We have shown previously that coumarin derivative LM-031 enhanced CREB/BDNF/BCL2 pathway. In this study we explored if LM-031 analogs LMDS-1 to -4 may act as TRKB agonists to protect SH-SY5Y cells against Aβ toxicity. By docking computation for binding with TRKB using 7,8-DHF as a control, all four LMDS compounds displayed potential of binding to domain d5 of TRKB. In addition, all four LMDS compounds exhibited anti-aggregation and neuroprotective efficacy on SH-SY5Y cells with induced Aβ-GFP expression. Knock-down of TRKB significantly attenuated TRKB downstream signaling and the neurite outgrowth-promoting effects of these LMDS compounds. Among them, LMDS-1 and -2 were further examined for TRKB signaling. Treatment of ERK inhibitor U0126 or PI3K inhibitor wortmannin decreased p-CREB, BDNF and BCL2 in Aβ-GFP cells, implicating the neuroprotective effects are via activating TRKB downstream ERK, PI3K-AKT and CREB signaling. LMDS-1 and -2 are blood-brain barrier permeable as shown by parallel artificial membrane permeability assay. Our results demonstrate how LMDS-1 and -2 are likely to work as TRKB agonists to exert neuroprotection in Aβ cells, which may shed light on the potential application in therapeutics of AD. We are freakin' robots. For the LMD arc, Joshua points out two knock-off Captains America as well as the shell game that is LMDs and Doombots in the 616. Then Lani leads the conversation about identity, bad girl shenanigans, ill conceived ships, and the missed opportunities in LMD.
S4:E22 - Jess and Jared recap and discuss “World's End." As Jess puts it so well in the recap to the finale of this final third of season four, the show sort of loses its connection to reality from now on, and it changes fundamentally. Yo-yo is in the Framework and trying to save Mack before the Framework goes down for good, and Radcliffe's ghost in the machine is there to help. Robbie shows up to stop Ivanov's stupid robot face in his tracks, and uses his hellfire and badassedness to put Aida in her place, and really gets under her skin. He seems to be able to hurt her, but appears impervious to her. And this wraps up the season perfectly, connecting AIDA and Robbie's arcs with the Darkhold and the Ghost Rider curse in a full circle. Talbot has some hard news for Coulson, there is no S.H.I.E.L.D., and he needs him immediately to argue on its behalf. Robbie and Daisy quasi-flirt, and we were ROBBED. Mack doesn't recognize Yo-yo, and Radcliffe tries to help bridge that gap, but the dissolving Framework and Radcliffe's vibe are making the Framework Hope figure out what's going on. Jared is slightly confused by Ivanov and his Russian analyst LMD buddy's strategy, but we're happy to move on and not delve into Ivanov any more than we have to. At the meeting with Talbot Ivanov drops the Darkhold on the U.N. task force and Talbot calls "a load of L. Ron Horsecrap!" before being shot in the face by a Daisy LMD, framing S.H.I.E.L.D. and making Inhumans look bad. AIDA is a weirdo, reveling in torturing Daisy as Robbie destroys the hell out of the Daisy LMD. Daisy and Robbie tag team fight a buttload of LMDs and Coulson is super bummed to have missed the superhero team up of his (and our) OTP. We digress slightly because we lost Robbie's Ghost Rider spin-off, and that SUCKS. Coulson has a sweet, but difficult moment with Fitz and Simmons trying to get them to push through their issues and get them back to working in tandem. Yo-yo understands Mack's difficulty letting go, but can't let him die for Hope's framework self. Coulson mentions Robot May being more supportive, tipping May off to Coulson and Robo-May's connection. Daisy hacks the crap out the Framework to save Mack and Yo-yo, and makes some goo-goo eyes with Robbie. Yo-yo debates leaving with Radcliffe, and Hope's fears are confirmed, she knows she's not real (violating the first rule of Early Childhood Education). Fitz tries to reason with AIDA, and Jemma won't have coddle her, and AIDA doesn't respond super well. Coulson can't quite explain almost getting with Robo-May, but May agrees that they both deserve a do-over with each other. We have a JemmAndroid death fakeout, and AIDA is stopped by Coulson possessed by the Rider. Ghost Rider Coulson burns the crap out of