L., Valdes, P., Huang, D., Caflisch, A., Aguzzi, A. Abstract: The cellular prion protein PrPC mediates the neurotoxicity of prions and other protein aggregates through poorly understood mechanisms. Antibody-derived ligands against the globular domain of PrPC (GDL) can also initiate neurotoxicity by inducing an intramolecular R208-H140 hydrogen bond (H-latch) between the alpha2-alpha2 and beta2-alpha2 loops of PrPC. Importantly, GDL that suppress the H-latch prolong the life of prion-infected mice, suggesting that GDL toxicity and prion infections exploit convergent pathways. To define the structural underpinnings of these phenomena, we transduced nineteen individual PrPC variants to PrPC-deficient cerebellar organotypic cultured slices using adenovirus-associated viral vectors (AAV). We report that GDL toxicity requires a single N-proximal cationic residue (K27 or R27) within PrPC. Alanine substitution of K27 also prevented the toxicity of PrPC mutants that induce Shmerling syndrome, a neurodegenerative disease that is suppressed by co-expression of wild-type PrPC. People with anxiety often close off their Crown and Upper Chakras and feel disconnected from Source.  This Chakra also feeds energy to all other chakras in the body. Step 7: Massage the thumbs and fingers with Peace and Calming essential oil blend. while saying or thinking the affirmation, “I am real.”This is to release victim consciousness and be self-responsible and empowered to make choices, which can be needed for people feeling trapped, unsafe or anxious. Step 8: Massage the middle of the palms again,  while spraying a spritzer of Lavender essential oil, and say, “I learn from all of life's experiences, I learn from all of life's experiences, I learn from all of life's experiences,"  and then, "From all of life's experiences I learn, from all of life's experiences I learn, from all of life's experiences I learn."  This addresses stress, cramping, and tension in the abdominal region, which often manifests when people have anxiety. Step 9: Go to the K27 points in the upper chest, just at the indentation lateral to where the clavicle and the sternum meet. Tap or make finger circles at the Elegant Mansion or K27 points. Working this area balances the overall energy in the body. These points strengthen immune function and relieve breathing difficulties, chest congestion, asthma, coughing, anxiety and depression. I want to talk with you today about aging. Aging is something we do from the moment we come into this world. When we are really little we just can't wait until we are older. There are so many things that we are looking forward to and time seems to stand still when you are young. I remember as a kid, when I got 30-minutes to go play, it felt like forever! Well, these days, 30-minutes seems to go by pretty darned quick. There are other things that come along with aging, as you get in the older years of aging that is. Maybe you start to notice you have more body aches and pains. I know I have, riding a horse isn't as easy as it used to be, but it is still really fun. Maybe you have less energy and find you need to have your siestas mapped out for yourself. Or, maybe you are noticing you have more wrinkles, or grey hairs, or maybe you dye it. There can be so many different things that change in our lifetime. You know, what is possible, when we are able to stay in relationship with ourselves, as well as with others, we can learn to listen to our body and honor our bodies wisdom. To rest when you need to rest, to be intentional with your exercise and your diet. To really notice, how does it serve your body? Sometimes things might taste really great, yet, it's important to be able to know how whatever you ingest is going to impact your body in the long run. In a sense we become what we eat, and it truly does impact us. I know I have found it to be so important to have good, clear water, lots of water. Not pure, because purity would remove the minerals that are important for us to have. We are actually made up of mostly water. What is helping me a lot is to be very intentional when I wake up. There is something that I do that Donna Eden recommends, she offers Eden Energy Medicine, which is starting my day with the 4 thumps. You start by thumping the points right under your eyes, I use three fingers to be sure I'm tapping the meridian point. This helps to wake our system up. The next place you tap in on the