Abstract: BackgroundAlzheimers disease (AD) is characterized by progressive neurodegeneration, but the specific events that cause cell death remain poorly understood. Death Induced by Survival gene Elimination (DISE) is a recently discovered powerful cell death mechanism mediated by short (s) RNAs including micro (mi) RNAs acting through the RNA induced silencing complex (RISC). G-rich 6mer seed sequences in the sRNAs (position 2-7) target hundreds of C-rich seed matches in genes essential for cell survival resulting in the simultaneous activation of multiple cell death pathways. The RISC of most cells is occupied by miRNAs with nontoxic 6mer seeds, which may protect them from DISE by blocking loading of toxic sRNAs. However, during aging when miRNA expression decreases, toxic sRNAs may enter the RISC more readily leaving cells primed for DISE. Whether DISE contributes to neuronal loss in a neurodegenerative disease such as AD has not been evaluated. MethodsUsing Ago precipitation and RNAseq (Ago-RP-Seq) combined with SPOROS, a recently developed bioinformatics pipeline to analyze small RNAseq data with respect to 6mer seed toxicity, we analyzed RISC bound sRNAs (R-sRNAs) in in vitro models and in the brains of multiple in vivo AD mouse models, aged mice, and AD patients. ResultsWe find that in in vitro cell line studies, in mouse models that show neurodegeneration, and in the aging brain R-sRNAs shift to more toxic seeds. In contrast, in cells that survived in post-mortem brains of AD patients and the brains of "SuperAgers", individuals over age 80 who have superior memory performance, R-sRNAs shift to more nontoxic seeds, supporting a protective function of miRNAs. ConclusionOur data provide first evidence of a contribution of DISE to the neurotoxicity seen in AD suggesting that increasing the levels of protective miRNAs in the brain or blocking the activity of toxic R-sRNAs could lead to a novel way of treating the disease. My name is Henry Sink, CRNA, a pseudonym. HENRY stands for “high earner not rich yet.” SINK stands for “single income no kids.” CRNA, as we all know, stands for “Certified Registered Nurse Anesthetist.” I graduated from my CRNA program in May 2020 and started working in August 2020 after passing the NBCRNA in June 2020. I earned my MSN.  I realized from an early point in my nursing career that though I was making good money, whether at the bedside or as a travel RN, that it was not about how much I was making but instead how much I was saving and investing for my future.  As a full time CRNA, with over 6 years as a bedside RN and over a year and a half as a CRNA, I have taken what I have learned and am passing it on to the next generation of nursing students, RNs, and APRNs. This is in an attempt to educate these individuals about the link between their income and financial independence. I created my platform, all on Instagram, during the summer of 2020. I had just finished my board exam, the NBCRNA, and realized I finally had time back on my hands. I knew that I wanted to talk about financial issues in the nursing community but wanted to specifically speak to SRNAs and CRNAs about their current and future financial positions.  In the future, I want to work to create financial literacy and financial independence resources for individuals at all levels of their nursing career. Whether it is a pre-nursing student who is nervous about taking out student loans to pay for nursing school, or a newly graduated CRNA who wants to know how to navigate their workplace retirement accounts, investing in different asset classes, or paying down their loans from their graduate program. Support the show

Well, this episode is for you! Morgan Fitterer and Cynthia Shen are two SRNA moms spilling all of the secrets and discussing common struggles SRNAs face while in school. They sit down and discuss mom guilt, finding balance, the motivation to pursue this profession, handling tough rotations, and more!  Both Cynthia and Morgan became pregnant before being accepted to CRNA school. Despite their fears, this didn’t hold them back from pursuing their dream. The ladies contribute a lot of their success to the support of their husbands and family during this time. They are both set to graduate this spring! The ladies discuss having mom guilt. How leaving their child at daycare to study was oftentimes a struggle for them. Whether it was focusing on relationships, money, parenting, school work, or time for themselves, focusing on everything causes nothing to be done well.  19:40 How did you find balance?  Morgan knows this time in her life is temporary and breaks this large goal up week by week. She spent time figuring out how to achieve these large goals. One of the biggest things was working on time management and overcoming mom guilt.  