AIDA, and puts an end to the Darkhold's creation once and for all. Mack looks for Hope in the Framework and tells her that she's the most real thing in the world, and Yo-yo won't leave him. Framework Hope begins to disappear, and we watch her die, erased from the program in Mack's arms, and we are destroyed. They go through Daisy's back door, and Mack remembers both Yo-yo, and his Framework life with Hope, and hopes to have that life with Yo-yo now. We go on for a bit about Mack's amazingness, and how is the perfect S.H.I.E.L.D. Director, and Jared's perfect choice for Captain America one day. Ghost Rider has an amazing exit as he opens a gateway to another dimension with his hellfire and chains, and we're so sad to him go. Daisy forgives Fitz on the team's behalf and Coulson decides they need a break and a meal, so they go out for dinner. We hear our favorite sentient Chronicom and trusted friend Enoch for a moment before everyone but Fitz disappears, and then Coulson's in space, and that's it! Also, congratulations to S.H.I.E.L.D. alumnus J. August Richards for coming out to his audience and the public as the proud Gay man he's been privately his entire life. We love our Deathlok! S4:E21 - Jess and Jared recap and discuss “The Return." We get into the penultimate episode of this...third of the season, but also, since it's the *final* third, the penultimate episode of the season in general! A long-absent non-Framework'd May and Coulson must reunite while fighting off an army of Ivanov-bots (Anton Ivanov? A.I.? *Really*, S.H.I.E.L.D. writers?) The Zephyr's literally on fire but Daisy, Jemma, Yo-yo and Davis fight the impending disasters and manage to pull through. Fitz is disoriented as the newly (In?)human AIDA teleports them to some picturesque beach. AIDA continues to mess with Fitz's brain by compounding his guilt, reinforcing that he made all his own choices in the Framework. Yo-yo yells at Daisy for leaving Mack behind, but understands why she had to let him make the choice, though against Daisy's request, really wants to jump into the collapsing Framework to try once more to pull Mack back out. While fending off the Robo-Russians Coulson finds deflecting May's questions to avoid addressing his feelings for her, and how he told them to Robo-May, more difficult than fighting off the scores of attacking androids. May kicks the crap out of a ton of A.I.vanovs in a hallway/doorway shot while Coulson holds his own, and it's freaking rad. Ivanov floods their facility at this point, though as Jess points out, what was he waiting for? Though, we maybe cracked the character's essence: he's really bad at planning/illogical/maybe just stupid. Fitz tries to reason with, and teach AIDA, who's almost infant-like emotional state combined with her Inhuman abilities leaves her exceptionally dangerous, though he manages to appeal to her growing empathy and she saves Mack with her teleportation. Jemma has no patience for either AIDA or Fitz and has them immediately taken into custody. May is told about Robo-May's sacrifice as they return to the base, and place AIDA & Fitz into the containment capsule as they pass an emotionally poignant fallen photo of Mace's heroic moment in Vienna. May and Coulson try to explain to Jemma what their Framework lives were like, and what they mean to them, in order to process what Fitz has become, but can't handle it, and Daisy and Yo-yo hit the nail on her head, explaining that she both worries about who Fitz is, and who he may love. We see Fitz try to let AIDA down gently, even relating himself to Ward. But as misguided as she is, and as immature, AIDA simply cannot deal with his rejection, and goes berserk. Talbot shows up and makes Jared's day when he tells Coulson to "shut his robot mouth, or I'll have my men weld it shut for you!" The superpowerful AIDA uses Lincoln's electricity to bust out as Coulson tries to convince Talbot he's not an LMD. AIDA cuts a swath through the playground to get to a fleeing Fitz who barely manages to escape on the Zephyr to a conflicted Jemma. There she comforts an emotionally destroyed, sobbing Fitz, and Yo-yo does what Daisy begged her not to before, and goes after Mack in the Framework. AIDA gets her emotions going by briefly smooching and then totally