Pedro Sánchez, ha anunciado su decisión de continuar al frente del Ejecutivo después de cinco días de reflexión sobre su continuidad. El presidente del Gobierno ha sido entrevistado en el TD2 de TVE para abordar esta cuestión. Esta es la primera entrevista que concede después de su anuncio. Pedro Sánchez, en su entrevista en TD2, ha llamado a la reflexión tanto a fuerzas políticas como a medios de comunicación para la regeneración de la democracia. "O estás con él o estás contra él, y no cabe nada más. Esa es la visión que tiene el señor Sánchez de la democracia", ha declarado la portavoz de VOX sobre las palabras del presidente del Gobierno. VOX se ha personado como acusación popular en el juzgado que investiga a Begoña Gómez. Pepa Millán ha asegurado que en su partido van a "llegar a las últimas consecuencias", pero que "dudan que se vayan a depurar responsabilidades políticas". "Exigimos que se esclarezca, por eso hemos acudido a los tribunales. No solo con Begoña Gómez, también con Illa", ha insistido. En 24 horas de RNE, Sara Bailac, portavoz de ERC en el Senado, ha reaccionado a esta entrevista: "Han faltado respuestas y aclaraciones sobre este parón de cinco días (…) Tampoco ha explicado cuáles van a ser sus propuestas para resolver esta situación que él denuncia". "ERC ha hecho una apuesta por la resolución del conflicto político entre Cataluña y el Estado español, hemos situado la negociación en el centro y esta vía ha dado frutos", ha afirmado la portavoz de ERC. Sara Bailac ha subrayado que esta estrategia de Pedro Sánchez es una manipulación a la ciudadanía: "Nos parece que lo que ha pasado estos cinco días ha sido manipular esa empatía que podría sentir mucha gente en Cataluña respecto a los ataques de la derecha y de la extrema derecha". Setting off early in his days to roam the world his experience was cut short due to lack of funds. After he spent some money buying a lot of type three parts posting them for sale in the magazine advert Paul came back to an answering machine full of request for parts. Asking his friend Ivan to use the name type three detectives Paul then kicked off a successful business supplying NOS type three parts in the early days of his career. Seeing the change evolve into lots of buses being put back on the road it was only natural that the transition was made to type two detectives. It's good to hear the stories from the enthusiast that turn their hobby into a career and the path he took to do it. Engineer and artist Andrew Johnson talks about his engineering work and his artistic practice, blurring the lines between both, at the intersection of engineering's crisp mathematics and physical materiality, and art, with its unbounded possibilities, creative expression, and playfulness.Johnson is an artist and engineer born and raised in Nebraska. He received an MAE degree in structural engineering from the University of Nebraska in 2006. He is a self-taught artist and works in a variety of media. Johnson has shown work throughout the metropolitan Omaha area, most notably at the Bemis Center for Contemporary Arts and The Union for Contemporary Art. As a practicing engineer working at TD2, Johnson works on a variety of project types and market sectors throughout the US, focusing primarily on local and regional work. He enjoys close collaboration with architects and designers and has worked on several award-winning designs. Johnson has extensive experience with structural designs for art and performance spaces, including the Jackson Dinsdale Art Center at Hastings College, Hoff Family Arts and Culture Center in Council Bluffs, The Union for Contemporary Art campus, Omaha Benson Theater, and, most recently, the new addition to the Joslyn Art Museum.

In dieser Arbeit wurde die zelluläre Cholesterin-Homöostase in menschlichen Zellen von Patienten mit der seltenen Tangier-Erbkrankheit (TD) untersucht. Diese Patienten besitzen verschiedene Mutationen im ABCA1-Gen, welches eine zentrale Rolle im Cholesterin-Export spielt, und haben aufgrund des niedrigen bzw. fehlenden HDL im Plasma ein erhöhtes Risiko, kardiovaskuläre Erkrankungen auszuprägen. Es ist jedoch unklar, wieso es trotz der relativ einheitlichen HDL-Defizienz zu völlig unterschiedlichen Identitätsmustern der Arteriosklerose kommt. Ziel der vorgelegten Arbeit war es, Auswirkungen des Funktionsverlustes von ABCA1 auf die Regulation der zellulären Cholesterin-Homöostase zu untersuchen. Dazu wurden Telomerase-immortalisierte Fibroblasten zweier Tangier-Patienten mit verschiedenen ABCA1-Mutationen und unterschiedlich ausgeprägter klinischer Manifestation der Arteriosklerose (TD1 und TD2) mit Fibroblasten eines gesunden Spenders verglichen. Der Cholesterin-Gehalt in TD-Fibroblasten im Vergleich zu Kontrollzellen war 1,4 bzw. 1,5-fach erhöht. Diese zelluläre Cholesterin-Akkumulation führte zur verminderten Expression der an der Cholesterin-Synthese und -Aufnahme beteiligten Gene, HMG-CoA-Reduktase und des LDL-Rezeptors. Daher war die endogene Cholesterin-Biosynthese im Vergleich zur Kontrolle um 27 % (TD1) bzw. 58 % (TD2) reduziert. Die Anreicherung von Cholesterin in den TD-Fibroblasten ging mit der verminderten Expression der Gene einher, die an der Regulation der Cholesterin-Homöostase bzw. dem Cholesterin-Export (ABCA1, ABCG1 und SREBP1c) beteiligt sind. Diese Störung war auch an einem entsprechend gegenläufigen Gehalt an Oxysterolen erkennbar (ein geringer Cholesterin-Export bewirkte einen höheren Oxysterol-Spiegel). Diese Untersuchungsergebnisse deckten jedoch gleichzeitig die Tatsache auf, dass keine strikte Korrelation zwischen einer verminderten Expression des defekten Cholesterin-Export-Gens (ABCA1) und der intrazellulären Cholesterin-Akkumulation existiert. Da die Expression von ABCA1, ABCG1 und SREBP1c laut den Ergebnissen anderer Arbeitsgruppen durch den Transkriptionsfaktor LXR reguliert wird, muß es –auf der Basis der hier vorgelegten Ergebnisse– LXR-unabhängige, aber wichtige Regulationsmechanismen des Cholesterin-Stoffwechsels geben. Die komplexe und unerwartete Regulation der LXR-Zielgene könnte erklären, warum Patient TD2 eine schwere Arteriosklerose aufweist, während Patient TD1 keinen klinischen Befund hat. Weitere Studien sind jedoch notwendig, um diese unbekannten und von Oxysterolen unabhängigen Regulationsmechanismen aufzuklären.
