Abstract: The critical brain hypothesis states that the brain can benefit from operating close to a second-order phase transition. While it has been shown that several computational aspects of sensory information processing (e.g., sensitivity to input) are optimal in this regime, it is still unclear whether these computational benefits of criticality can be leveraged by neural systems performing behaviorally relevant computations. To address this question, we investigate signatures of criticality in networks optimized to perform efficient encoding of stimuli. We consider a spike-coding network of leaky integrate-and-fire neurons with synaptic transmission delays and input noise. Previously, it was shown that the performance of such networks varies non-monotonically with the noise amplitude. Interestingly, we find that in the vicinity of the optimal noise level for efficient coding, the network dynamics exhibits signatures of criticality, namely, the distribution of avalanche sizes follows a power law. When the noise amplitude is too low or too high for efficient coding, the network appears either super-critical or sub-critical, respectively. Our work suggests that two influential, and previously disparate theories of neural processing optimization - efficient coding, and criticality - may be intimately related. Our guest today is Dr. Christopher Logothetis, one of the nation's foremost experts on prostate cancer. Chris has spent nearly five decades at MD Anderson in Houston developing therapies for prostate cancer as well as conducting research into the underlying biology of the disease. Aside from skin cancers, prostate cancer is the most common cancer among men, claiming a man's life every 15 minutes in the United States, according to the Prostrate Cancer Foundation. Since the 1970s when Chris joined the staff at MD Andersen, which is the nation's top-ranked hospital for cancer care, he has been dedicated to the treatment, research, and prevention of genitourinary cancers such bladder, kidney, testes and penis cancer. For the past 25 years, he has focused primarily on prostate cancer and the development of effective chemotherapy treatments. Today, Chris is the director of MD Anderson's Genitourinary Cancer Center and the director of the Prostate Cancer Research Program. [00:03:43] Ken asks Chris when he first became interested in science. [00:04:09] Dawn asks if there were a particular teacher or class that prompted Chris' decision to pursue medicine. [00:05:10] Dawn mentions that in the ‘60s and ‘70s, talking about cancer was almost taboo and asks Chris to talk about the stigma that surrounded cancer for quite some time. [00:05:57] Ken asks if Chris knew he wanted to specialize in cancers when he first started medical school in Athens or if that interest developed later. [00:07:06] Dawn mentions that Chris graduated from medical school in 1974 and then took off for Chicago where he had an internship at Cook County Hospital. Dawn asks about the experience, and if it were a culture shock to go from Athens, Greece to Chicago in the 1970s. [00:08:54] Dawn asks what took Chris to Texas and MD Anderson after his time in Chicago. [00:09:36] Dawn mentions that after Chris finished his fellowship, he joined the faculty at MD Anderson, and is now coming upon his 50th anniversary there. [00:09:51] Chris explains his view that we need to better understand the drivers of cancer and goes on to talk about what we currently know about these drivers. [00:12:06] Ken asks about the significance of the Human Genome Project on cancer research. [00:13:49] Dawn mentions that along with new technologies, there evolved a strategy of what is called co-clinical investigation where researchers study the mouse, but in parallel look at the difference and similarities with humans. She asks him about how that integrated data required a new language to bring it all together, which is now known as Prometheus. Dawn asks Chris to talk about Prometheus and how this has led to an accelerated understanding of cancer biology. [00:20:47] Dawn mentions that Chris has studied a range of genitourinary cancers throughout his career, such as germ cell tumors, bladder, and renal cancers, but that his interest in prostate cancer is a more recent development. Dawn asks what led to this specific interest. [00:23:12] Dawn explains that metastatic cancer was first cured in 1956 when methotrexate was used to treat a rare tumor called choriocarcinoma. She goes on to say that since then, chemotherapy drugs have been used to treat mixed germ-cell tumors and has led to dramatically improved survivorship among patients with metastatic germ-cell tumors. She also mentions that in 1982 Chris published a paper in the journal Cancer titled, “The growing teratoma syndrome,” at which time, tumor growth following chemotherapy for mixed germ-cell tumors had been considered a reliable indicator of a persistent active carcinoma, with the rule being that if the cancer didn't respond to treatment that operations were fu...