K27, this is located right underneath where your collar bone is. Tap with some good force. The next is your thymus, which is right on your sternum, this supports your immune system. I know when I caught a cold and it took a couple of months for my system to clear it out. So, I became intentional with the energy medicine in order to boost my immune system. The last place is your spleen points, under the center of your breasts. The spleen is important to flush the toxins out of your body. Just doing that I feel so much more energized and alive and alert. I do that before I get out off bed in the morning, and it makes such a difference for the rest of my day. Notice, what do you like doing, what do you really enjoy? Begin to set things out for yourself to look forward to. One of the things I discovered I wanted to be able to do for myself was to let myself start singing again. So, practicing that for 15 minutes every day, just because I like to sing. Then maybe I'll sing for 20 minutes the next week, or maybe for 25, but to start to build and grow that which I really enjoy. To cultivate that in this world. I encourage you to recognize the direction that you would like to move in with more and more expansiveness, and then give yourself permission to exist in whatever it is that you are doing, or however you are being. The practice of acknowledging what is, is a powerful one. Be kind to yourself today. Thanks for being on this journey with me today. Startup company Trojan Energy is installing 200 of its chargers across Brent and Camden. Each charge point is slotted into the ground with a flat and flush connection. The technology has no permanent footprint or street clutter as the hardware is only visible when a vehicle is charging." according to Electric Vehicle Research: "The technology consists of 2 parts - a charge point slotted into the ground, and a 'lance' which is inserted into the charge point in order to charge. A drivable prototype is to be available shortly." says electrive: "JLR was among the carmakers that benefitted from £73.5 million in funding through the Advanced Propulsion Centre that the British government announced in June. In the case of Jaguar Land Rover, the company is using the money to further project Zeus to develop fuel cell technology. Autocar now reports that progress is being made as the first fuel cell concept to emerge from this project will probably be an SUV the size of the Range Rover Evoque. The next-generation Range Rover Evoque’s arrival is expected in the middle of the 2020s. It's nothing spectacular, but there's certainly interest." says CNET: "The company said Friday it has 7,062 reservations for the Ocean. Compared to recent news surrounding preorders (I'm thinking Tesla Cybertruck and Ford Bronco) it's a drop in the bucket. However, it goes to show there is an audience for Fisker's SUV with oodles of sustainable materials inside. The reservations come from 30 countries and Fisker said the figure virtually sells out the first planned production run in late 2022. Additionally, Fisker talked about a second SUV for the first time it dubbed a "sports crossover." The image shows a swoopy design sort of reminiscent of the Volkswagen ID 4. Sounds like a big problem." according to Jalopnik: "This could be a huge problem for Fisker Inc., as it’s already advertised and sold deposits for a vehicle with certain projected performance—deposits it accepted without even knowing for certain what the fundamental hardware of the vehicle would be. Any major changes from here could mean major changes to the Ocean as advertised. We are committed to taking any actions that advance our objective of delivering the Ocean as early as the end of 2022.”"   TESLA RELEASES Q2 2020 SAFETY REPORT "In the 2nd quarter, we registered one accident for every 4.53 million miles driven in which drivers had Autopilot engaged. For those driving without Autopilot but with our active safety features, we registered one accident for every 2.27 million miles driven. For those driving without Autopilot and without our active safety features, we registered one accident for every 1.56 million miles driven. By comparison, NHTSA’s most recent data shows that in the United States there is an automobile crash every 479,000 miles."   In comparison, Tesla had an accident on Autopilot every 3.27 million miles in Q2 2019.   