This training class builds on the earlier information for fate and transport and site characterization presented in the PFAS 101 CLU-IN training. It provides more in-depth information for fate and transport, site characterization, source identification and some introductory information on environmental forensics. These topics will be presented along with options and a framework for data visualization. This training will focus largely on PFAS fate and transport in groundwater. The goal is to provide detailed information about the inputs practitioners can use to develop a robust conceptual site model to help understand fate and transport at PFAS sites, and how to gather evidence of multiple sources. Resources and further details for the topics included in this training are in the ITRC Guidance Document (PFAS-1) in Sections 2.6, 3, 4, 5, 10, 11 and 15 along with referenced tables. Our next poster is titled Characterization of Concussions in the National Basketball Association and the 90-day Period Following Return to Play: Analysis of Musculoskeletal Injury Risk and Player Performance. This study was performed at the Mayo Clinic in Arizona and sought to determine the incidence of musculoskeletal injury in the immediate 90-day period following return to play from concussion. The secondary outcomes were to evaluate player performance within the 90-day. As well as time lost after musculoskeletal injury.Concussions have steadily garnered increased attention over recent years. We actually did an episode with Dr. Rachel Frank focused on concussions and sideline evaluation. This is episode 98 if you want to go check it out. That episode focused primarily on identifying and appropriately managing concussions. This study aims to evaluate the impact of a concussion on risk of musculoskeletal injury. Concussions have previously been shown to have persistent neurological effects that may lead to slower reaction times and may compromise postural stability in high-level athletes. Therefore, it stands to reason that a player may be at greater risk for musculoskeletal injury and may exhibit poorer performance upon return from a concussion.The authors identified NBA players who sustained a concussion between 2015-20 22 using a publicly available database. The database was also queried to identify any musculoskeletal injury in the 90-day period following return to play after the concussion, as well as time lost after the subsequent injury. Performance statistics were obtained from each players preinjury season and post-injury season. Each concussed player was matched with a non-concussed control player using position, win shares, player efficiency rating and points per game as metrics.So, what did this study find? A total of 70 concussions were identified during this time period. 70% of concussed players sustained a musculoskeletal injury in the 90-day period following return to play. Compared with controls, the odds of sustaining a musculoskeletal injury in the concussed cohort were 11.3 times greater. There was no significant difference between the type and location of injury, with ankle injuries being the most common in both groups.Now let's talk about performance. Interestingly, there were no significant changes in points per game, minutes played per game or true shooting percentage between the concussed and control groups. When compared with the controls, no changes in performance statistics were significantly different. Games missed after subsequent musculoskeletal injury were similar between the concussed and control groups.

BUFFALO, NY—February 12, 2025 — A new #research paper was #published in Aging (Aging-US) on January 3, 2025, in Volume 17, Issue 1, titled “Characterization of DNA methylation clock algorithms applied to diverse tissue types.” Researchers Mark Richardson, Courtney Brandt, Niyati Jain, James L. Li, Kathryn Demanelis, Farzana Jasmine, Muhammad G. Kibriya, Lin Tong, and Brandon L. Pierce from the University of Chicago and University of Pittsburgh, studied how biological aging is measured using DNA. Their study found that while commonly used “DNA methylation clocks” work well for blood samples, they may be less reliable for other tissues like the lungs, colon, and kidneys. DNA methylation clocks are widely used in forensic science, epigenetics and longevity research to estimate a person's biological age based on chemical changes in DNA. These epigenetic clocks help scientists predict age-related diseases and assess how lifestyle factors, such as smoking, impact aging. Most were originally developed using blood samples, and their effectiveness in other tissues remains unclear. This study tested eight different DNA methylation clocks across nine human tissue types, including the lungs, prostate, ovaries, skeletal muscle, and kidneys. The researchers analyzed data from 973 tissue samples collected through the Genotype-Tissue Expression (GTEx) project. Their analysis revealed significant differences in biological age estimates across tissues. While blood samples provided the most reliable results, other tissues showed noticeable variations. For example, testis and ovary tissues appeared younger than expected, while lung and colon tissues appeared older. “These differences across tissue types were most apparent for clocks trained using DNAm from blood only (e.g., Hannum), but also present for clocks trained on multiple tissue types (e.g., Horvath, a clock designed for pan-tissue age prediction.” These findings suggest that aging may not occur at the same rate in every organ, and that standard DNA methylation clocks may not always give accurate biological age estimates outside of blood samples. The authors suggest that new, organ-specific epigenetic clocks may be needed to improve biological age prediction. Creating tissue-specific aging clocks could also improve medical diagnostics, age-related disease prevention, and health monitoring. The researchers emphasize that larger studies with more tissue-specific DNA methylation data are needed to refine these aging clocks. By improving these tools, scientists can better understand how aging affects different organs and develop more reliable methods for measuring biological age. In 2020, ITRC's 