Abstract: The insula has been classically divided into vast granular, dysgranular and agranular sectors. Over the years, several distinct studies proposed subdivisions of these sectors, with however no consensus. We recently proposed a cyto- and myelo-architectonic partition in which each sector contained sharply delimited areas (Evrard et al. 2014 J Comp Neurol 522: 64-97). Some of these areas were further divided into distinct subareas with obvious functional implications. Here, we examined the spatial relationship between architectonic boundaries and tract-tracing labeling in the insula in the macaque monkey. Injections of neuronal tracers in distinct areas of the prefrontal or anterior cingulate cortices produced heterogeneous and discontinuous patterns of anterograde and retrograde labeling in the insula. These patterns were made of sharply delimited patches forming anteroposterior stripes across consecutive coronal sections. While the overall pattern of labeling varied with the injection site, the patches systematically coincided with specific architectonic subareas, particularly in the dysgranular insula. This unequivocally validates our prior architectonic partition and strongly supports the idea of a refined modular Bauplan of the primate insula. This modular organization may underlie a serial stream of integration of interoception with self-agency and social activities across distinct insulo-prefrontal processing units that need to be explored. Abstract: Digitized neuroanatomical atlases are crucial for localizing brain structures and analyzing functional networks identified by magnetic resonance imaging (MRI). To aid in MRI data analysis, we have created a comprehensive parcellation of the rhesus macaque subcortex using a high-resolution ex vivo structural imaging scan. The structural scan and its parcellation were warped to the updated NIMH Macaque Template (NMT v2), an in vivo population template, where the parcellation was refined to produce the Subcortical Atlas of the Rhesus Macaque (SARM). The subcortical parcellation and nomenclature reflect those of the 4th edition of the Rhesus Monkey Brain in Stereotaxic Coordinates (RMBSC4; Paxinos et al., in preparation). The SARM features six parcellation levels, arranged hierarchically from fine regions-of-interest (ROIs) to broader composite regions, suited for fMRI studies. As a test, we ran a functional localizer for the dorsal lateral geniculate (DLG) nucleus in three macaques and found significant fMRI activation in this atlas region. The SARM has been made openly available to the neuroimaging community and can easily be used with common MR data processing software, such as AFNI, where the atlas can be embedded into the software alongside cortical macaque atlases. Abstract: The brainstem noradrenergic locus coeruleus (LC) is reciprocally connected with the prefrontal cortex (PFC). Strong coupling between LC spiking and depolarizing phase of slow (1 - 2 Hz) waves in the PFC field potentials during sleep and anesthesia suggests that the LC drives cortical state transition. Reciprocal LC-PFC connectivity should also allow interactions in the opposing (top-down) direction, but prior work has only studied prefrontal control over LC activity using direct electrical (or optogenetic) stimulation paradigms. Here, we describe the physiological characteristics of naturally occurring top-down prefrontal-coerulear interactions. Specifically, we recorded LC multi-unit activity (MUA) simultaneously with PFC single unit and local field potential (LFP) activity in urethane-anesthetized rats. We observed cross-regional coupling between the phase of ~5 Hz oscillations in LC population spike rate and the power of PFC LFP oscillations within the high Gamma (hGamma) range (60 - 200 Hz). Specifically, transient increases in PFC hGamma power preceded peaks in the ~5 Hz LC-MUA oscillation. Analysis of cross-regional transfer entropy demonstrated that the PFC hGamma transients were predictive of a transient increase in LC-MUA. A ~29 msec delay between these signals was consistent with the conduction velocity from the PFC to the LC. Finally, we showed that PFC hGamma transients are associated with synchronized spiking of a subset (27%) of PFC single units. Our data suggest that, PFC hGamma transients may indicate the timing of the top-down excitatory input to LC, at least under conditions when LC neuronal population activity fluctuates rhythmically at ~5 Hz. Synchronized PFC neuronal spiking that occurs during hGamma transients may provide a previously unknown mode of top-down control over the LC. Copy rights belong to original authors. I feel a great sense of gratitude to have had a chance to interview Leon Logothetis in the episode you are about to hear. Leon is a global adventurer, motivational speaker & philanthropist but It wasn’t always that way. He used to be a broker in the city of London where he felt uninspired and chronically depressed but he gave it all up for a life on the road. This radical life change was sparked by the inspirational movie The Motorcycle Diaries Leon recently returned from filming the second season of his Netflix TV show The Kindness Diaries. In season one he circumnavigated the globe on his vintage yellow motorbike named Kindness One, and in season two he drives a vintage yellow Volkswagen Beetle convertible which he names Kindness 2 all the way from Anchorage, Alaska down to Argentina. Along the way in both season 1 and 2, Leon gives a life changing gift to an unsuspecting good Samaritan in each episode. All of this while relying on the kindness of strangers to get him through each day. Leon is no stranger to adventure. He teamed up with First Book® & drove a car from London to Mongolia raising money to buy 10,000 books for underprivileged children in America. He also drove a vintage London taxi across America, giving free cab rides to the needy and working with Classwish to bring hope back to the schools of America. Leon has documented his travels for numerous media outlets including Good Morning America, Los Angeles Times, San Francisco Chronicle, THE BBC, FOX NEWS, CNN , Psychology Today, and The New York Times. Leon just released his 5th book which is entitled Go Be Kind This book isn’t just a journal or another how-to guide to getting what you want. Formerly a London broker, he gave up his comfortable life to travel and find real human connection. Born in London England, of Greek descent, Logothetis now lives in Los Angeles California. He was educated in London and graduated from Babson College in Boston Massachusetts with a Business degree. After graduating college, Logothetis was a successful broker. But his real education started in the school of life—traveling the world. He had a radical life change after stumbling upon an inspirational movie, The Motorcycle Diaries, which chronicled the story of Che Guevara. On Che’s journey, he crossed South America relying solely on the kindness of strangers, and his story lit up Logothetis' mundane existence. Alain Moussi has enjoyed a rapid rise from stuntman to starring roles to leading his own martial arts movie franchise.
