Abstract: Neutrophils are essential for host defense against Staphylococcus aureus (S. aureus). The neuro-repellent, SLIT2, potently inhibits neutrophil chemotaxis, and might therefore be expected to impair antibacterial responses. We report here that, unexpectedly, neutrophils exposed to the N-terminal SLIT2 (N-SLIT2) fragment kill extracellular S. aureus more efficiently. N-SLIT2 amplifies reactive oxygen species production in response to the bacteria by activating p38 mitogen-activated protein kinase that in turn phosphorylates NCF1, an essential subunit of the NADPH oxidase complex. N-SLIT2 also enhances exocytosis of secondary granules. In a murine model of S. aureus skin and soft tissue infection (SSTI), local SLIT2 levels fall initially but increase subsequently, peaking {approx} 3 days after infection. Of note, neutralization of endogenous SLIT2 worsens SSTI. Temporal fluctuations in tissue SLIT2 levels may promote neutrophil recruitment and retention at the infection site and hasten bacterial clearance by augmenting neutrophil oxidative burst and degranulation. Collectively, these actions of SLIT2 coordinate innate immune responses to limit susceptibility to S. aureus. Copy rights belong to original authors. LexaGene Holdings (CVE: LXG – OTCQB: LXXGF) CEO Dr Jack Regan joined Steve Darling from Proactive to share news the company has announced the release of a news test panel within the MiQLab testing unit. Regan telling Proactive, the next-generation test panel will screens for 10 pathogens and 30 gene variants. The new panel is expected to detect 96% of bacterial UTI's & 93% of bacterial SSTI's and pick up a much higher percentage of genes that confer resistance to some of the most commonly prescribed antibiotics.
Azithromycin is a macrolide antibiotic used to treat many bacterial infections. Some common uses are for Community Acquired Pneumonia (CAP), Group A Strep, Acute Exacerbations of COPD, STDs, among other bacterial infections. One of the most commonly known dosing regimens is 500 mg on day one, then 250 mg on days 2-5. When using the reconstituted suspension it must be refrigerated once reconstituted it must be discarded after 10 days. Azithromycin works through the inhibition of RNA-dependent protein synthesis by binding to the 50S ribosomal subunit. By blocking transpeptidation this inhibits the chain elongation step of protein synthesis. Azithromycin distributes well into most tissues and fluids with the exception of cerebrospinal fluid (CSF). Protein binding is concentration dependent varying from 7-51%. Azithromycin can be taken with or without food but food may decrease any GI distress. Ben Wolfe discusses how to prepare yourself to stand out in an audition. He talks about how to select the right material for musical theatre, how to make a good audition tape, and how to effectively present yourself in an audition. Ben is the Founder and Director at the Southeastern Summer Theatre Institute, a nationally ranked high school summer theatre intensive based in Hilton Head Island, SC. A native of Georgia, Ben spent his youth performing, and in college began to transition into directing, and soon after he settled into his natural habitat, producing. Although he was an entrepreneur from a young age, SSTI wasn't born to serve as a career, but rather as a one-time summer project. However, as one summer turned into two, then into ten, it became clear that SSTI was in fact a career. Working with a talented staff and student body drawn from all over the world, Ben and SSTI produced the first regional theatre for high school students. With new technical theatre programs and a professional division employing working professional actors throughout the year, SSTI continues to reach new heights. When not working around the clock May through August in South Carolina, Ben enjoys making his home in New York City, where he can keep a close watch on the industry he loves.