AMERICA’S CHEAPEST ELECTRIC VEHICLES ARE COMING COURTESY OF CHINESE AUTOMAKER KANDI "Chinese electric vehicle and parts manufacturer Kandi Technologies Group  is officially bringing two EVs to the United States through its subsidiary Kandi America — news that has prompted run up in its share price in the past day. " says TEch Crunch: "Kandi Technologies has been talking about bringing EVs to the United States for a couple of years. Now, two models are arriving as early as the end of 2020, beginning in a limited area in Texas. Both are priced under $30,000 before federal incentives. The two models heading to the U.S. are Kandi’s compact K27 vehicle that comes with a 17.69 kWh battery, The larger K23 is also coming to the U.S. market. This small electric SUV has a 41.4 kWh battery and a driving range of more than 180 miles. In most eukaryotes the histone methyltransferases SU(VAR)3-9, G9a and their orthologues play major roles in transcriptional regulation. Histone H3 lysine 9 methylation is associated to transcriptional silencing in vivo. SU(VAR)3-9 is the main H3K9 HMTase in Drosophila heterochromatin whereas G9a was found to be an euchromatic H3K9 methyltransferase in mammalian cells. In this work SU(VAR)3-9 and a new HMTase homologous to G9a were characterized in vitro. A detailed analysis of the reaction products shows that recombinant SU(VAR)3-9 adds three methylgroups to full-length H3 and only two methylgroups to an H3-tail peptide. The transfer of two methylgroups to an unmethylated H3-tail peptide is achieved in a nonprocessive manner. The full-length enzyme elutes with an apparent molecular weight of 160 kDa from a gel filtration column, which indicates the formation of a dimer. The N-terminus was shown to be required for this dimerisation and to retrieve full activity in vitro. We show that the interaction occurs by domain swapping of two motifs within the N-terminus. The fact that the N-terminus is responsible for a concentration dependent dimerisation of SU(VAR)3-9 may indicate a role for this domain in the dosage-dependent effect on position effect variegation. Drosophila G9a adds three methyl groups to unmethylated H3 in vitro as has been described for mouse G9a. In vitro, a N-terminal truncation of dG9a adds three methylgroups toward H3K9 and K27, with a preference for K9. Surprisingly, dG9a also methylates H4 with specificity for K8, K12 or K16. In vivo, dG9a is present in complexes with a molecular mass of 440-670 kDa and we show that it specifically interacts with the histone deacetylase Rpd3. HP1a is predominantly associated with centromeric heterochromatin in Drosophila. Supporting the histone code hypothesis, the chromo domain of HP1 recognises and binds H3K9 methylated peptides. Here we show the mechanism for binding to H3K9Me chromatin by recombinant Drosophila HP1a. HP1a requires a bimodal interaction of the chromo domain with H3K9Me and a simultaneous interaction of the chromo shadow domain with auxiliary factors (SU(VAR)3-9 and ACF) for stable association with H3K9Me chromatin. The two HP1 paralogs HP1a and HP1c bind to distinct chromatin structures and we identify distinct interaction partners for these two proteins.

In der vorliegenden Arbeit wurden die beiden Histon-Methyltransferasen Su(var)3-9 und E(Z) aus Drosophila melanogaster charakterisiert. Die Histonmethylierung als Modifikation war schon länger bekannt gewesen, bis zum Jahr 2000 war jedoch vor allem die Acetylierung etwas genauer untersucht worden. Su(var)3-9 war die einzige bekannte Histon-Lysin-Methyltransferase, als diese Arbeit begonnen wurde. Zur Charakterisierung wurde das myc-getagte Enzym aus Drosophila-Kernextrakt durch Affinitätschromatographie aufgereinigt und zunächst die Substratspezifität festgestellt. Wie das humane Enzym Suv39H1 methyliert es ebenfalls spezifisch H3-K9 (Lysin 9 im Histon H3). Das aus den Kernextrakten aufgereinigte Enzym besitzt aber auch die Fähigkeit, ein an H3-K9 präacetyliertes Substrat zu methylieren. Die Vermutung, dass Su(var)3-9 mit einer Histondeacetylase assoziiert ist, konnte durch Verwendung von TSA als HDAC-Inhibitor bestätigt werden. Es stellte sich heraus, dass HDAC1 (Rpd3) mit Su(var)3-9 assoziiert ist. Um das Enzym besser untersuchen zu können, wurde es als Volllängenprotein und als Deletionsmutante in E. coli exprimiert. Die Aufreinigung des rekombinanten Enzyms sowie seine Lagerbedingungen wurden optimiert. Das Volllängenprotein Su(var)3-9 liegt – wie durch Gelfiltration festgestellt - als