destroying the bodies of some of Ivanovs LMD duplicates, and Ivanov offers to help her gain control of this world as she had in the Framework. Talbot and his team abandon the ruins of the Playground, seeing no trace of the S.H.I.E.L.D. team or AIDA and her LMDs. An inter-dimensional gate left intact turns on and opens, with Ghost Rider stepping outside. As the episode closes we see Yo-yo strapped to a table in a Framework Hydra facility, falling apart at the seams. And other than our fictional law enforcement agency, fuck the police. S4:E15 - Jess and Jared recap and discuss “Self Control." Picking up where we left off, the team have all nearly been replaced by LMDs, and viewers and Fitz and Simmons are *freaking out* about it. This is a pretty dark/horror-y episode, and as Jess observed, this is a Jed Whedon joint, and he's said he loves Ward the most, so maybe he's a kind of a dark dude deep down. AIDA kills Radcliffe's human body, leaving his mind trapped in the Framework, Jemma and Fitz have a very standard "who's the robot" type exchange where Robo-Fitz is exposed and Jemma and Daisy (who's just found a room of Daisy LMD's) are *pretty much* the last real humans running around free in S.H.I.E.L.D., and Daisy is able to take out the LMD S.H.I.E.L.D. team to escape with the help of Robo-May, the most real and human of all the LMDs, who loves the real Coulson, not the LMD Coulson, breaking our collective heart something crazy. And shoutout to Jeff Steck and Ian Beavers, may they Rest in Peace in the Framework. S4:E14 - Jess and Jared recap and discuss “The Man Behind the Shield." Fitz explains the Framework as a VR training program, and he and Jemma plan to use it to locate the real May as the rest of the S.H.I.E.L.D. team searches for Mace and the Russian. We learn that people can get brain damage, or even die, in the Framework, and the stakes are just as high in their virtual world. Throughout the episode we see flashbacks to the beginning of May and Coulson's relationship, and it's heartbreaking. While Gemma and Fitz are unsuccessful in locating Radcliffe's other Framework, the team is able to save Mace and Daisy overpowers the Russian, and they return to the base. The episode concludes with Jemma and Fitz discovering that four of their friends had been replaced with LMDs, including Coulson, who boots up Robo-May. And we begin our discussion by going down a weird rabbit hole of references to Legends of the Hidden Temple, Guts, All That! and Ross Perot, before we ever get to the episode with one of our weirdest asides ever, brought to you by Nickelodeon™! Moon Knight finds himself wanting to join the West Coast Avengers and so it's in a provisional capacity that our Fist of Khonshu fights alongside the West Coast Avengers against the Zodiac, Scorpio, LMDs...and even the alien race, the Sligs! Josh brings his acumen for classic 80's run and both he and Rey get into the nitty gritty of how this arc pans out for Moon Knight. Finally - WE HAVE A PERIMETER BREACH! WE HAVE A PERIMETER BREACH! Meanwhile, Taurus becomes the sole survivor of the Zodiac cartel when Jake Fury (masquerading as Scorpio within the group) reveals himself and sets his own team of LMD Zodiacs upon them. Jake reveals that he too is an LMD, and his band of Zodiacs kill all but Taurus, who manages to flee. Taurus approaches the West Coast Avengers, asking for their help to put a stop to Jake Fury and his LMD Zodiac squad. Elsewhere, Bobbi Morse aka Mockingbird, laments not having told Hawkeye the entire truth about her final confrontation with the Phantom Rider and his seemingly innocent fate. The Avengers tussle with the Zodiac at a cattle breeding auction of all places, but are ultimately unsuccessful in capturing them. A short while after, Hawkeye is ambushed by the 12 members and he’s abducted and replaced by an LMD version, unbeknownst to his teammates. Reconvening with the West Coast Avengers, Taurus predicts the Zodiac are to strike a mint much to suspicion of Moon Knight. The White Knight does not entirely trust Taurus but entertains Taurus’ insistence that his assistance with the West Coast Avengers is entirely altruistic. Moon Knight shares a kiss with Tigra, whilst Mockingbird finally confronts (the LMD) Hawkeye about the truth