Abstract: Neurogenesis occurs in the adult brain in the hippocampal dentate gyrus, an area that contains neurons which are vulnerable to insults and injury, such as severe seizures. Previous studies showed that increasing adult neurogenesis reduced neuronal damage after these seizures. Because the damage typically is followed by chronic life-long seizures (epilepsy), we asked if increasing adult neurogenesis would prevent epilepsy. Adult neurogenesis was selectively increased by deleting the pro-apoptotic gene Bax from Nestin-expressing progenitors. Tamoxifen was administered at 6 weeks of age to conditionally delete Bax in Nestin-CreERT2Baxfl/fl mice. Six weeks after tamoxifen administration, severe seizures (status epilepticus; SE) were induced by injection of the convulsant pilocarpine. Mice with increased adult neurogenesis exhibited fewer chronic seizures. Postictal depression was reduced also. These results were primarily female mice, possibly because they were the more affected by Bax deletion than males, consistent with sex differences in Bax in development. The female mice with enhanced adult neurogenesis also showed less neuronal loss of hilar mossy cells and hilar somatostatin-expressing neurons than wild type females or males, which is notable because these two cell types are implicated in epileptogenesis. The results suggest that increasing adult neurogenesis in the normal adult brain can reduce experimental epilepsy, and the effect shows a striking sex difference. The results are surprising in light of past studies showing that suppressing adult-born neurons can also reduce chronic seizures. Copy rights belong to original authors. When Justin Botterill first arrived at the University of Saskatchewan, he took what he described as a 'shotgun approach' to choosing classes. Midway through his second year, his psychology professors introduced him to neuropsychology, and to psychiatric and neurological disorders. He was hooked. Botterill soon began working with rodent models, later focusing on the hippocampus and pathways involved in forming memories and spatial navigation. "The hippocampus is widely implicated in a variety of neurological as well as psychiatric disorders," said Botterill. "It's a really important and critical structure." After completing his PhD in cognition and neuroscience at the University of Saskatchewan, Botterill went on to work as a postdoctoral fellow at the Centre for Dementia Research at the Nathan Kline Institute for Psychiatric Research in New York, then at the University of Toronto Scarborough. In January, he was hired as an assistant professor at Anatomy, Physiology and Pharmacology at the University of Saskatchewan's College of Medicine. Today, he's setting up his laboratory and hiring researchers to help examine the dentate gyrus, its granule cells and the diverse roles mossy cells play across the hippocampus. "Several lines of research over the last five or ten years have really shown that [mossy cells] contribute to a lot of important functions. That's why they're very exciting to study," said Botterill. "It's a really interesting feedback loop." He said mossy cells are not a single homogenous group of neurons. "Although historically mossy cells have been treated as a single class of neurons in the hippocampus, several lines of research suggest they differ in their gene expression, morphology, physiological properties, and anatomy," he said. "No one's really investigated these potential subcategories of mossy cells at a deep level yet." In this episode, Botterill explains the role mossy cells in the dorsal hippocampus play in spatial and cognitive functions. He said those in the ventral hippocampus show strong links to emotions such as anxiety. He also is looking at the role these excitatory cells play in Temporal lobe epilepsy, a disease characterized by seizures, along with debilitating cognitive and behavioural effects. "One in three people with epilepsy don't respond to medication," said Botterill, who said he's hoping to focus on translational research in this area. Botterill hopes to better identify the cells, circuits and proteins inside the dentate gyrus that generate seizures. Alongside researchers including John Howland and Robert Laprairie, he's hoping to eventually test new therapeutics on these target areas, to see which ones best prevent or reduce seizure activity. "Can we develop treatments that would improve care for individuals, reduce the severity of their seizures, and allow them to lead healthier lives?"

