Abstract: While past studies have suggested that plasticity exists between dermal fibroblasts and adipocytes, it remains unknown whether fat actively contributes to fibrosis in scarring. We show that adipocytes convert to scar-forming fibroblasts in response to Piezo-mediated mechanosensing to drive wound fibrosis. We establish that mechanics alone are sufficient to drive adipocyte-to-fibroblast conversion. By leveraging clonal-lineage-tracing in combination with scRNA-seq, Visium, and CODEX, we define a mechanically naive fibroblast-subpopulation that represents a transcriptionally intermediate state between adipocytes and scar-fibroblasts. Finally, we show that Piezo1 or Piezo2-inhibition yields regenerative healing by preventing adipocytes activation to fibroblasts, in both mouse-wounds and a novel human-xenograft-wound model. Importantly, Piezo1-inhibition induced wound regeneration even in pre-existing established scars, a finding that suggests a role for adipocyte-to-fibroblast transition in wound remodeling, the least-understood phase of wound healing. Adipocyte-to-fibroblast transition may thus represent a therapeutic target for minimizing fibrosis via Piezo-inhibition in organs where fat contributes to fibrosis. Jenn's parents separated when she was young and it took a toll on her well being and learning to grow up fast was a must.  Jenn had come to accept that her life would consist of being in and out of rehab/ hospitals due to her drinking and meth use.  Jenn thought for sure her daughter would provide the strength and insight for her to get and stay sober but it didn't. After 11 rehabs Jenn still struggled to stay sober and it wasn't until she surrendered and became willing to work a 12 step program that her life changed. Jenn is a strong example of sobriety and is a real light in this world. Jenn has been sober for 4 years and her life has completely changed. This is Jenn's story on the sober motivation podcast. "Beyond Your News Feed: Understanding Contemporary Politics" is a podcast by the Department of Political Science. This episode of Beyond Your News Feed explores the fate of local news. Dr. Andrea McDonnell and Dr. Matt Guardino, both associate professors of political science, join host Dr. William Hudson, professor of political science, for a conversation about the dire straits of local newspapers and the impact on American democracy. Christian is a very talented, charming young man. He's a musician and his instrument of choice is his voice In 2017 he had a good run on U.S TV's family entertainment juggernaut America's Got Talent. But as you know, here at Studio 1, we're not interested in such shallow fripperies. People involved in or watching these so-called reality television shows often talk of the participant's “journey” - which generally means didthey got to Round 3 or the quarter finals and did they shout at anyone or cry like a baby on the way But in the case of Beth and Christian, a much more interesting and significant journey actually began twenty one years ago when Christian was born with a severe visual impairment. As Beth says in the course of the interview you're about to hear… “I started out holding this infant and not knowing if he would ever see my face or what life would look like for him… I never knew anybody else who was legally blind” Blindness and low vision are often seen very much as “other” something that happens to someone else. One of the things we try to do here at Studio 1 is to get across the message that the people this stuff happens to are just ordinary people like you with feelings, hopes, dreams and busy busy lives.
