Koiran ruokaa pitäisi miettiä kokonaisuutena. Piatsi kun se on pakko pilkkoam osasiin. Oikeammin kyse on silloin valikoinnista niistä syistä ja seurauksista miksi jotain ruokaa annetaan koiralle. Silloin ruoka lakkaa olemasta ruokaa koiralle ja käyttötarkoitus onkin olla ravintolisä. Kaksi yleistä esimerkkiä ovat voi ja siirappi, joita kumpaakin kannattaa harkita koiran ongelmissa. Voi: näppärä rasvalisä koiralle Voi on katoavaa kansanperinnettä suomalaisessa keittiössä. Ketoilun ja muiden VHH-ruokavalioiden muodikkuuden myötä voi on hieman palautunut muuhunkin ruokakäyttöön kuin perinteisen leivonnan rasvalisänä, mutta silti kovien rasvojen käyttö vähenee kokonaisosuudesta. Koirille voita ei kannata käyttää perusruokinnassa, koska rasva olisi syytä saada perusruuasta. Mutta voi saattaa olla käyttökelpoinen energialisä, varsinkin suolisto-ongelmaisilla, närästäville tai muutoin rasvarajoitteisilla koirilla. Voi on syystä tai toisesta koirilla kohtuullisen hyvien siedetty. Kukaan ei oikeastaan tiedä miksi, mutta ylipäätään kovat rasvat aiheuttavat vähemmän ongelmia kuin öljyt. Yksi syy siihen saattaa olla niinkin yksinkertainen asia kuin rakenne. Se sulaa ja käyttäytyy kuin muukin ruoka, vaikka taatusti sulaa nopeammin, mutta silti olomuodon muutos juoksevaksi kestää aikansa. Silloin voi (ja muut kovat rasvat) ehtivät eräällä tavalla sekoittua ruokamassaan. Öljyt sen sijaan pysyvät erottuneina pidempään ja juoksevina myös nousevat helpommin eräänlaisena ohivirtauksena vatsalaukun supistuessa. Kun öljy, vatsahappojen takia matalamman pH:n saaneena, pusertuu koiran normaalistikin ihmistä väljemmän ylemmän vatsaportin läpi nieluun, niin se tuntuu närästyksenä. Suurin syy voin sopivuuteen rasvaongelmaisille koirille lienee kuitenkin yksinkertaisempi. Se on maitorasvaa ja ylipäätään koostumukseltaan erilaista kuin lihan tai kasvien rasvat. Lisäksi se mielletään puhtaaksi rasvaksi, mutta ei se ole. Voissa on noin 20 prosenttia vettä joukossa. Maidossa rasva on miselleinä, eräänlaisisan rasvapalloina. Jokaista palloa ympäröi fosfolipideistä ja proteiineista muodostuva kalvo. Kun maito aletaan hapattamaan ja sen jälkeen kirnuamaan, niin nuo kalvot rikotaan. Silloin rasvapallukat alkavat yhdistymään ja aletaan saamaan voita. Nestettä, vettä ja heraa, kaadetaan pois, kirnuamista jatketaan ja loppuvaiheessa voimassaa jopa hieman huudellaan vedellä vaivaamisen yhteydessä, että saadaan viimeisetkin maidon, kerman tai piimän jäämät pois. Voissa rasva on kolmessa eri olomuodossa: vapaana maitorasvana rasvakiteinä alkuperäisinä rasvamiselleinä Tuon kolmikon keskinäiset suhteet vaikuttavat voin rakenteeseen, kuten mitä enemmän rasvaa on kiteinä, niin sitä kovempaa se on. Entisaikoina itsekirnutun voin koostumus riippui siitä miten kukakin maitoa oli käsitellyt, ja kuinka paljon voihin jätettiin vettä. Teollinen voin valmistus tuottaa melkoisen tasalaatuista jälkeä, ja jos eroja on, niin se löytyy enemmältkin meijereiden väliltä, ei meijerien omien erien väliltä. Tuo kaikki yhdessä - rasvan rakenne, rasvahappokoostumus sekä vesimäärä - selittävät hyvin pitkälle miksi voi on usein vatsaystävällisempää kuin läski tai öljy. Voi eroaa rasvakoostumukseltaan jonkun verrankin niin öljyistä kuin lihankin rasvoista. Jotta asia olisi hieman hankalampaa, niin rasvakoostumus eroaa myös naudoittain ja varsinkin millaista rehua ne ovat saaneet. Mutta koska nautojen pitäminen ja ruokkiminen on hyvin samanlaista kaikilla navetoilla, niin ravinnolla ei ole suurtakaan käytännön merkitystä voin rasvakoostumukseen. Paitsi jos joku meijeri päättää haluta tuottaa hieman toisenlaista maitoa rasvan suhteen. Siirappi: nopeaa energiaa koiralle Siirappi on nopeita hiilihydraatteja. Se on niiden voima koiran ruokinnassa, sekä suurin syy omistajille olla käyttämättä sokereita. Ihmisten ylipainon ja ahneuden takia hiilihydraatit on demonisoitu ja niiden potentiaali jätetään hyödyntämättä silloin kun edut koiralle olisivat selvät. Siirappi säilyy, on kohtuullisen edullista ja helposti annosteltavaa. Sen koostumuksesta iso osa on glukoosia, joka imeytyy suoraan. Loput ovat muuten vain yksinkertaisia ja helposti hyödynnettäviä sokereita. Siirapilla haetaan niitä kahta ainoaa asiaa, jonka takia hiilihydraatteja ylipäätään syödään: energiaa ja insuliinin nostamista. Toki kumpikin onnistuu ilmankin sokereita, mutta nopeus ei ole sama. Lisäksi