1,4-Dioxane team created multiple tools and documents that provide information to assist all interested stakeholders in understanding this contaminant and for making informed, educated decisions. Since the 1950s, 1,4-Dioxane has seen widespread use as a solvent stabilizer. The use of solvents through the 1980s suggests its presence at thousands of solvent sites in the US; however, it is not always a standard compound in typical analytical suites for hazardous waste sites, so it previously was overlooked. The U.S. EPA has classified 1,4-Dioxane as "likely to be carcinogenic to humans." Some states have devised health standards or regulatory guidelines for drinking water and groundwater standards; these are often sub-part per billion values. These low standards present challenges for analysis, characterization, and remediation of 1,4-Dioxane. The 1,4-Dioxane: Science, Characterization & Analysis, and Remediation training is a series of six (6) modules. The six individual modules will be presented together live, and then archived on the ITRC 1,4-Dioxane training webpage for on demand listening. My son and I love a few certain characters from the books we've read aloud over the years. Gum-Baby, from Tristan Strong, Boots, from Gregor the Overlander, Maniac Magee. For my daughter, it's Junie B. Jones and Ramona from their named series collections. For me, it was always Anne (of Green Gables) I returned to growing up, and Jo from Little Women. Oh, and of course, Calvin from Calvin and Hobbes. Incredible characters are everywhere we turn in literature, and they make such an impact on us. We see through their eyes, experience their transformations, build empathy through their experiences. Maybe that's why when I think about characterization, I tend to think about activities that showcase characters visually. That come at them from many angles. That require students to consider their evolution, their growth, their nature vs. their nurture. Because sure, by all means, let's talk about what it means to be flat or round, static or dynamic. But then let's go much further. Today on the podcast, I'm sharing six creative characterization projects I've come up with over the years, in hopes that one (or two, or three) will fill a hole for you. Over 2,000 garbage trucks worth of plastic waste enter our oceans, lakes, rivers, and estuaries every day. Of the 300 million tons of plastic produced annually, only 10% is recycled. Due to the inertness and durability of plastic materials, current industrial recycling processes employ energetically demanding processes such as combustion and melting which further the global climate crisis by releasing greenhouse gases into the atmosphere. As such, enzymatic degradation of plastic has emerged as a promising alternative to traditional plastic recycling approaches since enzymes operate under significantly milder and less energetically demanding conditions. The Caribbean presents a fascinating paradox in the sea turtle conservation story. On one hand, Costa Rica hosts the world's longest running and most iconic sea turtle conservation initiative. Meanwhile, just up the coast in the neighbouring Nicaragua, one of the world's few legal sea turtle fisheries flourish.So how does this contradiction play out for sea turtles of the Caribbean? As we know by now sea turtles are global citizens, caring not for our political boundaries. The Caribbean green turtles are no different, for they nest in massive numbers in Costa Rica and forage in Nicaragua. Many listeners of this show will be familiar with precision fermentation, or turning microbes into factories to produce proteins like those proteins that have historically been produced inside of chickens and cows. Think of companies whose founders we've had on, like Perfect Day and The Every Company. But, what if instead of using microbes as protein factories—and all the associated costs of bioreactors and other capex—you could simply turn plants into protein factories, and make actual animal proteins inside of the plants, which can then be extracted and sold? That's exactly what Israeli startup PoLoPo is doing inside of potatoes. Their first protein: Ovalbumin, or the protein that makes up most of the egg white's protein content. If you pay attention to ingredient decks on food packaging, you've probably noticed that albumin is an ingredient in many foods, often serving to help color and texturize foods, as well as serving as a high-quality source of protein. In fact, the global egg albumin market is valued at billions of dollars, with some estimates around $5 billion USD and others as much as $30 billion USD. Founded in 2022, PoLoPo has already raised a couple million US dollars to scramble that market with real egg proteins grown inside of potatoes. Since the process is totally animal-free, it should go over easy as a vegan ingredient, but since it's an actual egg protein, those with egg allergies will still want to avoid cracking open a food with PoLoPo's Ovalbumin. In this episode, PoLoPo CEO Maya Sapir-Mir and I chat about her work as a plant biologist, how she teamed up with a vegan scientist to co-found this company, her passion for using bioengineering to help save the planet, and of course, how she plans to use the humble potato to displace some of the need for chickens in our food industry. More about Maya Sapir-Mir Maya Sapir-Mir is CEO and co-founder of PoLoPo, a molecular farming pioneer producing proteins directly in common crops, beginning with egg protein (ovalbumin) grown in potatoes. She has nearly ten years of experience in the biotech industry and agricultural R&D, including senior management at a small cannabis industry startup. In addition to leading R&D on plants with commercial and medical applications, she managed collaborations with partners and customers. She holds a PhD in plant sciences and an MSc in plant genetics from the Hebrew University of Jerusalem, and a BSc in biochemistry from Ben-Gurion University of the Negev. She performed post-doctoral work at the Volcani Institute, Israel's leading agricultural R&D facility, creating a new area of research for the organization in Protein Identification, Extraction, and Characterization in plants and microorganisms.
