Jim Ridl is a great pianist! He's also a composer and bandleader and teacher. I recently had the pleasure of hearing him play at Furman University with the Tim Armacost Quintet. (I just interviewed Tim recently. See Ep 44.) I was particularly struck by Jim's touch and lyricism, by his command of harmony, and by his ability to improvise at length on a theme. He played a beautiful solo piano intro to Wayne Shorter's Infant Eyes that displayed all of these facets of his playing. He also displays great empathy and sensitivity, and strength, as an accompanist and ensemble player. The group also held a workshop for students and faculty. I had the opportunity to talk with Jim briefly and he graciously agreed to this interview. I think you will find some fascinating and insightful comments here, and a good introduction to this great musician. Then he brings us the news of the heart, reading poems from How to Love the World: Poems of Gratitude and Hope. An episode full of inspiration, invitation and devotion--an episode to send you to the canvas or stage or blank page with a renewed sense of what’s at stake. Jack Ridl, Poet Laureate of Douglas, Michigan (Population 1100), in April 2019 released Saint Peter and the Goldfinch. Jack’s Practicing to Walk Like a Heron was awarded the National Gold Medal for poetry by ForeWord Review/IndieFab. His collection Broken Symmetry was co-recipient of The Society of Midland Authors best book of poetry award for 2006. His Losing Season was named the best sports book of the year for 2009 by The Institute for International Sport, and The Boston Globe named it one of the five best books about sports. Jack and his wife Julie founded the visiting writers series at Hope College where he taught for 37 years. The students named him both their Outstanding Professor and Favorite Professor, and in 1996 The Carnegie (CASE) Foundation named him Michigan Professor of the Year. He is the author of several collections of poetry, and has also published more than 300 poems in journals and has work included in numerous anthologies. He has given readings of his work and led workshops at colleges, universities, art colonies and other venues around the country. More than 85 of Ridl's former students are now published authors, and nine of his students appeared in “25 under 25,” in blind judging, edited by Naomi Shihab Nye. Ridl grew up in both the world of basketball where his father was a well-known head coach at Westminster College and the University of Pittsburgh, and the world of the circus, inherited from his mother's family. Ridl lives a short walk from Douglas Beach, arguably the most beautiful of Lake Michigan's disappearing public beaches, with his wife, the writer and artist Julie Ridl, and a few barely domesticated beasts. I dagens avsnitt berättar Emelie om att tävlingarna på Brolöten var tvungna att ställas in på grund av översvämning. Sommarn är verkligen här! Emelie har två hästar klara för Falsterbo och om en vecka börjar Brolötens Ridläger, och det finns fortfarande platser kvar på några av lägren. Victoria njuter av sina sista K1-ritter innan hästarna går på sommarbete. Hon har även svårt att välja namn på sitt föl som för tillfället är lite blygt och planerar många besök för att vänja in sig på människor och fölgos. Veckans fråga handlar om hur man ska tänka kring sitt egna tävlande nu när det inte längre är tävlingsstopp.

Den internationella ögondagen firas den 8 oktober och i samband med den har Synskadades Riksförbunds ögonvårdspris delats ut. I år är det två personer som delar på priset och Radio SRF följer med på prisutdelningarna i Göteborg och Linköping. I september anordnade SRF ett ridläger för barn- och ungdomsmedlemmarna på Örebro Fältrittklubb. Vi hängde med till stallet. Trots att Wilmer Bäckman är nästan blind anser Region Stockholm att han inte behöver färdtjänst. Förbundsjurist Anna Quarnström berättar om fallet som också uppmärksammats av Svenska Dagbladet och vi pratar om varför det blivit svårare för personer med synnedsättning att få färdtjänst.