yksinkertaisilta sokereilta puuttuvat usein ne haitat, jonka takia koira ei kykene syömään riittävästi rasvaa - närästys ja suolisto-oireilu. Koiran ruokaa katsotaan aina laajemmin+ Voi ja siirappi ovat hyelppoja esimerkkjä siitä miten ruokaa on mietittävä laajemmin kuin uskomusten tai jonkun yhden nimetyn ominaisuuden kautta. Toki se vaatii erilaisten asioiden syvempää osaamista ja perusteita parempaa osaamista, mutta melkoinen osuus tuostakin nippelistä löytyy jo pelkän perustiedon kautta. Niitä ei vain osata tai tajuta kytkeä koiran ruokintaan. Abstract: CD45 is an abundant and highly active cell-surface protein tyrosine phosphatase (PTP) found on cells of hematopoietic origin. CD45 is of particular importance for T-cell function, playing a key role in the activation/inactivation cycle of the T-cell receptor signaling complex. The extracellular domain of CD45 is comprised of an N-terminal mucin-like domain which can be alternatively spliced to a core domain (RO) consisting of four domains with fibronectin 3 domain (FN3)-like topology. The study of CD45 has been hampered by a small set of publicly available antibodies, which we characterized as specific to the N-terminal FN3 domains of CD45 RO. To broaden the human CD45 reagent set, we identified anti-CD45 single domain VHH antibodies from a post-immune llama phage display library. Using a yeast display domain mapping system and affinity measurement we characterized seven unique clonotypes specific for CD45 RO, including binders that target each of the four FN3-like domains. "Bei uns bekommt jede Probe eine Art Barcode", so Seitz. Die WissenschafterInnen müssten dann nicht mehr jeden Test einzeln auswerten - was lange dauert - sondern könnten alle Proben gleichzeitig analysieren. Theoretisch Millionen auf einmal. Lexogen ist eines von mehreren vielversprechenden Startups, die im "Vienna Health Hub" (VHH) im 3. Wiener Bezirk angesiedelt sind. Der Health Hub ist ein "Accelerator". Er unterstützt besonders innovative Unternehmen aus dem medizinischen Bereich, ein Produkt zu entwickeln. VHH-CEO Irene Fialka kann eine ganze Menge von ihnen aufzählen, die sich jetzt dem Kampf gegen das Coronavirus bzw. Covid-19 verschrieben haben. A clinic participant asked a great question: “What do you do about a horse that ‘sets back’ when tied?” In this episode we talk about this from the perspective of safety and understanding the horse’s instincts, and offer specific suggestions about how to avoid the problem, and how to address it in a horse that’s already developed the habit. Preventing–or Curing–the Horse that “Sets Back” When Tied Some horses react badly to being tied up, using all their strength to pull against the restraint. Can this be prevented? Can it be “cured”? Why does a horse pull back when tied? It’s because the horse doesn’t respect the halter, so it all starts with proper halter training. Flight or fight response The key to understanding this problem is understanding the horse’s instincts. By evolutionary development, a horse is a prey animal whose first, preferred response to any threat is to flee–get away first, check it out later. When we tie up our horses, we take that defensive option away, leaving them only with their second choice: to fight. The objective of early training is to teach the horse to overcome that instinct to flee in response to the pressure they feel when tied. Prevention is best The keys to preventing this habitual response are: 1. Due diligence (understanding the horse’s natural, instinctive responses) and careful training. In this episode we outline the process of training the horse to respect the halter. 2. Don’t provoke the flight or fight instinct by tying too hard and fast (use a knot with some give). It’s a safety issue This response of setting back, or pulling back, is dangerous for us, for our equipment, and for our horses. We discuss some safety tips: 1.  Use a knot that can easily be untied. Check back here for a short instructional video that demonstrates how to tie the knot I recommend. 2.  Use a halter and lead rope with no metal fittings that can break. 3.  Always carry a safe type of knife that can quickly be used to cut the rope if necessary. Check back here for a photo of what I carry for this purpose.   