This week, we were all over the place with a bunch of different designs and experiments. After last week's analysis of the TLV320DAC3100, we made some updates to the design and re-booked prototype PCBs. We also designed a triple-matrix bonnet: with our latest work on getting HUB75 RGB matrices working on the Raspberry Pi 5, we can now do matrix control on the latest Pi 5 chip. But we're limited by the RP1 chip, so to get big displays going, we'll need multiple strands—these don't use significantly more bandwidth because half of the pins are shared. Finally, we ended the week by getting another older prototype working: the SAM-M8Q is an entry-level all-in-one GPS from u-blox. It comes with both UART and I2C interfaces, plus a built-in antenna, so it's ready to go out of the box. The NMEA interface is trivial, but we also wanted to try out the UBX interface, and thankfully, Claude 3.7 was able to vibe-code it for us in a jiffy.

Heeeey, we're just having a super chill vibe here at the desk of Ladyada—writing a driver for the uBlox M8Q https://blog.adafruit.com/2024/09/10/a-mini-gps-from-ublox-with-i2c-and-uart/ which has both I2C and UART interfaces. As expected, it can do everyday NMEA output, but it can also do UBX, a "compressed" protocol for advanced data reads and writes over I2C/UART—or even SPI on some other chips. However, the UBX protocol is a hugely complex driver to implement, with dozens of commands and hundreds of flags. But why stress when you can viiiibe? We're using this beast of a spec as an excuse to try out the new Claude 3.7, which is doing great at chomping through the UBX documentation roughage and giving us some nice code on the other side. Within an hour, we're able to connect and switch to UBX mode by sending a well-formed message and receiving an ACK. What we like about coding with a good LLM is that it does the work we sometimes get lazy over, like handling various error conditions, timeouts, and verbose error messages. Senior executives and heads discussed how banks and financial institutions redefine business models to be more inclusive, innovative, and resilient to volatile market conditions for SMEs. They also highlighted the opportunities and challenges of utilizing a composable design to succeed in the market. Aswin phlaphongphanich of DeeMoney, Edu Trinidad of UBX, Frankie Wai of Temenos, Prashant Chauhan of Tata Capital, Sokha ROS of SBI LY HOUR Bank, and Srinivasan Shanmuganathan ( Shan ) of Standard Chartered shared their perspective on how can the composable design enable financial institutions to better address the needs of SMEs. Composable designs through interconnected components of microservices which can be assembled and reassembled quickly to go live fast are going to be the future. They also discussed how traditional banks have to evolve to compete with new market entries amid technological disruption and regulatory changes. To serve smaller-sized MSMEs a change of the game is needed in distribution and infrastructure. Wai discussed that new players keep raising the bar for customer experience. Whenever there is a new positive experience regardless of industry, customers expect the same standards. Sokha pointed out that most incumbents only look at the top SMEs which leaves MSMEs behind. Aswin revealed that the pandemic brought to the fore pain points on the payments side where MSMEs were unable to reach their banks for remitting fund transfers within and across borders, and were facing huge forex volatility impacting the cash flow and liquidity of MSMEs. Trinidad commented that embedded finance allows the ability to provide quick and specific solutions right to the customers. Shanmuganathan noted that infrastructure is more about accessibility not about technology enablement and how to gain access and speed to market that is critical. API connectivity and fintech collaboration are critical in building a self-service model for MSMEs to empower them to pick and choose what they need. He also highlighted that the biggest challenge with open banking is that there are many regulatory market restrictions, including how to utilize data that have not yet been addressed. To create service and product flexibility at the front end, Prashant shared that for agile capabilities one needs to build a layer on top of legacy systems, an open network of APIs for various functions. But to succeed in this, the most important part is to change the process, the policies and the digital mindset of the company accordingly. In the future, there is no brand loyalty only service loyalty and those FIs that serve the best and provide the best rates in the shortest time will win the race Key discussion points: How can the composable banking approach address the needs of SMEs and promote financial inclusion? How can traditional banks evolve to compete with challenger banks amid technological disruption and regulatory changes in digital payments and lending? How do organisations build and redefine business models to be more inclusive and resilient to volatile market conditions? How organisations are balancing corporate responsibility and profitability to achieve greater stakeholder satisfaction and engagement?