Jack Ridl was walking with his 7-year-old daughter when she said "with" was the most important word because people always have to be with something, someone, or themselves. When she added that it meant everyone has to makes sure they have a good "with," Jack's perception of the world changed. We were 7- 2. I waited for a brother. My father kept to the hard schedule. Waking the morning of the tenth game, I thought of skipping school and shooting hoops. My cornflakes were ready, soggy. There was a note: "The baby may come today. Get your haircut." We were into January, and the long December snow had turned to slush. The wind was mean. My father was gone. I looked in on my mother still asleep and hoped she'd be OK. I watched her, dreamed her dream: John at forward, me at guard. He'd learn fast. At noon, my father picked me up at the playground. My team was ahead by six. We drove toward the gym. "Mom's OK," he said and tapped his fist against my leg. The Plymouth ship that rode the hood pulled us down the street. "The baby died," he said. I felt my feet press hard against the floorboard. I put my elbow on the door handle, my head on my hand, and watched the town: Kenner's Five and Ten, Walker's Hardware, Jarret's Bakery, Shaffer's Barber Shop, the bank. Dick Green and Carl Stacey waved. "It was a boy." We drove back to school. "You gonna coach tonight?" "Yes." "Mom's OK?" "Yes. She's fine. Sad. But fine. She said for you to grab a sandwich after school. I'll see you at the game. Don't forget about your hair." I got out, walked in late to class. "We're doing geography," Mrs. Wilson said. "Page ninety-seven. The prairie." That night in bed I watched this kid firing in jump shots from everywhere on the court. He'd cut left, I'd feed him a fine pass, he'd hit. I'd dribble down the side, spot him in the corner, thread the ball through a crowd to his soft hands, and he'd loft a star up into the lights where it would pause then gently drop, fall through the cheers and through the net. The game never ended. I fell into sleep. My hair was short. His collection Broken Symmetry (WSUPress) was co-recipient of The Society of Midland Authors best book of poetry award for 2006. His Losing Season (CavanKerry Press) was named the best sports book of the year for 2009 by The Institute for International Sport, and The Boston Globe named it one of the five best books about sports. In 2017 it was developed into a Readers Theater work. Winner of The Gary Gildner Prize for Poetry, Jack has been featured on public radio (“It’s Only a Game with Bill Littlefield,” “The Story with Dick Gordon,” and Garrison Keillor’s “The Writer’s Almanac.”) Then Poet Laureate Billy Collins selected his Against Elegies for The Center for Book Arts Chapbook Award. He read in NYCity with Billy Collins and Sharon Dolin at Christmas after 9/11. He and Peter Schakel are co-authors of Approaching Poetry and Approaching Literature, and editors of 250 Poems, all from Bedford/St. Martin’s Press. With William Olsen he edited Poetry in Michigan in Poetry (New Issues Press). He has done readings in many venues including being invited to read at the international Geraldine R. Dodge Poetry Festival, and was one of twelve people in the arts from around the U.S. invited to the Fetzer Institute for their first conference on compassion and forgiveness. In 2014, Jack received the “Talent Award” from the Literacy Society of West Michigan for his “lifetime of work for poetry literacy,” and The Poetry Society of Michigan named him “Honorary Chancellor,” only the second poet so honored. His poem “Remembering the Night I Dreamed Paul Klee Married the Sky” was selected by Naomi Shihab Nye and featured in The New York Times Sunday Magazine for November 3, 2019. Following the presidential election in 2016 he started the “In Time Project,” each Thursday sending out a commentary and poem. Christian Zaschke, the NYC based U.S. correspondent for the leading German Newspaper Sueddeutsche  Zeitung, wrote a feature about his work. Jack and his wife Julie founded the visiting writers series at Hope College where he taught for 37 years. The students named him both their Outstanding Professor and Favorite Professor, and in 1996 The Carnegie (CASE) Foundation named him Michigan Professor of the Year. Nine of his students are included in the anthology Time You Let Me In: 25 Poets Under 25 edited by Naomi Shihab Nye. More than 90 of Jack’s students have earned an MFA degree and more than 90 are published authors, several of whom have received First Book Awards, national honors. The Gram-negative bacterium Legionella pneumophila naturally parasitises environmental