set the lighting to that floor. Senthil Kumar used the best available equipment to shoot this film. Since Yamalokam set demanded a great amount of lighting, we procured almost all available lighting equipment from Hyderabad movie infrastructure providers. We had to borrow some of the lighting equipment from Chennai too. Graphics work plays a very vital role because it is a socio fantasy. We employed various e-fx companies. Adel Edeli - a renowned 3D animator from Iran - did the 3-D work for the exterior of Yamalokam. EFX studio did the major work (around 90%) of the graphics part. They have been working on the film from April to August. Rajamouli did online editing and made sure that graphics work is done simultaneously along with the shooting. A team of 80 professionals spread over three outsourcing houses spearheaded by EFX are working on the graphics and the production house is confident of coming out with the most impressive visuals. Keeravani started the work on 2 July and will complete it on 7 August. It is taking over a month. Keeravani is immensely enjoying rerecording as it is a challenging task to work for a socio-fantasy It is partly true. Prabhas did the role of Viswamitra. But not for Yamadonga film. When Rajamouli wanted to design the logo of Viswamitra Creations, he needed a model for Viswamitra photo. He requested Prabhas as his physique is very good. Prabhas obliged for a photo shoot. The designer cut the physique of Prabhas and inserted in the logo of Viswamitra creations. We started the shooting on 2 January 2007 and completed it by the end of July. It took us 148 working days spread over a span of seven months. We exposed around 2 lakhs feet of negative. We shot most of the film indoors by erecting massive sets in Ramoji Film City, Annapurna studios and Rama Naidu studios (Nanakram Guda). Climax was shot in Talakona forest. A song and some scenes were shot at Golconda Fort in Hyderabad. Audio launched on July 14h, 2007 at Hotel Marriot in Hyderabad. Finally movie released on Movie was a hit at the box office collecting 29 crores share and ran for 50 days in 400 centers and 100 days in 61 centers. 3rd consecutive hit in SSR NTR combination.

Last year, Stephen Harrison expounded on a string of press releases presaging some of the most promising data of the last decade in NASH drug development. As Principal Investigator, he reviewed on the podcast Phase 2b results from Akero's HARMONY Trial. This is a 24-week study evaluating the efficacy and safety of the FGF-21 agonist efruxifermin (EFX) in patients with clinically relevant NASH (F2-F3). The study met its primary endpoint for both the 50mg and 28mg EFX dose groups. Respectively, 41% and 39% of EFX-treated patients experienced at least a one-stage improvement in liver fibrosis with no worsening of NASH by week 24 (compared with 20% for the placebo arm).Stephen walks through his presentation on Akero's lead product candidate EFX and two important points emerge. First, Stephen and Sven Francque share the idea that “not all FGF-21's are created equal.” FGF-21's themselves need to be stabilized due to their two-hour half-lives. The first FGF-21, pegbelfermin, was PEGylated and revealed significant challenges in clinical development. EFX is a bivalent structure – two molecules stabilized by a fusion protein. This appears to work far better. The conversation continues with more points and questions about the study results and implications. An interesting idea surfaces: EFX might function better as an induction therapy to be followed by an oral maintenance therapy and a long-term monotherapeutic solution. As the session winds down, the group speculate the dynamics of what happens when disease regresses to a point where there is far less liver damage.

This week attendees from around the world convened at the 73rd Annual AASLD Liver Meeting for a lively and momentous event in Washington DC. On the final morning, Stephen Harrison, Sven Francque, Jörn Schattenberg, Ian Rowe and Jeff McIntyre join Roger Green to discuss key takeaways. Sven is first to expand on a presentation that struck notable interest. He chooses to highlight a presentation by Laurent Castera, which Mazen Noureddin anticipated in our preview coverage. This focuses on a prospective head-to-head comparison of the different non-invasive tests in a large cohort of T2D patients with NAFLD. The paper concluded that F-AST and MAST scores outperformed MEFIB, FIB-4 and NFS for identifying fibrotic NASH in T2D patients with NAFLD. Stephen broadens Sven's analysis by noting F-AST as a standout test that is generally available for multiple different applications. The group puts forth commentary on the cost effectiveness and accuracy of these tests within enriched populations. As this subject winds down, Ian points to the particular challenge of selecting patients for treatment. The reality, he says, is that it's not a one size fits all approach. The conversation transitions from diagnostics to the topic of drugs. Stephen walks through his presentation on the efficacy and safety of Akero's lead product candidate efruxifermin (EFX) in patients with clinically relevant NASH (F2-F3). The results point to EFX significantly improving liver histology, liver fat content and noninvasive markers of liver injury, fibrosis, and glucose and lipid metabolism in patients with F2/F3 fibrosis due to NASH. Stephen shares his optimism for this particular FGF-21 agonist while Sven agrees that “not all FGF-21's are created equal.” The group explores the science behind the structure of the EFX compound before Roger recalls the question of where the right use for such a drug might exist. It's been previously suggested on this podcast that a medication not entirely suited for long-term therapy due to cost, dosing or side effects might serve best as induction therapy. With this in mind, the group addresses the dynamics of what happens when disease regresses to a point where there is far less liver damage. From here, Roger introduces a brief assessment of Arun Sanyal's presentation of topline results from a new analysis of the REGENERATE Trial.The last major theme in this episode involves the Nomenclature session. Jeff offers an objective account of how this subject reported before disclosing some concerns from the patient perspective. His primary concern lies in coming to a decision that causes regulatory agencies to require new clinical trials, thereby slowing approvals by several years. The three discuss the work of this clinical-stage biopharmaceutical company and its relevance to Fatty Liver. Inventiva develops novel and differentiated oral small molecule therapies for patients suffering from diseases with significant unmet medical need. The discussion centers mostly around clinical trial development for the pan-PPAR lanifibranor.

Doug Voet has an illustrious stage career appearing in numerous productions. Singer, actor, producer, horse lover and close friend of David's, they met in 1983 when David took over the lead role on Broadway in Joseph and the Amazing Technicolor Dreamcoat. Doug taught David the role and their friendship grew. Some years later David, who was starring in EFX in Las Vegas, offered Doug a new direction in his career inviting him to produce a new show, At The Copa, he had created. They also worked together on The Rat Pack Is Back and Doug went on to be David's tour manager on his US and UK tours. Robyn Hadden was personal assistant to David Cassidy when he took over the starring role in EFX, the biggest show in Las Vegas in 1996. David replaced Michael Crawford who had hand-picked Robyn to be his personal assistant. She continued working in that role with David throughout his time in Vegas where he also was behind successful productions, At The Copa and The Rat Pack Is Back. When David returned to concerts, Robyn toured both the United States and the UK with him making lifelong friendships with many fans. Robyn had planned to be a pharmacist and explains her journey into the world of entertainment, later working in Special Events. She earned her Certified Special Events Professional (CSEP) designation in 2000 and her Masters Degree from the University of Nevada in 2005. She served as Director of Alumni & College Relations at the UNLV College of Hospitality and continued her career at the Ohio State University where she was Associate Director (Special Events). In our amusing and, at times, emotional conversation, Robyn shares anecdotes from her years knowing David and explains why at 40 she decided to follow her dream of becoming a mother through IVF and an anonymous donor. She became mother to Jack in 2011, and later moved back to Ohio having met the love of her life.
