Når vi taler om mental træning af atleter, handler det om at hjælpe topatleter med at holde fokus på opgaven, målet eller deres gameplan på et ekstremt højt plan. Men man behøver ikke at være topatlet for at kunne lære noget om ens hverdagsadfærd og hvordan man kan drage fordel af at kunne holde fokus på opgaven, selv på en almindelig grå mandag i november. At håndtere det mentale pres fra en hektisk hverdag eller presset under et langt løb, er en af de mest kritiske succesfaktorer, når vi taler om at stå i såvel forandringens behag som ubehag på vej til at opnå vores mål. Dette gælder både når vi skal forandre adfærd og gøre mere af noget for at nå vores mål, f.eks. at øge den distance vi vil løbe, eller gøre mindre af noget for at nå vores mål, f.eks. ikke at løbe 80 km hver dag, men kun løbe det hver anden dag.I dag skal vi altså tale om, hvordan man bruger mental træning til adfærdsforandringer, både når vi vil gøre noget mere eller mindre af noget. Spørgsmålet er: Hvordan får du noget til at ske, der ikke sker af sig selv, men som er afgørende for at du kan opnå dit mål? Til at hjælpe os med at udfolde dette emne og svare på dette spørgsmål, taler jeg i dag med søstrene Camilla og Johanne Prisak. Til daglig driver I firmaet Mental Atlet og har begge en kandidatgrad i Idræt og Sundhed med specialisering i sportspsykologi fra SDU. Og så er de begge ultraløbere.Lyt med og få:- Forståelse af adfærdsændringer: Få en klar forståelse af, hvordan mental træning kan hjælpe med at tackle adfærdsændringer, både når man ønsker at øge eller mindske en bestemt aktivitet eller vane- Anvendelighed i hverdagen: Opdag hvordan teknikkerne inden for mental træning ikke kun er forbeholdt eliteatleter, men kan anvendes af alle for at forbedre hverdagsadfærden og håndtere presset fra en travl hverdag.-3 R'er modellen: Lær om den effektive 3 R'er-model (Registrér, Release, Refokusér) og hvordan den kan hjælpe med at opretholde fokus på opgaven og målet, selv når distraktioner opstår.- Konkrete løsninger: Opnå konkrete løsninger til hver af de tre faser i 3 R'er-modellen gennem forskellige mentale træningsøvelser, herunder mindfulness, distancering og visuel koncentration.- Fremtidig praksis og refleksion med søstrene Prisaks få-det-til-at-ske-liste: Reflekter over den erhvervede viden og tænk over, hvordan man kan integrere disse teknikker i ens egen praksis for at opnå ønskede adfærdsændringer og mål.

Weber, from the University of Cincinnati Academic Health Center, describes a research perspective he authored that was published by Oncotarget in Volume 14, entitled, “Crossroads: the role of biomarkers in the management of lumps in the breast.” DOI - https://doi.org/10.18632/oncotarget.28402 Correspondence to - Georg F. Weber - georg.weber@uc.edu Abstract Premalignant lesions in the breast pose a difficult decision-making problem, whether to treat proactively and accept the side effects or to engage in watchful waiting and possibly encounter a later diagnosis of invasive cancer. A biomarker or set of biomarkers to inform on the individual progression risk would be beneficial to the patient and cost-effective for the healthcare system. The gene products of tumor progression may be expressed in early non-cancerous (“premalignant”) lesions, where they are associated with a high probability for full transformation in breast cancers. One such molecule is the OPN splice variant-c. OPN-c is also present in a fraction of the premalignant lesions, where it reflects an elevated risk for progression to cancer within 5 years, regardless of the lesion's subtype. This marker has the properties needed to facilitate decisions to treat at the premalignant stage. Rain chances overall for the day is 60 percent, but it appears from their analysis that rain possibilities will likely cease by 4 p.m.Crowding and capacity needs continue to stress the main University of Iowa Hospitals and Clinics, compelling administrators to expand the emergency department, reorganize existing services and — now — open a new “short-stay unit.”UIHC opened a new 30-room short-term unit Monday aimed at freeing up patient beds for sicker patients; streamlining the hospital experience for short-term patients requiring observation before heading home; and allowing providers to focus on either critical-care or short-term needs.Examples of UIHC patients who might require a short hospital stay are those needing further evaluation to determine whether they're stable enough to be released or should be admitted; patients recovering from surgery; or ER arrivals requiring observation, like those with chest pain, for example.Patients might find themselves in the short-stay unit after the operating room or a trip to the emergency room — which has become increasingly stretched in recent years.The family of a Muscatine man who went missing from Iowa City over the weekend hosted a prayer vigil Tuesday evening, and has been working with police on ongoing search efforts.Cristian Martinez, 20, had gone to Iowa City with some friends who last saw him in the alley behind Bardot