What do you think? In the early episodes of Ride Every Stride we’re working on laying a foundation for what comes later. As I’ve said many times over the years, when it comes to horsemanship (and maybe life?), there’s nothing beyond the basics. For horsemanship, those basics are what I call the Absolute Basic Criteria. Picking up where we left off last time, in this episode of Ride Every Stride we’re talking about the second of those ABCs. The Absolute Basic Criteria of Horsemanship, part 2 — Control of Forward Movement As we said last week, even the most complex, sophisticated maneuvers on horseback are refinements of the basics. Understanding those basics–the ABCs of horsemanship–will equip us to work with our horses to accomplish whatever our horsemanship goals might be. What Are the ABCs of Horsemanship? forward movement (we discussed this in episode 2) control of forward movement stopping of forward movement (we’ll discuss this next time)   Once we’ve achieved the first of the ABCs–forward movement–we want to be able to control that forward movement, both its direction and its speed. In this episode we talk about what cues we use, when, and why. Directional Control One component of controlling forward movement is controlling its direction. We want to be able to turn our horses and get them to go where we want them to go. In learning the techniques for doing this, remember: our hands control what happens from our horse’s shoulders (withers) forward, and our legs control what happens from the withers back.   There’s no magic spot where your hands should be–you need to pay attention to your horse and make small adjustments as needed to get the response you want. Both by instinct and, hopefully, by good training, the horse will be inclined to move away from pressure, so think: what do I need to do to create an opening for my horse to go where I want him to go? We use the image of a river flowing from the horse’s hindquarters (where all the power comes from), through your legs (“big rocks on the banks of the river”) forward to the bit. You can control the horse’s movement–the flow of the river–by moving the rock out of the way on the side you want the horse to go. In other words, if you want the horse to go right, it’s not that you push from the left, but that you take the pressure away from the right, opening a pathway for the horse to go where you want it to go. Speed Control Although beginning riders are concerned about a horse running off with them, I’m more concerned about a horse that won’t move, in part because of safety issues (like the story I tell about a confrontation between a well behaved, but a bit lazy, horse I was riding and a cow). I want my horses to be calm and well behaved, but when I signal that it’s time to go, they need to respond immediately. To train that habit, I use the approach I call “squeeze, bump, kick, and kill.” For your safety’s sake, you need to understand the concepts of speed (and directional) control we talk about in this episode. Use the horse’s instinctive desire to conserve energy to help her learn to follow your cues (make it harder for her to go at the pace she wants to go if it’s faster than you want her to go) Learn how to make sure you are the one establishing the rhythm, which helps set the pace   What do you think? As I’ve said many times over the years, when it comes to horsemanship (and maybe life?), there’s nothing beyond the basics. For horsemanship, those basics are what I call the Absolute Basic Criteria. In this episode of Ride Every Stride we’re talking about the first of those ABCs. The Absolute Basic Criteria of Horsemanship Even the most complex, sophisticated maneuvers on horseback are refinements of the basics. Understanding those basics–the ABCs of horsemanship–will equip us to work with our horses to accomplish whatever our horsemanship goals might be. What are the ABCs of Horsemanship? forward movement control of forward movement stopping of forward movement   Although most beginning riders are especially concerned about the last of those ABCs–stopping–we actually want our horses to move. Forward movement generally constitutes one of the three basic gaits: the walk, the trot, or the lope (or canter). If we understand the mechanics of each gait, we can cooperate with our horse to make it easier for her to move forward easily at the gait we want. In this episode Van shares a simple tip for knowing where your horse’s feet are without leaning over to look. Using this method keeps you centered on your horse’s back, which is safer for you and more comfortable for your horse, and helps you use a simple technique to encourage that forward movement that’s the starting point to accomplishing our horsemanship goals. Ride Every Stride is about empowering you with knowledge. Understanding the mechanics of how your horse moves is key to letting you work with your horse instead of against him.  