Full Moon Monday!!! Yowza!!!!!!I’m so glad that when I decided to put this here podcast on the internets that I decided early on that Monday was the day to do it. I don’t know about you, but Mondays are generally tough days for me, so I’m really glad that no matter what, I will always have this tiny piece of joy and excitement no matter what happens, because it’s podcast day. And today is extra, extra rad because I got to sit with someone who I’ve admired from afar for quite some time and then the podcast was the vehicle for me to swallow my nerves, reach out, and be blown away by the conversation I got to have.Our guest today, the AMAZING Shari Hampton, is a podcaster, on and off again blogger and, founder of Served Up Sober. Shari battled alcohol and drug addiction for over 35 years until one day five years ago, she landed in the arms of a retired psychiatrist and anthropologist turned holistic wellness enthusiast. She worked with this husband and wife team who merged their perspectives into a regime that combined various holistic modalities and focused on introspective examination and healing. It was through that experience she was able to build a life of recovery and gained awareness around how recovery exists at the crossroads of traditional methods and holistic ones. Shari soon felt a deep calling to create her own program that blended the two together.She founded Served Up Sober because there is a stigma attached to healing that needs to be broken. Shenoa and Erin have a very powerful and insightful conversation regarding their expectations of one another and the business in general.
In this episode Manan Sharma from USDA ARS fills in for Don who is away in Brazil. Manan grew up in Alabama and studied Microbiology as an undergraduate at the University of Florida. He received his M.S. and Ph.D. in Food Science from the University of Georgia where he focused on food safety microbiology. Manan and Ben talked about formative experiences in their careers including Manan’s high school internship at Research Genetics where he had great mentors who helped him appreciate molecular and microbiology. Research Genetics was later bought out by Invitrogen). Manan gave a shout out to his great mentors Bob Zahorchak, his first laboratory mentor, and Jim Hudson the president and CEO of Research Genetics. Manan also talked about his advisor at the University of Georgia, the great Dr. Larry Beuchat who encouraged him to take chances with his ideas and explore. Manan talked about his first project at USDA ARS: blowing up meat using hydrodynamic pressure processing (HDP) - different from hydrostatic pressure processing. HDP, where an explosion in a vessel of water is used to create a highly energetic wave is featured in an episode of MythBusters. Manan had to obtain a license from the ATF to use the explosives needed for this technology. Manan reported that while HDP was effective at blowing the roof off of his test facility it wasn't great at inactivating of food pathogens. Manan and Ben talked about food safety for leafy greens, particularly on the persistence of pathogens in manure and biological amendments. The guys talked about two a 2006 E. coli O157:H7/leafy green outbreaks, one connected to spinach and another linked to shredded lettuce leading to a research focus and the creation of the Center for Produce Safety, the Leafy Green Marketing Agreement. The conversation went to further impacts of big events including a 2012 Salmonella Outbreak linked to peanut butter manufactured by Sunland Farms in New Mexico which continues to impact growers. Manan talked about a paper, Bacterial Occurrence on Kitchen Hand Towels and extra-intestinal pathogenic E.coli (ExPEC). Extraintestinal pathogene Escherichia coli (ExPEC) sind die häufigsten Erreger von Harnwegsinfektionen beim Menschen. Darüber hinaus können sie eine Vielzahl weiterer Erkrankungen wie Pneumonie, Wundinfektion, Neugeborenenmeningitis oder Sepsis hervorrufen. Hierzu verfügen ExPEC über ein breites Repertoire von Pathogenitätsfaktoren wie Adhäsine, Toxine und Eisenaufnahmesysteme. Wesentlicher Bestandteil von Eisenaufnahmesystemen sind Rezeptoren in der bakteriellen Außenmembran, wie z. B. die Siderophorrezeptoren FyuA und IroN sowie der Häminrezeptor ChuA. Ihre Assoziation mit der Virulenz von ExPEC und die Lokalisation in der Außenmembran bzw. an der bakteriellen Oberfläche lassen Eisenaufnahmerezeptoren als geeignete Zielstrukturen eines Impfstoffes gegen ExPEC erscheinen. Die vorliegende Arbeit erbrachte grundlegende Erkenntnisse hinsichtlich der Eignung von Eisenaufnahmerezeptoren als potentielle Zielstrukturen, indem Seren von Patienten mit einer E. coli-Infektion und Seren aus dem Mausinfektionsmodell im Immunoblot auf Antikörper gegen FyuA, ChuA und IroN untersucht wurden. Hierzu wurde eine Sammlung von E. coli-Patientenisolaten und der entsprechenden Patientenseren angelegt. Die Isolate wurden mittels PCR hinsichtlich mehrerer ExPEC-assoziierter Gene, u. a. fyuA, chuA, iroN und usp charakterisiert. Im Immunoblot wurden dann die Seren auf Antikörper gegen die rekombinanten Proteine FyuA, IroN, ChuA und Usp getestet. Im Unterschied zu FyuA, ChuA und IroN handelt es sich bei dem zusätzlich untersuchten Usp um ein ExPEC-Protein mit bisher weitgehend unklarer Funktion. Die unterschiedliche Prävalenz von ExPEC-assoziierten Faktoren unter den Patientenisolaten konnte bestätigt werden. Es gelang erstmals, bei Patienten spezifische Antikörper gegen Eisenaufnahmerezeptoren von ExPEC nachzuweisen. Für die hierzu durchgeführten Immunoblot-Untersuchungen konnte in dieser Arbeit erstmals der Häminrezeptor ChuA rekombinant exprimiert und aufgereinigt werden. Die humorale Immunantwort gegen die einzelnen Eisenaufnahmerezeptoren wies ein relativ heterogenes Bild auf. Während bei den Patienten einerseits Antikörper unter der Infektion gebildet wurden (ChuA), waren sie andererseits bei IroN offenbar schon vorher vorhanden oder konnten, wie für FyuA gezeigt, nicht nachgewiesen werden. Auch im Mausmodell der ExPEC-Peritonitis fanden sich nur Antikörper gegen IroN, die in einem entsprechenden Tiermodell der ExPEC Harnwegsinfektion nicht darstellbar waren. Aufgrund der Ergebnisse dieser Arbeit ist es denkbar, dass die Bildung von Antikörpern gegen ExPEC-Außenmembranproteine zum einen von der Intensität und der Dauer der Infektion und zum anderen von Unterschieden in der Expression und in der Immunogenität der einzelnen Eisenaufnahmerezeptor-Proteine abhängig ist. Es ist jedoch nicht völlig auszuschließen, dass auch methodische Gründe(z. B. bei der Durchführung des Immunoblots) die Antikörpernachweise negativ beeinflusst haben. Die Ergebnisse dieser Arbeit konnten die Bedeutung von Eisenaufnahmerezeptoren als potentielle Zielstrukturen eines Impfstoffes bestätigen. Insbesondere die Rolle von IroN wurde durch die Beobachtungen unterstrichen. Die unterschiedliche Prävalenz der einzelnen Eisenaufnahmesysteme unter ExPEC und die Unterschiede der humoralen Immunantwort deuten jedoch darauf hin, dass es nötig sein wird, einen polyvalenten Kombinationsimpfstoff herzustellen, der gleichzeitig gegen mehrere Eisenaufnahmerezeptoren sowie weitere Zielstrukturen wie Adhäsine und Toxine Antikörperantworten erzeugt