Iowa, a bar at 347 S. Gilbert St., sometime before 1 a.m. Saturday. His phone had died earlier in the evening, according to the Iowa City Police Department.Martinez was reported missing Saturday after his family noticed he hadn't come home with his friends. Iowa City police investigators found security footage that showed Martinez at 12:56 a.m. in the 100 block of W. Harrison Street, walking west.Family, friends and other community members gathered Tuesday at the downtown Pedestrian Mall in Iowa City to hand out flyers and show support.The family has started a GoFundMe account to help with hiring a private investigator, and any other expenses as the search continues. As of Wednesday afternoon, the account had raised almost $10,000.Iowa City is taking the first step in exploring space and department needs of the City Hall building on Washington Street, which houses various city departments including police and fire.The city will be working with OPN Architects over the next six months on the master plan and feasibility study project, which the Iowa City Council approved as part of the consent agenda during Tuesday's meeting. Since it was on the consent agenda, there was no council discussion.OPN will review the current and projected space and needs of departments — including whether it would be possible to relocate the police department to a stand-alone building.

Myra, Dei & Mojo discuss the Weeknd's fifth album is both a sonic and thematic follow-up to After Hours. It also has him riding the cinematic and retro synth-pop sounds first explored in 2016's Star Boy to its zenith. This is immortalized by the A-list personnel behind the new project, from Swedish pop maestro Max Martin and EDM trio Swedish House Mafia to U.S. producer and film composer Daniel Lopatin, who goes by Oneohtrix Point Never, or OPN. You could argue that Dawn FM is merely a flowering of ideas that were there all along. House of Balloons creates chilly R&B out of samples of  Beach House, and Aaliyah and elsewhere seems to nod to the moody drums & bad vibes of Phil Collins classics and the bedroom antics of '90s R&B legends like Jodeci. Elsewhere, pop hitmaker Max Martin takes guitars and synths and transforms them into disco-inflected bangers such as the album's lone single, "Take my Breath." The rhythm guitars resemble a mix of '80s funk and Daft Punk on "Sacrifice," which was built atop a sample of Alicia Myers' 1981 track "I Want to Thank You." And on "Less Than Zero," a tender acoustic guitar opens a new-wave lane that we've never seen the Toronto singer explore before, a rare moment of sonic fun (it's also one of two tracks on this entire album that is in a primary key). There was even be a global early, live broadcast today on Amazon Music for those who are extra eager to share the listening experience with fans across the world. As we count down the hours until the official drop at midnight, I am extraordinarily thankful for The Weeknd's spontaneity, for I am sure we are about to experience music history in the making. Listen to The Weeknd on Apple music. Also, they discuss. Gunna has come a long way, solidifying himself as one of the most prominent hitmakers coming out of Atlanta. Though he didn't release much music last year, he appeared on several other compilations like Young Thug's Punk, Pop Smoke's Faith, Doja Cat's Planet Her, and many others. Now, it's time for young Wunna to shine. Additionally, he further explores the themes of people he can no longer trust and how that has changed him. In the chorus, he raps, "You know b*****s talk, but you were the boss, so we keep our business private." He backs up these lyrics by acknowledging how he has helped others with "I got some slimes out a beehive, I know they are dying 'bout it." Overall, this song is a coming-of-age track for Gunna and is a perfect opener for the final chapter of the Drip Season series. On the next track, "poochie gown," there is a lot of anticipation and beef surrounding this track. However, it seems that, while there may be a lot more behind the scenes of this track, the main point that comes to attention is his apparent diss at Freddie Gibbs—produced by Wheezy, this song bass-heavy track with Gunna delivering fast flows. Gunna drops the bar in the song, "Hang above the rim. I can't fuck with Freddie Gibbs." An opportunity came up to invest in this new emerging company that had some great IP. The company had found an investor that would help them with the production. The investor was going to contribute considerable funds too. Getting deeper into the business ideaJeffery went through this process of doing a deep dive and analysis around the business. He requested that he meet with the investor who would pick the ball up and put a lot of money in to make this company successful. Jeffery met the investor, and as he started to pick their brain and learn more about them, he asked them questions because he had doubts. However, he never admitted his misgivings; he just kept going forward. Ignoring hisJeffery’s gut still told him that something was