Cynthia had a wake-up call at the beginning of the program. She started by modifying her study habits. She recognized she was an auditory learner, and she made sure she used her time wisely. She was always looking for smaller chunks of time to maximize her study time. Having clear communication with her spouse was another major factor in her success. They spent 30 minutes together on Sundays planning the week out.  27:40 What made you interested in CRNA school? Morgan found that she was most happy when she was being challenged, and CRNA school did this for her. One step pushed her to master the next, and having a mentor that encouraged her helped Morgan feel motivated. She had a fire inside her that pushed her to learn and grow. She was drawn to the autonomy that being a CRNA provided.  Cynthia's interest was originally in neuroscience. She was searching for something that overlapped her interest in neuroscience and nursing. Her desire to become a CRNA was solidified when she shadowed a CRNA and got to see her mentor put a “patient’s brain to sleep.” She was sold and has loved how her two interests have messed together to fuel a lifelong passion.  32:00 How did you handle your pediatric rotation as a mom? Cynthia says that her peds rotation was a struggle for her as a mom. She remembers crying on her way home from these cases as she thought about the mom in the hospital with her ill child as she gets to go home and be with hers. She became so appreciative of her healthy child. She salutes those moms who struggle with their sick kids. Morgan put things into a different perspective after having her own child. She never wanted to make a promise to a parent that she couldn’t keep. She has been vocal with her preceptors about what she is and is not comfortable with.  39:00 What advice would you give to the parent or soon-to-be parent thinking about going to CRNA school? Morgan states that “it's not about how smart you are, it's about how much dedication you put in. It takes time, dedication, and grace.” She encourages those interested in CRNA school to take one thing at a time, and anyone who has the motivation to continue can do this.  Cynthia feels that timing is never quite right. She never felt 100% ready to go. She encourages those on the fence to just do it. What is the worst that could happen? There is no downside to trying. Cynthia Shen: Hello! R., Vanderpool, C. K., Fei, J. Abstract: Small RNAs (sRNAs) are important regulators of gene expression in bacteria, particularly during stress responses. Many genetically and biochemically well characterized sRNAs regulate gene expression post-transcriptionally, by affecting translation and degradation of the target mRNA after they bind to their targets through base pairing. However, how regulation at each of these levels quantitatively contributes to the overall efficacy of sRNA-mediated regulation is not well understood. Here we present a general approach combining imaging and mathematical modeling to determine kinetic parameters at different levels of sRNA-mediated gene regulation. Unexpectedly, our data reveal that certain previously characterized sRNAs are able to regulate some targets co-transcriptionally, rather strictly post-transcriptionally, and suggest that sRNA-mediated regulation can occur early in the mRNA's lifetime, perhaps as soon as the sRNA binding site is transcribed. In addition, our data suggest several important kinetic steps that may determine the efficiency and differential regulation of multiple mRNA targets by an sRNA. Particularly, binding of sRNA to the target mRNA is likely the rate-limiting step and may dictate the regulation hierarchy observed within an sRNA regulon. Copy rights belong to original authors. Abstract: MicroRNAs (miRNAs) are small non-coding RNAs that are key players in the regulation of gene expression. In the last decade, with the increasing accessibility of high-throughput sequencing technologies, different methods have been developed to identify miRNAs, most of which rely on pre-existing reference genomes. However, when a reference genome is absent or is not of high quality, such identification becomes more difficult. In this context, we developed BrumiR, an algorithm that is able to discover miRNAs directly and exclusively from sRNA-seq data. We benchmarked BrumiR with datasets encompassing animal and plant species using real and simulated sRNA-seq experiments. The results demonstrate that BrumiR reaches the highest recall for miRNA discovery, while at the same time being much faster and more efficient than the state-of-the-art tools evaluated. The latter allows BrumiR to analyze a large number of sRNA-seq experiments, from plants or animals species. Moreover, BrumiR detects additional information regarding other expressed sequences (sRNAs, isomiRs, etc.), thus maximizing the biological insight gained from sRNA-seq experiments. Finally, when a reference genome is available, BrumiR provides a new mapping tool (BrumiR2ref) that performs an a posteriori exhaustive search to identify the precursor sequences.
