Despite advancements in genetic testing, people with rare diseases often face a prolonged diagnostic odyssey involving multiple physician visits and misdiagnoses. Genetic testing company GeneDX is working to shorten the path to a diagnosis by expanding access to sequencing, collaborating with researchers, and accumulating data to better understand gene-disease relationships. We spoke to Katherine Stueland, CEO of GeneDx, about the state of genetic testing, what its 2022 acquisition by the AI-drive genomics company SEMA4 has meant to it, and what she thinks it will take to make meaningful change to the diagnostic odyssey.

As the summer was ending,I sat down with Isaac Ro, Life Sciences Executive, who I originally met at the Blue Genes Bash several years ago.  Isaac left Goldman Sachs for Thrive Early Detection, where Bert Vogelstein and the group were working on liquid biopsy.  After Thrive was acquired by Exact Sciences, Isaac left and became CFO at Sema4, which was a company where I worked as well.  After taking Sema4 public, Haystack oncology is his newest venture which works on MRD in cancer.  We talk data, the financial climate in genetics, and what it's like to pivot to a management role where you're relying on everyone in the team for company success.

Called the GUARDIAN study, the project is the brainchild of Wendy Chung, Professor of Pediatrics at Columbia University. The study will take place in New York State and is somewhat similar to an ongoing project in the U.K. being done by Genomics England.

Today, we hear about an 80 year tradition at the Bethlehem Connecticut post office that has earned it the nickname Christmastown. Thousands of cards and packages come through this tiny post office every year to get one of their cachets. On Friday, the Connecticut Mirror reported that Sema4, the laboratory based in Stamford responsible for managing 15 out of the 23 state testing sites, has pulled out of it's contract with the state. The company will exit the testing business by mid-January. With the holidays coming up and latest surge in cases, this could not have come at a worse time. Connecticut Mirror investigative reporter Dave Altimari joins us. Nevertheless, the improving affordability of newborn screening is expanding access to these tests for parents seeking them. We spoke to Eric Schadt, CEO of Sema4, about the changing landscape for newborn screening, the company’s Natalis test, and how far off we are from routine screening of newborns with comprehensive genetic sequencing.

"We like to refer to it as consumer initiated, but physician supervised,” says Eric Schadt today when asked if his new test is direct-to-consumer. Eric is the Dean for Precision Medicine at the Icahn School of Medicine at Mount Sinai in New York and since 2016 has served as CEO of Sema4, a spinoff that he founded out of the Mount Sinai Health System. Sema4 launched a newborn screening panel, Sema4 Natalis, in February of this year covering over 190 disorders.

During the course of his 20-year career, Dr. Eric Schadt has built genetics and systems biology groups at Merck. He built the computational biology group at Rosetta. He has served as co-founder of Sage Bionetworks and as Chief Science Officer of Pacific Biosciences. He now serves as the founder and CEO of Sema4. He has published more than 300 peer-reviewed papers in leading scientific journals, and he has contributed to a number of discoveries relating to the genetic basis of common human diseases such as diabetes, obesity and Alzheimer’s. In today’s conversation, we explore the information technology of biology – DNA – the world of genomics, where big data looms large. We begin by mapping the territory of the human genome and exploring the pathways of disease. We look to understand the ways in which complex genetic combinations express themselves as phenotypes such as height, bone structure, intelligence, and personality. How are these traits coded for? What are the instructions our body uses to repair a damaged cell? What blueprint does it consult before trying to grow new arteries? How does it know to regulate our appetite or when start us down the path of puberty? What happens when these instructions are damaged? How can the smallest difference in the order of life’s code make all the difference for our success, our happiness, and even our survival? 50 years have passed, between the discovery of the double-helix and the mapping of the first human genome.