not right. But because he had been working with the startup for such a significant amount of time, he trusted that they had picked the right investor. Jeffery did not want to slow down the progress, so he ignored his gut, did all of the analysis, came back, and signed off on the deal. The truth comes outJeffery signed everything off, and everything was good. About six months into the project, when the final handoff was supposed to happen, the unforeseen happened. Once everything was solidified and sorted out, the investor ended up having creditors coming after them, and they went bankrupt. Everything they had worked hard for went down the drain just because of one person. If only Jeffery had listened to his gut. Abstract: In the mouse, retinal output is computed by over 40 distinct types of retinal ganglion cells (RGCs) (Baden et al., 2016). Determining which of these many RGC types project to a retinorecipient region is a key step in elucidating the role that region plays in visually-mediated behaviors. Combiningretrograde viral tracing and single-cell electrophysiology, we identify the RGC types which project to the olivary pretectal nucleus (OPN), a major visual structure. We find that retinal input to the OPN consists of a variety of intrinsically-photosensitive and conventional RGC types, the latter a diverse set of mostly ON RGCs. Surprisingly, while the OPN is most associated with the pupillary light reflex (PLR) pathway, requiring information about absolute luminance, we show that the majority of the retinal input to the OPN is from single cell type which transmits information unrelated to luminance. This ON-transient RGC accounts for two-thirds of the input to the OPN, and responds to small objects across a wide range of speeds. Introduction: Alexander Caillet is renowned for his pioneering approach to team coaching and his state of mind research with organisational Leaders. He is an organisational psychologist, consultant, executive coach and team coach, working internationally. Alexander is CEO of Corentus, a coaching and consulting organisation dedicated to transforming teams. He is also faculty at Georgetown University, He is a frequent international speaker on subjects such as  change, Teams, Team Coaching and Leadership. Alexander has co-authored a mini book called Corentus Team Coaching, a fascinating read. Alexander is a dual citizen of France and the US and he lives in Boston with his wife and 3 beautiful daughters.   Podcast episode summary: In this episode Alexander Caillet communicates with enormous clarity & passion his approach to team coaching, the power of this methodology and the gift it can afford teams and organisations especially in answer to our VUCA world. Alexander describes the company's approach to team interventions, the models he deploys and the role of the team coach. He shares many vignettes to illuminate the power of team coaching and how by way of this work the teams work is sustainable.    Noteworthy points of discussion Alexander Caillet saw the power of Group Dynamics in his studies at Columbia University and he wanted to bring this power into his work with teams.  Team Coaching is the product of integrating a real time approach in group dynamics in a business or organisational setting with the team is doing its work so that they can adapt and get at improved performance  Teams and groups today are brought into the work using tools to support the building of team charters, defining a purpose, clarifying goals understanding roles etc. but they stumble on communication, trust and respect and although we have tools for these ideas they are seemingly insufficient when the team is back in the workplace.  The real translation of those tools around communication trust, psychological safety happens in the moment when the team has a breakdown back at work and they can see how to navigate these issues with real time coaching.  There is almost an implicit assumption at work on teams that “we don't do that kind of work it is too dangerous” but group dynamics in terms of norms/patterns/outcome are unavoidable and without it we lose opportunity to get a HPTs.  There is a role with teams for Team Facilitation, Team Building, Team Teaching and Consulting and there is also room for Team Coaching.  Corentus offers a customised approach that is a combination of all four modalities Team Coaching is an engagement over time sometime a year-that allows for team observation and analysis of its processes and patterns to further sustained difference  Corentus employs a definitive model to support teams engage with their work: The model is described as Sensing/Being/Making moves.  