Abstract: Consolidation of recent memory depends on hippocampal activities during resting periods that immediately follows the memory encoding. There, Slow Save Sleep phases appear as privileged periods for memory consolidation as hosting the ripple activities, which are fast oscillations generated within the hippocampus whose inactivation leads to memory impairment. If a strong correlation exists between these replays of recent experience and the persistence of behavioural adaptations, the mobilisation, the localization and the importance of synaptic plasticity events in this process is largely unknown. To question this issue, we used cell-surface AMPAR immobilisation to block post-synaptic LTP within the hippocampal region at various steps of the memory process. 1- Our results show that hippocampal synaptic plasticity is engaged during the consolidation but is dispensable during the encoding or recall of a working memory based spatial memory task. 2- Blockade of plasticity during sleep leads to apparent forgetting of the encoded rule. 3- In vivo recordings of ripple activities during resting periods show a strong impact of AMPAR immobilization solely, prominent when a rule has been recently encoded. 4- In situ examination of the interplay between AMPAR mobility, hippocampal plasticity and spontaneous ripple activities pointed that post-synaptic plasticity at CA3-CA3 recurrent synapses support ripple generation. As crucial results were reproduced using another AMPARM blockade strategy, we propose that after rule encoding, post-synaptic AMPAR mobility at CA3 recurrent synapses support the generation of ripples necessary for rule consolidation. Abstract: The value of rewards and punishments - namely, how good or bad they are perceived - guides approach or avoidance behaviors. Valence refers to the negative or positive "sign" of the state elicited by an event, whereas salience refers to the amount of attention an event attracts, disregarding its valence. While identifying these signals conveys critical information for understanding circuits involved in emotional processing, they are often confounded due to their underlying correlation. Moreover, whereas the study of the neural basis of value coding has been intensively investigated in the appetitive domain, the neural substrates for how aversive values are established for different threat intensities and guide defensive behavior have yet to be discovered. The dorsomedial prefrontal cortex (dmPFC) is a key region in the control of defensive actions, although how different aversive values are encoded at the neuronal level within this region and drive defensive behaviors remains unknown. Here, we developed an instrumental approach/avoidance task in mice that, by matching motivational salience levels elicited by cues predicting rewards or punishments, allows univocally disentangling the presence of either salience, valence, or value coding from brain signals. We performed freely moving large neuronal population calcium imaging in the dmPFC of mice performing our task, conducting appetitive/aversive outcome devaluation/revaluation behavioral tests. We found that, while a similar fraction of single neurons decoded valence and value information, and only a minor fraction decoded salience, value coding was observed at the neuronal population level. Moreover, different value representations of the same valence lay within similar subspaces of the neural state space while values of opposed valence were encoded in orthogonal subspaces, unveiling how the brain stores associative appetitive and aversive information in medial prefrontal networks. Florence Herry  auparavant infirmière, et son mari, désormais associé, lancent Libheros en 2017. Pour répondre à cette problématique des soins à domicile post-hospitalisation, ils développent une plateforme d'intermédiation pour proposer à l'offre et à la demande de se rencontrer. En 2018, ils font le constat que le modèle BtoC à ses limites. Pourtant, en un an, la startup a su créer un réseau national de professionnels de santé qualifiés et des solutions tech pour répondre au marché. La valeur est là, mais le modèle est à revoir. Le pivot BtoB est amorcé. Les premiers contrats importants sont signés avec hôpitaux, entreprises et labo pharmaceutiques. Solution tech, offre, réseau de soignants, tout est déjà en place lorsque la vague Covid déferle. Jusque dans les aéroports, on recherche des infirmières pour prendre la température des voyageurs ou leur délivrer des tests PCR.
