"The Girl Who Cried Pearls" is a Canadian animated short film, directed by Chris Lavis and Maciek Szczerbowski. Made through stop-motion animation, the film centers on a young boy who meets and falls in love with a girl who secretly weeps pearls at night. The initial inspiration for the film came during the production of the duo's 2007 short film "Madame Tutli-Putli," when a prop necklace unexpectedly fell apart, scattering pearls across the set as they filmed the climactic scene. The film has received critical acclaim and is nominated at the 98th Academy Awards in the category of Best Animated Short Film. Franco-Belgian-Luxembourgish artist Marie Lavis joined Simon on the Breakfast Show today. Marie is an illustrator, animator, graphic designer who is keen to build bridges between visual art and music. Abstract: One-third of the mammalian proteome is comprised of transmembrane and secretory proteins that are synthesized on endoplasmic reticulum (ER). Here, we investigate the spatial distribution and regulation of mRNAs encoding these membrane and secretory proteins (termed 'secretome' mRNAs) through live cell, single molecule tracking to directly monitor the position and translation states of secretome mRNAs on ER and their relationship to other organelles. Notably, translation of secretome mRNAs occurred preferentially near lysosomes on ER marked by the ER junction-associated protein, Lunapark. Knockdown of Lunapark reduced the extent of secretome mRNA translation without affecting translation of other mRNAs. Less secretome mRNA translation also occurred when lysosome function was perturbed by raising lysosomal pH or inhibiting lysosomal proteases. Secretome mRNA translation near lysosomes was enhanced during amino acid deprivation. Addition of the integrated stress response inhibitor, ISRIB, reversed the translation inhibition seen in Lunapark knockdown cells, implying an eIF2 dependency. Altogether, these findings uncover a novel coordination between ER and lysosomes, in which local release of amino acids and other factors from ER-associated lysosomes patterns and regulates translation of mRNAs encoding secretory and membrane proteins. Abstract: Genetically encoded fluorescent calcium indicators have revolutionized neuroscience and other biological fields by allowing cellular-resolution recording of physiology during behavior. However, we currently lack bright, genetically targetable indicators in the near infrared that can be used in animals. Here, we describe WHaloCaMP, a modular chemigenetic calcium indicator built from bright dye-ligands and protein sensor domains that can be genetically targeted to specific cell populations. Fluorescence change in WHaloCaMP results from reversible quenching of the bound dye via a strategically placed tryptophan. WHaloCaMP is compatible with rhodamine dye-ligands that fluoresce from green to near-infrared, including several dye-ligands that efficiently label the central nervous system in animals. When bound to a near-infrared dye-ligand, WHaloCaMP1a is more than twice as bright as jGCaMP8s, and shows a 7x increase in fluorescence intensity and a 2.1 ns increase in fluorescence lifetime upon calcium binding. We use WHaloCaMP1a with near-infrared fluorescence emission to image Ca2+ responses in flies and mice, to perform three-color multiplexed functional imaging of hundreds of neurons and astrocytes in zebrafish larvae, and to quantitate calcium concentration using fluorescence lifetime imaging microscopy (FLIM). Luke Lavis is a senior group leader and head of molecular tools and imaging at HHMI's Janelia Research Campus. Luke Lavis works at the interface of chemistry and biology, developing small-molecule fluorescent dyes to illuminate biological samples. Lavis and his team use modern organic chemistry to design and synthesize bright fluorescent labels for imaging structures inside living cells. By combining these novel compounds with advances in instrumentation, protein engineering, and genetic manipulation, the Lavis Lab and their collaborators devise sophisticated bioimaging experiments at different scales ranging from tracking single molecules to measuring neural activity in cells and animals. “I sometimes call Luke our secret weapon here. Inoltre conoscono e comprendono i vari tipi di sistemi di irrigazione e come funzionano, in modo da poter installare il sistema corretto e riparare tutto ciò che non funziona correttamente. Un tecnico dell'irrigazione potrebbe essere responsabile dell'irrigazione di fattorie, parchi, strutture, giardini di hotel o campi da golf. In alcuni casi, a un tecnico esperto di irrigazione potrebbe essere richiesto di progettare un sistema di irrigazione e dovrà soddisfare le esigenze del terreno, nonché eventuali specifiche locali e federali. Molti elementi contribuiscono a un sistema di irrigazione altamente efficiente, come il tempo di irrigazione, il tipo di vegetazione, il suolo e il clima. Problemi come l'irrigazione eccessiva, le perdite sotterranee e la carenza d'acqua sono problemi con cui i tecnici dell'irrigazione potrebbero dover affrontare. In qualità di esperti nella risoluzione dei problemi quando si tratta di acqua e dei danni che può causare, agiscono in modo rapido ed efficiente per mitigare il problema il più rapidamente possibile. I tecnici dell'irrigazione sono abili nell'utilizzare vari tipi di macchinari come falciatrici, trattori o trencher (una macchina che installa linee di irrigazione). Azionano anche programmatori di irrigazione, tubi e teste di irrigazione, oltre a lavorare con tutti i cavi elettrici necessari per far funzionare il sistema. La tecnologia più recente consente ai tecnici dell'irrigazione di monitorare le linee di irrigazione da un computer, in modo che possano trovare un problema più velocemente e anche tenere traccia delle prestazioni del sistema di irrigazione. Un'eccellente tenuta dei registri è essenziale per tutte le attività di gestione dell'irrigazione e per gli ordini completati. I tecnici lavorano anche con i gestori di proprietà o i proprietari per garantire il corretto coordinamento delle attività. Poiché le esigenze di un'azienda o di un'organizzazione possono variare a seconda dell'occasione o della stagione, sono responsabili del mantenimento della comunicazione e di un forte rapporto di lavoro con il proprietario o il gestore.