[05:40] Nigel's First Proudest Moment Nigel's first proudest moment is winning the Wind Cup. It's the first trophy he won as an aspiring football player. Growing up, he wanted to be a professional football player. He was so proud when he was invited to the Saturday soccer at his primary school. Experiences like these shaped him as he grew to do bigger and better things. Now, he helps other people become entrepreneurs. [08:42] The Beginning of Entrepreneurs Circle The Entrepreneur's Circle was created in 2010 through an accident. After losing his business to the economic turndown in 2008, Nigel wanted to help other franchisees prepare for challenging times through recession training. His team mistakenly sent a video meant for the franchisees to the franchisees' customers. After seeing the learning gap and ‘appetite' of the email recipients, Nigel offered training for the customers of the franchisees. The Entrepreneurs Circle now has over 1,000 members from all over the world who are living more fulfilled, more comfortable, and happier lives. [15:14] Nigel's Second Proudest Moment All businesses need a team that works together towards the same goal. The biggest challenge is building and keeping the team. Nigel is grateful and proud that he could do this with his team at Entrepreneurs Circle. His team's dedication energises and inspires him. [19:31] Making the Most Out of Life Attack every day with gusto and find things that energise you. Life is too short to spend it doing things that do not make you feel better about yourself. Always be prepared so you can overcome adversity, including those that are not of your own doing. Nigel used to work for large corporations and abide by policies and decisions that did not align with him. Having his own business allowed him to ‘do things the right way.' Money will flow if your business is doing the right thing. [22:58] Nigel's Third Proudest Moment In 2015, Nigel's business lost £600,000 to a ‘systematic management of accounts and bookkeeping.' This forced him to take the Entrepreneurs Circle to a Creditors Voluntary Agreement (CVA). CVAs often end in the company closing and people losing money. However, with hard work, Nigel cleared the CVA a year early. Nigel considers the exit certificate as his single proudest moment as a business owner. He displays the exit certificate in his office as a reminder of his triumph. [31:42] Lessons from Major Adversities Major adversities will change you for the better and teach you the biggest lessons. Doing the right thing and using a values-driven approach to any adversity will create trust between you and your clients. The only way out is through. No matter how empathetic you are, you can never understand a situation as well as somebody who experienced it. [33:22] Business and Numbers You can never avoid numbers as a business owner. Any business owner can learn to drive their numbers with focus and guidance. [34:58] Nigel's Secret to Success The secret to success all comes down to how you use your time. People have built businesses bigger than themselves by spending time on the right things. Get away from the daily tasks and work on moving the business forward. Mistakes are normal. Just make sure to spend enough time on the things that will make a difference. When asked about what it is like to work with national teams, professional athletes and emergency room physicians, Cal will tell you that, “the stuff is the same, but the people are all different.” Adrienne will tell you one thing she has always admired about Cal is that he does what is right for the human being that sits in front of him. Cal is there for the people he works with, he speaks from the heart and he will tell you the truth, even when it may not be what you want to hear. Cal grew up on a farm with five brothers and sisters. He told us that the only way to survive in that setting was to have humility, a strong work ethic, and to care for the people around you. He mentioned that humility is not a lack of confidence, but a quiet version of it, and, that having gratitude for your life and those around you is key. His knack for caring for others transferred into all his work. When he spoke about his ability to have an impact on teams and groups he mentioned that ultimately what he did was make people feel needed and excited about what they could contribute to the process and then he helped them understand what they needed to do to get better. Of course, this is a very simple explanation of the work Cal has done and continues to do, but the message here is that caring for those around him is at the heart of his work. Upon completing his PhD, Cal sought out the five best sport psychologists in the world to learn the applied side of the job. He spent time with each of them and what he learned from that process helped him create his own style. Throughout his career he had the opportunity to work with several experts including Terry Orlick and Geoff Gowan. Now, in the sunset of his career, Cal reminds us that there are no ordinary moments. Every breath we get is a privilege, so enjoy the moment. He also suggests that recovery is the key to sustainable performance telling us that, "looking after yourself first may be the most unselfish thing you ever do." An inspiring leader, proud father, and grandfather, engaging storyteller, and undoubtedly a hero in our midst. We hope you enjoy our conversation with Cal Botterill.

The Sabres sent shockwaves through the hockey world as they fired Jason Botterill and named Kevyn Adams as his replacement. Much of Buffalo's scouting staff, player development and minor league coaching staffs were also released in the afternoon, effectively nuking the hockey department from the top down. Abstract: Glutamatergic dentate gyrus (DG) mossy cells (MCs) innervate the primary cell type, granule cells (GCs), and GABAergic neurons which inhibit GCs. Prior studies suggest that the net effect of MCs is mainly to inhibit GCs, leading one to question why direct excitation of GCs is often missed. We hypothesized that MCs do have excitatory effects, but each GC is only excited weakly, at least under most experimental conditions. To address this hypothesis, MC axons were stimulated optogenetically in slices. A brief optogenetic stimulus to MC axons in the inner molecular layer (IML) led to a short-latency field EPSP (fEPSP) in the IML, suggesting there was a direct excitatory effect on GCs. Population spikes were negligible however, consistent with weak excitation. FEPSPs reflected AMPA/NMDA receptor-mediated EPSPs in GCs. EPSPs reached threshold after GC depolarization or facilitating NMDA receptors. GABAA and GABAB receptor-mediated IPSPs often followed EPSPs. At the network level, an optogenetic stimulus led to a brief, small facilitation of the PP-evoked population spike followed by a longer, greater inhibition. These data are consistent with rapid and selective GC firing by MCs (MC [->] GC) and disynaptic inhibition (MC [->] GABAergic neuron [->] GC). Notably, optogenetic excitation was evoked for both dorsal and ventral MCs, ipsilateral and contralateral MC axons, and two Cre lines. Together the results suggest a way to reconcile past studies and provide new insight into the balance of excitation and inhibition of GCs by MCs. Copy rights belong to original authors. The trade deadline is next week and the Sabres are in a position where they almost have to make some waves in order to begin the process of finding upgrades to their forward group. We discuss the handful of UFAs the Sabres have to market along with floating a few potential targets Jason Botterill could chase in order to add to his roster. It's a big week for Botterill and the Sabres as the team floats on the periphery of the playoff race while struggling to gain a steady foothold in the Eastern Conference.
Much like the Sabres, the boys are struggling as they battle through some technical difficulties in this eppy. Morale is at an all time low as Ethan and Sam discuss how the Sabres dropped 3/4 games this past week against bottom-tier competition. The Sabres hot start has been fueled by a number of things, strong play from the blueline. Buffalo's group has been a strength and it stands to get stronger as Brandon Montour is nearing a return. Jason Botterill also confirmed that Zach Bogosian has started skating as well, that means the GM will have some tough decisions to make regarding his defense corps. We address what options Botterill has in thinning out a potentially overcrowded blueline before running through the highs and lows the Sabres have had in the third period this year.

Carolin is a 51-year-old ultra-runner and multi-day stage race specialist, however, this wasn’t always the case. When Caroline was 36 years old, she was a stay at home mum to three daughters. Wanting to feel better in herself, and wanting to get fitter and lose some weight, she started on a journey which would ultimately lead to her becoming an ultra-runner….accidentally!! She didn’t tell anyone what she was doing, and it was tough going at the start, she started with walking, then gentle jogging and gradually build it up over time to running further and further distance, until she was training for her first marathon! She persevered with it, even through injury and kept on challenging herself more and more. Since then Carolin has gone on to participate in some of the toughest multi-day ultras on the planet, from running the Ice Ultra in the Arctic Circle to the Jungle marathon in Peru.
