In this week's episode we'll learn about Azacitidine in VEXAS syndrome. Treatment can provide responses in patients with this complex autoinflammatory disorder. But relapse rates were high, so long-term therapy may be required to maintain disease control. After that: A step forward in precision blood matching. High-throughput array genotyping enables extended matching to reduce antibody formation. The results show the potential for reducing harm in regularly transfused patients. Finally, identifying a new vulnerability in TP53-mutated AML. Loss of the tumor suppressor BAP1 defines a unique subtype of TP53-mutated de novo AML. BAP1 loss also confers sensitivity to BCL-xL inhibitors in vivo, opening a new therapeutic avenue.Featured ArticlesEfficacy and safety of azacitidine for VEXAS syndrome: a large-scale retrospective study from FRENVEXArray genotyping of transfusion-relevant blood cell antigens in 6946 ancestrally diverse study participantsLoss of BAP1 defines a unique subtype of TP53-mutated de novo AML and confers sensitivity to BCL-xL inhibitors

Gavin Newsom; and the rear-wheel drive trim of the Tesla Cybertruck lasted about five months before it was unceremoniously removed from online sales. How is it that during the era of Trump and Con Inc. Twitter, we seem to embrace so many good ideas, and Trump even embarks on implementing some of them, yet over time we continue to slide backward? It's because there is no follow-through and the Left evolves to bigger playing fields. I explain the "small step forward, giant leap backward" dynamic in terms of immigration, crime, abortion, culture, techno-fascism from COVID, and digital currency. Also, what is the endgame of the AI data-center agenda? There is some good news, as local residents in some counties are successfully fighting back. Melanoma is the fifth most common cancer in the UK, accounting for around 4% of all new cancer cases. While immunotherapy is a standard treatment for advanced melanoma patients, only about half of patients respond well, leaving the rest at risk of disease progression and metastases. In a new pharmaphorum podcast, web editor Nicole Raleigh speaks with Phillip L'Huillier, CEO of Scancell, about how the company develops novel immunotherapy products for the treatment of multiple cancers, as well as its recently announced partnership with the NHS Cancer Vaccine Launch Pad, and latest data release from the Phase 2 clinical SCOPE study in melanoma. L'Huillier discusses their melanoma-specific vaccine, and more broadly this pivotal time in immuno-oncology and possibilities on the near horizon.