Being describes the way a Coach shows up-important to be fit for the work as this work is not for the faint of heart-essential to be grounded and to understand triggers – being calm and clear gets noticed and helps the team Sensing is about gathering data either quantitatively or qualitatively and then feeding it back to the team in a neutral manner . Corentus uses many tools but important to them is the idea of the relationship between norms/patterns and outcomes or the OPN model – all teams have patterns laid down by norms, assumptions and beliefs that generate definitive outcomes  Making Moves is about using the data in the various domains of decision making/accountability etc for difference  Having data and asking powerful questions in the moment when the team is doing its work can often be a real turning point for a team and the real value of Team Coaching  We walk into systems very quickly and often we become blind or go native to the system we are in. Team Coaching allows us to pause and notice and ask the team to do the same- to notice what is happening, what patterns are emerging and what needs to be changed if anything?  Important to this work is Leadership Coaching in addition to working with the Leader and team publicly, Corentus will work with the Leader and their impact offline over time.  We are living in a VUCA world  Volatility represents the relentless speed & pace of change, Uncertainty meaning things are harder to predict, Complexity making things difficult to analyse and Ambiguity making things harder to interpret.  VUCA drives a lot of overwhelm.  Corentus and Alexander sees a real need for teams to focus, for teams to invest time together, to come together to work things out.  Alexander sees the work of Team Coaches as  instrumental and of value in this VUCA world if we can transfer  the knowledge and skills we use with teams so that they can sense/be and make moves for themselves.  Alexander shared a powerful story of work with a with a well-known NGO and how his approach and models supported the Executive Team of this entity ultimately make decision four times faster than they ordinarily were able to do. This ability and skill learnt over time  was of enormous value when in reality the NGO was faced with a humanitarian crisis after the Tsunami in 2008.  Alexander is intent on building Corentus Globally , writing and speaking more, stepping out of delivery and having an impact in terms of increased global collaboration and collective intelligence . Ena od iniciativ za razvoj prostora je Automatik delovišče, skupina 20 akterjev z različnih področij, ki si prizadevajo za med seboj povezan razvoj mesta in ustanovitev neodvisnega urbanističnega sveta. Why is the transparency agenda critical to rebuilding citizens’ trust in our governance institutions? How have international movements been working to promote a new form of relationship between citizens and their governments? And how do Parliaments fit into this agenda? Citizen trust in democratic institutions, including parliaments, is critical to the functioning of democracy and effectiveness of governance. As the institution that represents citizens and translates their interests to legislation, parliaments are particularly affected by declining trust and blurred perceptions about their core functions. As the institution that is closest to citizens, it is therefore not surprising that parliaments are emerging at the center of international platforms and national efforts to retain and restore public confidence in democratic institutions by, for example, adopting measures to provide information on their day-to-day activities in more accessible and transparent ways. The Open Government Partnership (OGP) is an example of a multilateral initiative that aims to restore public confidence by securing concrete commitments from governments to promote transparency, fight corruption, empower citizens, and harness new technologies to strengthen democratic governance. The OGP currently has 75 participating countries including Canada, which have collectively made over 2,500 related commitments. The transparency agenda – and its pursuit through platforms like the OGP – are not exclusive to the executive branch of government. Parliaments in many countries – particularly those in the Americas – have been driving this agenda by working with citizens to develop their own internal action plans, and by functioning as models for transparency in their country. Increasing numbers of parliaments are also developing standalone chapters as part of their country’s OGP commitments. Through leadership roles in the OGP Legislative Openness Working Group and the ParlAmericas Open Parliament Network (OPN) Senator Hernán Larraín has been instrumental in positioning parliaments at the centre of this transparency agenda. In his address, he will consider progress achieved and upcoming opportunities, like the next Summit of the Americas about Democratic Governance against Corruption in April 2018 in Peru, to incorporate a parliamentary perspective and a focus on citizen trust in the shaping of the transparency agenda. This intervention was recorded during the ParlAmericas Canadian Section Annual Parliament Hill reception where Senator Larraín was invited to be the Guest Speaker to discuss how a number of parliaments in the Americas and the Caribbean are leading by example within the Open Government Partnership to be at the cutting edge of the transparency agenda.