Homeotic (Hox) genes are transcription factors that dictate the development and compartmentalization (regionalization) of body parts in animals along the anterior-posterior (head to tail) axis. Using various insects and crustaceans, Dr. Nipam Patel studies how alterations in the expression of Hox genes could explain the evolution of specialized body parts in arthropods. Patel describes the spatially restricted patterns of Hox gene expression, explains the effects of Hox gene deletions, and how these phenotypes help us understand the manner in which Hox genes act to control the insect body plan. Nuclear positioning and functional regulation of endogenous genes and transgenes in the fruit fly Drosophila melanogaster and in mammalian cells

The goal of the work was to address the role of higher order nuclear architecture in the functional regulation of endogenous genes and transgenes in different species. In the first part of the work, 3D distance measurements were performed to analyze in WT flies and in different transgenic lines of Drosophila melanogaster (Cavalli and Paro, 1998; Zink and Paro, 1995) the nuclear localization of endogenous genes and transgenic constructs containing the Polycomb Response Element (PRE) Fab-7 relative to the nuclear periphery and heterochromatin. Transgenic constructs containing the Fab-7 element and three endogenous genes, Abd-B, sd, and Ubx, were first analyzed at their inactive state. The results showed that they were preferentially associated with the nuclear periphery and did not display specific associations with heterochromatin. The localization of the transgenic Fab-7 element was further analyzed at different states of activity. Activation of the transgenic Fab-7 element resulted in frequent (up to ~50%) association with the boundary of the heterochromatic domain. The percentages of such associations were tissue- and fl y line-dependent. Further investigations of the boundary of heterochromatin showed that this region has a complex organization, where euchromatic sites enriched in the active form of RNA pol II and trimH3K4, sites enriched in dimH3K4 and Pc-binding sites, as well as pericentromeric satellite DNA are exposed and juxtaposed towards each other. The concentration and specific architecture of such sites at the boundary of heterochromatin might help to maintain the equilibrium between activation and repression at this boundary. This specific environment might be favourable for maintaining PREs in the active state. I also investigated in three transgenic lines whether endogenous and transgenic copies of the Fab-7 element interact physically, using 3D distance measurements. In five tissues analyzed, no pairing between endogenous and transgenic copies of the Fab-7 element was observed. Also enhancement of Pc-mediated silencing did not induce pairing. Additionally, the general organization of Pc-binding sites was addressed in six larval tissues. We used different methods of microscopy and image analyses to count the numbers of Pc foci in nuclei from these tissues. Our data did not indicate clustering of Pc-binding sites and formation of so called „Pc bodies“. However, corresponding analyses are not without problems at the current stage of methodology and results must be interpreted carefully. Together with the results of previously published studies, which investigated pairing between PREs (Bantignies et al., 2003; Vazquez et al., 2006), my data demonstrated that such pairing is a highly tissue-specific phenomenon and is likely not involved in the regulation of PREs in various tissues. Activity-related positioning of transgenes was also addressed in transgenic porcine cell lines (Hofmann et al., 2003). Results of 2D erosion analyses showed that the LV-PGK transgene was associated with the nuclear periphery in its inactive state, while it occupied more interior positions in its active state. This corresponds to my results obtained with transgenic Drosophila lines. My data also suggested that the active LV-PGK construct might be associated with heterochromatin in one case. However, further experiments would be necessary to confirm such associations. The results obtained with Drosophila and porcine cells suggested conserved mechanisms for tethering inactive loci to the nuclear periphery. These were further addressed using the human CFTR locus as a model, which is closely associated with the nuclear periphery in its inactive state (Zink et al., 2004). The question was addressed whether Tpr, a protein associated with the nuclear basket, plays a role in the perinuclear localization of the inactive CFTR locus. CFTR showed a high degree of association with the nuclear periphery in control cells in accordance with previous data (Zink et al., 2004). After knock-down of Tpr via RNAi CFTR displayed a more interior positioning. This suggests that Tpr is involved in the organization of inactive gene loci at the nuclear periphery. Moreover, since Drosophila Tpr has a high level of homology to the mammalian Tpr (Zimowska et al., 1997) and as it has been shown that the yeast Tpr homologs Mlp1 and Mlp2 are involved in tethering of inactive loci to the nuclear periphery in yeast cells (Galy et al., 2000), it is possible that Tpr is a part of a conserved mechanism anchoring inactive loci to the nuclear periphery in eukaryotic cells.