Knowledge of how your horse works (and thinks) leads to greater confidence, which leads to greater enjoyment. It’s been months in the planning, but we are excited to finally launch Ride Every Stride, a podcast about horsemanship . . . and more. In episode 1 we share a little bit about who I am, what my horsemanship background is, and what you can expect from me and this podcast. In this and future episodes, you’ll get lots of practical horsemanship tips–tools you can take with you to the barn and put to use to improve your working relationship with your horse. But I hope you’ll also be inspired as we talk about the many ways the concepts and principles we use to improve our horsemanship also apply to our journey as humans. As I’ve said many times, horsemanship is an opportunity to practice humanship every day. My purpose in creating this podcast is simple: to serve you. I can do that best if you let me know how. What questions do you have? What challenges are you facing in your growth as a horseman or horsewoman? What topics would you like to hear me cover in future episodes? Mitä suosittelette tankkaukseen, jos haluaa noudattaa paleo-tyyppistä ja nk. hiilihydraattitietoista ruokavaliota ja välttää turhia sokereita myös ennen suorituksia ja niiden aikana. Ketoosiin ei ole tarkoitus pyrkiä eikä noudattaa VHH dieettiä vaan välttää keinotekoisia hiilihydraattilähteitä myös urheillessa. Lihakset ja maksahan ottavat vain rajoitetun määrän hiilareita vastaan, joten energiaa pitäisi saada irti rasvoista. Kuulisin mielellänne ajatuksianne tankkaamisesta ennen urheilusuoritusta ja mitä tehdä, jos tuntuu, että puhti loppuu suorituksen aikana. Generation and characterization of heavy chain antibodies derived from Camelids

Antibodies and antibody fragments are essential tools in basic research, diagnostics and therapy. Conventional antibodies consist of two heavy and two light chains with both chains contributing to the antigen-binding site. In addition to these conventional antibodies, camelids (llamas, alpacas, dromedaries and camels) possess so-called heavy chain antibodies (hcAbs) that lack the light chains. The antigen binding site of these unusual antibodies is formed by one single domain only, the so called VHH domain. The VHH domain represents the smallest intact antigen binding fragment (~ 15 kDa) and is characterized by very high stability, solubility and specificity. These unique features render VHHs a promising alternative to conventional antibodies and antibody fragments with a multitude of possible applications. In the course of this thesis, we aimed to generate and characterize VHHs suitable for different applications ranging from biochemical studies to immunofluorescence assays and live cell imaging. In order to meet the diverse requirements of the intended downstream applications, a new selection method differing from traditional phage display, called native panning, was developed and established. In combination with the protein-protein interaction fluorescent two-hybrid (F2H) assay, a new process to identify antigen specific VHHs functional inside living cells was developed. We used the immune system of alpacas to generate VHH libraries against various antigens, ranging from small peptides to large proteins. The libraries were screened by phage display and antigen specific VHHs were identified by phage ELISA. We were able to identify antigen specific VHHs against different antigens which were characterized and their functionality was tested in various applications. Furthermore, we could demonstrate that the selection method influences which VHHs are identified and therefore needs to be chosen very carefully with regard to the intended biochemical and cell biological application. In summary, we developed and established an efficient and versatile process to screen and identify antigen specific VHHs suitable for different downstream applications.