During her tenure at Earthbound Farm, she worked with leading retailers throughout the US and Canada to develop and build organic programs and markets. Tonya was an instrumental leader in helping to build this California brand to its half billion-dollar sales level prior to her departure.Tonya brings an unabashedly positive outlook and can-do attitude to everything she does and understands the power of the judicious use of humor, even in business, which means that she’s in high demand. She has also consulted for leading US Venture Capital firms on investments in the organic space. Tonya, is the 2017 recipient of the Ag Against Hunger, Ag Women of the Year, the 2016 co-recipient, along with her husband, Rick, of the United Fresh, Lifetime Achievement Award, and the 2003 recipient of the Women in Produce achievement award. In 2010, she joined the Ag Business Department at Cal Poly, San Luis Obispo as an Adjunct Professor in Fresh Fruit and Vegetable marketing and has also served the university as a member of the President’s Council. Tonya also enjoys guest lecturing at Hartnell Junior College, Ag Business Department and most recently at CSUMB, School of Business. Tonya also serves on the CSUMB Foundation Board. Tonya holds a Bachelors of Arts degree in Communications from the University of California at Irvine. Osteopontin represents a multifunctional molecule playing a pivotal role in chronic inflammatory and autoimmune diseases. Its expression is increased in inflammatory bowel disease (IBD). The aim of our study was to analyze the association of osteopontin (OPN/SPP1) gene variants in a large cohort of IBD patients. Genomic DNA from 2819 Caucasian individuals (n = 841 patients with Crohn's disease (CD), n = 473 patients with ulcerative colitis (UC), and n = 1505 healthy unrelated controls) was analyzed for nine OPN SNPs (rs2728127, rs2853744, rs11730582, rs11739060, rs28357094, rs4754 = p.Asp80Asp, rs1126616 = p.Ala236Ala, rs1126772 and rs9138). Considering the important role of osteopontin in Th17-mediated diseases, we performed analysis for epistasis with IBD-associated IL23R variants and analyzed serum levels of the Th17 cytokine IL-22. For four OPN SNPs (rs4754, rs1126616, rs1126772 and rs9138), we observed significantly different distributions between male and female CD patients. rs4754 was protective in male CD patients (p = 0.0004, OR = 0.69). None of the other investigated OPN SNPs was associated with CD or UC susceptibility. However, several OPN haplotypes showed significant associations with CD susceptibility. The strongest association was found for a haplotype consisting of the 8 OPN SNPs rs2728127-rs2853744-rs11730582-rs11439060-rs28357094-rs112661-rs1126772-rs9138 (omnibus p-value = 2.07×10⁻⁸). Overall, the mean IL-22 secretion in the combined group of OPN minor allele carriers with CD was significantly lower than that of CD patients with OPN wildtype alleles (p = 3.66×10⁻⁵). There was evidence for weak epistasis between the OPN SNP rs28357094 with the IL23R SNP rs10489629 (p = 4.18×10⁻²) and between OPN SNP rs1126616 and IL23R SNP rs2201841 (p = 4.18×10⁻²) but none of these associations remained significant after Bonferroni correction. Our study identified OPN haplotypes as modifiers of CD susceptibility, while the combined effects of certain OPN variants may modulate IL-22 secretion.