Dimer vor, die Interaktion mit sich selbst ist über den N-Terminus vermittelt. Su(var)3-9 bindet an sein eigenes, bereits methyliertes Substrat. Dies wurde an Peptiden untersucht, die den ersten 20 Aminosäuren des Histons H3 entsprechen, und entweder an Lysin 9 dimethyliert oder unmodifiziert waren. Die Interaktion mit dem methylierten Substrat ist auf die Chromodomäne von Su(var)3-9 zurückzuführen, ist jedoch schwächer als die Wechselwirkung von HP1 mit methyliertem H3-K9. Des weiteren wurde eine Drosophila-Zelllinie stabil mit Su(var)3-9 transfiziert. Das überexprimierte Protein ist jedoch nur schwach aktiv. Die Tatsachen, dass Su(var)3-9 mit HDAC1 interagiert sowie mit seinem eigenen Substrat assoziiert, ermöglichen die Aufstellung von Hypothesen über die bis jetzt kaum erhellte Ausbreitung von Heterochromatin in euchromatische Bereiche. Durch die Wechselwirkung mit der Deacetylase könnte Su(var)3-9 auch in aktiv transkribierte Bereiche vordringen und diese methylieren. Die Acetylierung, Zeichen für aktive Transkription, würde durch die Methylierung ersetzt werden. Die Interaktion mit seinem umgesetzten Substrat könnte verhindern, dass das Enzym sich nach der Reaktion entfernt, vielmehr könnte Su(var)3-9 entlang eines DNA-Stranges sukzessive alle Nukleosomen methylieren. Die darauffolgende Bindung von HP1 an methyliertes H3-K9 könnte den heterochromatischen Charakter des Chromatins verstärken und für längere Zeit festlegen. Aus Drosophila-Kernextrakten gelang es weiterhin, den E(Z)/ESC-Komplex über Säulenchromatographie aufzureinigen. Dieser enthält neben E(Z), ESC, p55 und Rpd3 auch Su(z)12. E(Z), ESC und Su(z)12 gehören der Polycomb-Gruppe an. Deren Funktion ist die dauerhafte Repression der homöotischen Gene. Sie spielen daher eine wichtige Rolle im „Zellgedächtnis“ während der frühen Entwicklung von Drosophila. Es konnte gezeigt werden, dass der E(Z)/ESC-Komplex Lysin 9 sowie Lysin 27 im Histon H3 methyliert. Außerdem wurde in vitro ein Teilkomplex aus rekombinantem E(Z), p55 und ESC rekonstituiert, der das Histon H3 methylieren kann. Ein Teilkomplex, der E(Z) mit mutierter SET-Domäne enthält, ist nicht in der Lage, H3 zu methylieren. Die Vorhersage, dass E(Z) aufgrund seiner SET-Domäne eine Methyltransferase sein müsse, konnte durch vorliegende Untersuchungen bestätigt werden. Polycomb ist ein weiteres Protein aus der Polycomb-Gruppe. In dieser Arbeit konnte gezeigt werden, dass dieses Protein spezifisch an das Histon H3 bindet, das an K27 trimethyliert ist. Polycomb besitzt wie HP1 eine Chromodomäne. Aus den vorliegenden Daten kann folgendes Modell aufgestellt werden: Nach der Methylierung von H3-K9 sowie H3-K27 durch den E(Z)/ESC-Komplex in homöotischen Genen, die schon abgeschaltet sind und weiterhin reprimiert werden müssen, bindet Polycomb an dieses Methylierungsmuster. Polycomb befindet sich in einem großen Komplex mit weiteren Polycomb-Gruppen-Proteinen. Die Bindung dieses Komplexes an Chromatin könnte ein denkbarer Mechanismus sein, wie die dauerhafte Repression der homöotischen Gene vermittelt wird. Um den E(Z)/ESC-Komplex genauer untersuchen zu können, wurden Viren für das Baculosystem hergestellt, so dass eine Einzel- oder auch Coexpression der Proteine möglich ist. Die Aktivität von E(Z), das im Baculosystem exprimiert wurde, ist nicht besonders hoch. Es bindet unter den in dieser Arbeit verwendeten Bedingungen weder an DNA, noch an Histone noch an H3-Peptide, die methyliert sind. Innerhalb des E(Z)/ESC-Komplexes bindet E(Z) an p55, Rpd3, ESC sowie Su(z)12. Su(z)12 interagiert mit p55, Rpd3 und E(Z). Die weiteren Interaktionen werden am besten durch eine bildliche Darstellung (siehe Abb. 86) vermittelt. In einem Luciferase-Assay wurde eine repressive Wirkung von E(Z) festgestellt. Dieses Experiment bedarf allerdings eines aktivierten Systems. Ferner muss durch Mutationsanalysen sichergestellt werden, dass die repressive Wirkung auf die Methyltransferase-Aktivität von E(Z) zurückzuführen ist. Kürzlich wurde entdeckt, dass E(Z) sowie Su(z)12 in verschiedenen Tumoren überexprimiert sind. Noch ist weder deren Funktion in den Tumorzellen klar, noch weiss man, ob die Überexpression der Grund oder eine Folge der Tumorbildung ist, noch kennt man alle Zielgene, die durch eine Überexpression von E(Z) und Su(z)12 beeinflusst werden. In nächster Zeit sind hier Einsichten in die Wirkungsweise von E(Z), Su(z)12 und anderen Polycomb-Gruppen-Proteinen zu erwarten.