with Phantom Rider. She soon discovers that he isn’t really Hawkeye and with the help of Tigra, they dispose of the android mole. A reveal shows that Tigra, too, is also an LMD - being a SECOND mole planted by the Zodiac to infiltrate the Avengers compound. The Avengers catch up with the Zodiac at the Denver Mint and another battle ensues. The Zodiac are able to escape to Death Valley via the Zodiac Key and Iron Man reveals his knowledge of the Zodiac Key. It essentially thrives on conflict, so when the realm from which it originated no longer satiated it’s needs, it was transported to Earth in search of conflict and sustenance. Moon Knight’s knowledge of astrology is used to confirm that Taurus’ next prediction of where the Zodiac will be, is correct, and with his suspicions eased somewhat the West Coast Avengers head to Death Valley and find what they are looking for - the Zodiac! Tigra reveals herself as an LMD, but the Zodiac and Jae Fury are defeated, only for them all to be resurrected by the Zodiac Key itself. They are all teleported away to the Realm of the Brotherhood, whilst Taurus uses the opportunity to escape. The Zodiac are all incapacitated as there is no Zodiacal energy in the Realm of the Brotherhood, but the Avengers find the real Hawkeye and Tigra and are reunited. They encounter the Brotherhood who wish not to fight them, lest they lose an opportunity to feed off conflict which may occur on Earth. So they return the West Coast Avengers back home, with the warning that they will send the key over to Earth again when they least expect it, to feed off the conflict which the Avengers or others may generate in the years to come. They are brought back, and having discovered Taurus’ escape, Moon Knight vows to track him down! The chase is on, and after a brief encounter with the Shroud, Taurus flees an ever encroaching Moon Knight. Their fight sees them high in the sky as Taurus tries to fly away on a hijacked plane, but he meets his death as the aircraft plummets out of control whilst a falling Moon Knight is saved by Iron man. Hawkeye is livid at the Fist of Khonshu for having not worked as a team, and for the apparent cold blooded murder of Taurus. Finally, after a bit of respite, Simon Williams is abducted by a race of aliens, the SLIGS, who want to analyse Earthly specimens before proposing the invade the planet. The West Coast Avengers come to Simon’s rescue but all are transported to another realm where they are each in turn tested by The Examiner. The Avengers manage to ultimately defeat the aliens and return home. The Sligs manage to send their data to their Supreme One and a Composite Avenger (or artist’s impression) is collated - giving enough reason for the SLigs to call off their plans to invade a planet with clearly formidable inhabitants. He shows up to stop her and get the Darkhold back. Once Aida sends her LMDs to a meeting with General Talbot, with the intention of getting the world to hate Inhumans, the team has to go on the run. But they may have a few surprises for Aida when she catches up with them. Jay and Josh, with Special Agent Tim, make a few deals and and practice their misdirection on the season finale of Agents of S.H.I.E.L.D.- Case Files.
Hello Marvel Universe!With the announcement of the new Guardians of the Galaxy ride at Disneyland (but not Disney World...), John and Jen try to untangle intellectual property rights when it comes to theme park rides. Universal Orlando has Spiderman....but Sony has the movie rights, which they're now sharing with Marvel Studios. Wait, what? We enthuse over concept art for Ragnarok and Black Panther, and remind ourselves that Marvel knows what they're doing, even if the Thor "mockumentary" shorts seem to be at odds with the epic fantasy tone of the series and the dark nature of the Ragnarok plotline. In the episodes "Boom" and "The Man Behind the SHIELD"...yeah. A lot of side plots that don't go anywhere, an extended Philinda flashback, and some head-scratcher moments where character motivations don't really seem to make a lot of sense. But there are also some great fight scenes, some good character moments, and a hell of a cliffhanger. How can Fitz and Simmons be sure that they aren't LMDs too? I guess we'll find out next week...