Investigation of the effect of low oxygen tension on the osteogenic differentiation of human mesenchymal stem cells

Osteogenic differentiation of hMSC into osteoblasts is a prerequisite for subsequent bone formation. Numerous studies have explored osteogenic differentiation under standard tissue culture conditions, which usually employ 21% of oxygen. However, bone precursor cells such as hMSC reside in stem cell niches of low oxygen atmospheres. Furthermore, they are subjected to low oxygen concentrations when cultured on three dimensional scaffolds in vitro for bone tissue engineering purposes, and even more so after transplantation when vascularisation has yet to be established. Similarly, hMSC are exposed to low oxygen in the fracture microenvironment following bony injury. Recent studies revealed that hypoxic preconditioning improves cellular engraftment and survival in low oxygen atmospheres. In the present study we therefore investigated the osteogenic differentiation potential of hMSC under 2% O2 (hypoxia) in comparison to a standard tissue culture oxygen atmosphere of 21% (normoxia). The success of differentiation was validated through Alizarin red staining and RT-PCR analysis of osteoblast markers ALP and OPN. We assessed osteogenic differentiation of hMSC following hypoxic preconditioning to address whether this pretreatment is beneficial for subsequent differentiation under low oxygen tension. To validate our findings we carefully characterised the extent of hypoxia exerted on cells with respect to cell survival (WST assay) and proliferation (growth curve). Furthermore we also tried to elucidate the role of HIF-1 alpha with respect to osteogenic differentiation under hypoxia via silencing RNA and DFO, a pharmacological agent. Finally we tested whether an immortalized hMSC-line (SCP-1) would serve as a model system for hMSC. We found that hMSC proliferate better if cultured under 2% of oxygen. We confirmed that osteogenic differentiation of hMSC is indeed inhibited under hypoxia. We showed for the first time that hypoxic preconditioning of hMSC prior to osteogenic induction restores osteogenic differentiation of hMSC under hypoxia. HIF-1 alpha seemed not to play a significant role in osteogenic differentiation under hypoxia, as transiently knocking down of HIF-1 alpha in preconditioned samples did not show any differences in their osteogenic differentiation. Moreover stabilising Hif-1 alpha in hypoxic samples did not yield any osteogenic differentiation either substantiating the notion that HIF-1 alpha does not have a direct role in the osteogenic differentiation of hMSC under hypoxia. Together our data suggest that hypoxia favours stemness over differentiation by upregulating embryonic stem cell markers like OCT-4 and NANOG. Hypoxic preconditioning may help to restore the otherwise reduced osteogenic potential of hMSC, either within a hypoxic fracture environment or at the site of implantation of tissue engineered bone constructs. We therefore believe that hypoxic preconditioning is a helpful tool for successful regenerative cell-based therapies in bone tissue engineering. SCP-1 cells might be used as a model system for hMSC as they are easy to handle, can be cultured to a desired cell number within a very short period of time, are relatively inexpensive and above all do not go into senescence as seen with hMSC after approximately 20 passages. Apart from their distinct advantages SCP-1 cells still maintain the specific CD markers characteristic for hMSC and are able to differentiate into adipogenic, osteogenic and chondrogenic lineages. However for in vivo experiments in animals a constant monitoring of neoplastic transformation is mandatory

Osteopontin and `Melanoma Inhibitory Activity': Comparison of Two Serological Tumor Markers in Metastatic Uveal Melanoma Patients

Background: Evaluation of the protein osteopontin (OPN) as a potential new marker in comparison to melanoma inhibitory activity (MIA) for screening and detection of metastatic uveal melanoma. Methods: Plasma levels of 32 patients with uveal melanoma were analyzed for OPN and MIA by enzyme-linked immunosorbent assay (ELISA). Fourteen of these patients had clinically detectable liver metastases. Results: Median plasma concentration of OPN in patients with metastatic disease was 152.01 ng/ml compared to 47.39 ng/ml in patients without clinically detectable metastases (p < 0.001). The difference between the median MIA plasma levels in patients with (13.11 ng/ml) and patients without (5.64 ng/ml) metastatic disease was also statistically significant (p < 0.001). No correlation could be found between MIA or OPN levels and tumor height in patients without clinically detectable metastases. Conclusion: The proteins MIA and OPN seem to be promising tumor markers for the metastasis screening in patients with uveal melanoma. Copyright (C) 2009 S. Karger AG, Basel