amoebae, but is also able to infect human alveolar macrophages in a mechanistically similar manner. This can result in the mild "Pontiac fever", a flu-like illness, or a potentially lethal pneumonia termed Legionnaires' disease". Crucial for establishing an intracellular replication niche is the Icm/Dot type IV secretion system (T4SS), which translocates approximately 300 different "effector" proteins into the host cell. These substrates enhance uptake efficiency into phagocytes and direct formation of a replication-permissive compartment, called the Legionella-containing vacuole (LCV), and ultimately the egress of the bacteria. Some of the effectors interfere with small GTPases, phosphoinositide metabolism or the ubiquitination machinery, and modulate host cell signalling and vesicle trafficking. We developed a method to isolate intact LCVs by using immuno-magnetic separation with an LCV-specific antibody followed by density gradient centrifugation. Proteomic analysis of the purified phagosomes together with findings of previous studies showed, that the vacuoles harbour markers of the endosomal network, associate with mitochondria, early secretory vesicles and the endoplasmic reticulum, but avoid fusion with lysosomes. Our investigations of the novel L. pneumophila effector RidL revealed that the LCV also communicates with the retrograde vesicle trafficking pathway of infected cells. This pathway recycles amongst others acid-hydrolase receptors, such as the cation-independent mannose 6-phosphate receptor (CIMPR), from the tubular endosomal network back to the trans-Golgi. This transport requires the multiprotein "retromer" complex, which consists of two major subunits: the heterotrimeric cargo-selective subcomplex comprising the proteins Vps26, Vps29 and Vps35 and the membrane-deforming heterodimeric subcomplex composed of any combination of the phosphoinositide (PI)-binding sorting nexins SNX1 or SNX2 plus SNX5 or SNX6. Pull-down experiments with lysates of RAW 264.7 macrophages or D. discoideum amoebae revealed Vps26, Vps29 and Vps35 to be retained by the then uncharacterised protein RidL, which represented an intriguing, novel effector interaction. Like most T4SS substrate mutants, L. pneumophila lacking ridL showed no phenotype for growth in liquid AYE medium and uptake into phagocytes compared to wild-type bacteria. However, intracellular replication was strongly impaired for the mutant strain in several host cell lines. RidL is preferentially expressed in the late post-exponential growth phase and translocated in an T4SS-dependent manner at early time-points of the infection, suggesting a role shortly after the uptake of the bacteria. The effector exhibited a bipolar localisation on the LCV membrane, but upon overexpression the protein covered the entire vacuole. Interestingly, RidL bound the lipid phosphatidylinositol 3-phosphate (PtdIns(3)P), a known eukaryotic endosomal membrane anchor, and also specifically bound to the retromer subunit Vps29. Although the protein had no effect on the acquisition of Vps26, Vps29 and Vps35, the percentage of LCVs positive for the retrograde cargo receptors CIMPR or sortilin was reduced in presence of RidL, suggesting interference with the retrograde transport pathway. Furthermore, significantly less SNX1- and SNX2-positive LCVs were detected in cells infected with wild-type L. pneumophila compared to the ridL mutant strain. Moreover, RidL competed with SNX1 for binding at PtdIns(3)P-positive membranes. To directly examine the influence of RidL on retrograde trafficking, the retromer-dependent transport of cholera and Shiga toxin inside cells was analysed in macrophages infected with wild-type or ridL L. pneumophila, and in HeLa cells ectopically producing RidL, respectively. In both cases, the trafficking was inhibited by RidL, and for cholera toxin the transport was arrested at the endosomal stage. In line with these findings, siRNA knockdown experiments revealed that a functional retrograde pathway restricted intracellular growth of L. pneumophila. Taken together, we postulate that RidL (Retromer interactor decorating LCVs) inhibits retrograde trafficking at endosomes by binding to the retromer subunit Vps26 and/or by competition with sorting nexins, thus promoting intracellular replication of L. pneumophila. Collectively, the results obtained in this thesis shed light on the host factor composition of LCVs and provide mechanistic insights into a novel L. pneumophila effector protein.

Bettan går till tandläkaren och ojar sig som vanligt över tonåringars svårighet att navigera mellan alla olika val de har. Ridläger eller anusblekning?
