It's Hump Day! Sam speaks with Sarah Lazare, editor of Workday Magazine and contributing editor at In These Times, to discuss her recent reporting on the UAW fighting to restore a shuttered Stellantis plant in Belvidere, Illinois. Sarah Lazare then joins, diving right into a history of Stellantis' plant in Belvidere, Illinois, from its opening in 1965 and through its peak in the late 2010s – when it employed nearly a third of the town's population – before the Corporation-formerly-known-as-Chrysler decided to idle the plant in 2023, essentially taking away thousands of jobs from the city. Continuing, Lazare then unpacks the plant's role in UAW negotiations with Stellantis in the wake of their overwhelmingly successful stand-up strike against the Big Three automakers (Stellantis, Ford, GM), with the reopening of the Belvedere plant one of many unprecedented wins for the union and emblematic of their emphasis on bringing workers into the manufacturing decisions that shape their lives. After briefly expanding on the details of the UAW's major wins from the stand-up strike, as well as some concessions, Sarah walks Sam through the devastating impact the idling of this plant has had on Belvidere, the incredible effort by other locals and the broader UAW to use their labor to demand the return of this plant, and the likelihood of the fight expanding into a broader strike, before wrapping up with a conversation on the ripple effect of labor rights and the importance of solidarity in building a better life for us all. BUFFALO, NY- March 20, 2024 – A new #research paper was #published in Oncotarget's Volume 15 on March 14, 2024, entitled, “Analytical validation of NeXT Personal®, an ultra-sensitive personalized circulating tumor DNA assay.” In this new study, researchers Josette Northcott, Gabor Bartha, Jason Harris, Conan Li, Fabio C.P. Navarro, Rachel Marty Pyke, Manqing Hong, Qi Zhang, Shuyuan Ma, Tina X. Chen, Janet Lai, Nitin Udar, Juan-Sebastian Saldivar, Erin Ayash, Joshua Anderson, Jiang Li, Tiange Cui, Tu Le, Ruthie Chow, Randy Jerel Velasco, Chris Mallo, Rose Santiago, Robert C. Bruce, Laurie J. Goodman, Yi Chen, Dan Norton, Richard O. Chen, and John M. Lyle from Personalis, Inc. describe the analytical validation of NeXT Personal®, an ultra-sensitive, tumor-informed circulating tumor DNA (ctDNA) assay for detecting residual disease, monitoring therapy response, and detecting recurrence in patients diagnosed with solid tumor cancers. “NeXT Personal uses whole genome sequencing of tumor and matched normal samples combined with advanced analytics to accurately identify up to ~1,800 somatic variants specific to the patient's tumor.” A personalized panel is created, targeting these variants and then used to sequence cell-free DNA extracted from patient plasma samples for ultra-sensitive detection of ctDNA. The NeXT Personal analytical validation is based on panels designed from tumor and matched normal samples from two cell lines, and from 123 patients across nine cancer types. Analytical measurements demonstrated a detection threshold of 1.67 parts per million (PPM) with a limit of detection at 95% (LOD95) of 3.45 PPM. NeXT Personal showed linearity over a range of 0.8 to 300,000 PPM (Pearson correlation coefficient = 0.9998). Precision varied from a coefficient of variation of 12.8% to 3.6% over a range of 25 to 25,000 PPM. The assay targets 99.9% specificity, with this validation study measuring 100% specificity and in silico methods giving a confidence interval of 99.92 to 100%. “In summary, this study demonstrates NeXT Personal as an ultra-sensitive, highly quantitative and robust ctDNA assay that can be used to detect residual disease, monitor treatment response, and detect recurrence in patients.” DOI - https://doi.org/10.18632/oncotarget.28565 Correspondence to - John M. Olympic silver medallist Dan Norton, born in Gloucester, is the the all-time record try scorer in the World Rugby Sevens Series, crossing the tryline 358 times for England over the course of 92 tournaments, totalling an enormous 1804 points in total (including 7 conversions - a 100% kicking record, he tells me!).Norton bowed out of the game as the fourth highest points scorer of all time, going down in history as a legend and one of the greatest sevens players ever, also playing for Team GB at two Olympic games.Shortly after we spoke, Dan announced his retirement from sevens after a 13-year career, and broke the news of a move into coaching, with China's rugby sevens squad.Dan's was the first episode recorded - back in spring 2022 - when I decided to embark on starting this podcast. Griffin captained Team USA at the Olympic Games in 2016, when rugby returned after a 92-year hiatus. Premier Rugby Sevens features 8 franchises with 16 teams in 2023 contesting five tournaments across the U.S. The season started on June 17 at Q2 Stadium (Austin, Texas) for the Eastern Conference Kickoff, then TCO Stadium (Minneapolis, Minnesota) hosted the Western Conference Kickoff on June 24. The Western Conference Finals are on July 15 at Paypal Park (San Jose, California) and the Eastern Conference Finals on July 23 at Highmark Stadium (Pittsburgh, Pennsylvania). The 2023 Championship will be played on August 6 at Audi Field in Washington, D.C., and will feature the top two teams from each Conference.
