In this episode of the mushroom show, we talk to Kevin Feeney who is the author of Fly Agaric: A Compendium of History, Pharmacology, Mythology & Exploration. He is an expert on the iconic Amanita muscaria (the mushroom emoji, in case you don't know) and we delve into the fascinating facts and myths about this mushroom, including his personal experience with it's psychoactive effects. Kevin Feeney, PhD, JD, is a cultural anthropologist and lawyer currently working as a Program Director and Senior Lecturer in Interdisciplinary Studies – Social Sciences at Central Washington University. His primary research interests include examining legal and regulatory issues surrounding the religious and cultural use of psychoactive substances, with an emphasis on peyote and ayahuasca, and exploring modern and traditional uses of Amanita muscaria, with a specific focus on medicinal use and preparation practices. His research has been published in the International Journal of Drug Policy, Journal of Psychoactive Drugs, Human Organization, and Curare, among other books and journals. He is a current member of Chacruna's Council for the Protection of Sacred Plants, and recently joined the Board of Advisors for Psyched Wellness, a Canadian health supplements company emphasizing medicinal mushrooms. Kevin is a cultural anthropologist and lawyer currently working as a Program Director and Instructor in Interdisciplinary Studies – Social Sciences at Central Washington University. His primary research interests include examining legal and regulatory issues surrounding the religious and cultural use of psychoactive substances, with an emphasis on peyote and ayahuasca, and exploring modern and traditional uses of Amanita muscaria, with a specific focus on medicinal use and preparation practices. His research has been published in the International Journal of Drug Policy, Journal of Psychoactive Drugs, Human Organization, and Curare, among other books and journals. C., Dorr, F. A., Pinto, E., Adachi, F. M. M., Hotta, C. T., Bastos, E. L. Abstract: L-DOPA extradiol dioxygenases (DODAs) catalyze the production of betalains and hygroaurins pigments. The sequence of the DODAs found in Caryophyllales and Basidiomycetes are not conserved, although betalains are produced both by plants and fungi. Here we revise the coding region of the dodA gene of fly agaric [Amanita muscaria (L.) Lam.] and describe an alternative start codon downstream that enables the heterologous expression of AmDODA, a promiscuous L-DOPA dioxygenase. AmDODA is 43-amino acid residues shorter than the recombinant DODA previously reported but catalyzes the formation of two isomeric seco-DOPAs that are the biosynthetic precursors of betalains and hygroaurins. The putative active site of AmDODA contains two distinct His-His-Glu motifs that can explain the dual cleavage of L-DOPA according to the mechanism proposed for non-heme iron-dependent dioxygenases. Upon addition of excess L-DOPA, both the betaxanthin and hygroaurin adducts of L-DOPA are produced. The kinetic parameters of enzymatic catalysis at pH 8.5 are similar to those reported for other L-DOPA dioxygenases. The rate constants for the conversion of L-DOPA into the betalamic acid and muscaflavin were estimated by kinetic modelling allowing the proposal of a mechanism of pigment formation. These results contribute to understanding the biosynthesis of bacterial, fungal and plant pigments, for the biotechnological production of hygroaurins, and for the development of more promiscuous dioxygenases for environmental remediation. Though incredibly versatile, the sustained energy in his sets and productions are equally exceptional. In the last two years, Agaric established the Club Tracks 10” series on his label We Are. Besides his own stellar productions, collaborations with close friends Shonky, Walker Barnard, Ed Davenport, and Beaner have also graced the Club Tracks series. International support came from leading DJs like Richie Hawtin, Seth Troxler, Loco Dice, Ben Klock, Luciano, Joris Voorn, Magda, Laurent Garnier, Sven Väth, and Adam Beyer. Above all, his productions were championed by Josh Wink, who went on to sign his album debut to his Ovum imprint. After more than 100 vinyl releases, Patrik Skoog can look back at quite a history: His harder Techno project Headroom on his label Native Diffusion, releases on Synewave, Loop, and many more long forgotten underground labels in the 90s and early 2000s left his first mark for those who know. From 2005 on, he evolved into his deeper Agaric project, with releases on his internationally celebrated We Are label, Adam Beyer’s Drumcode, Truesoul & Mad Eye, Raum…Musik, Sunset Diskos, Nummer, Kontra Musik, Treibstoff and Liebe Detail. As a technical DJ with a magical sense for momentum and selection, he’s been playing worldwide for more than 10 years, while his equally as stellar Live sets are one of the few that can rock a crowd throughout its duration. Relocating to Berlin in 2007, he quickly established himself as a celebrated constant inspiration, be it for extended Bar25 after hour sets, or club nights in Watergate, Arena, and Berghain’s Panoramabar, where he now holds his We Are label nights.
