Pro bono publico, a Latin phrase meaning “for the public good,” is most often associated with free legal services lawyers provide to people who need help but cannot afford to pay for it. No lawyer I know better reflects that commitment than Boston lawyer Susan M. Finegan, a Partner and Chair of the Pro Bono Committee of the Mintz law firm. In this episode of Higher Callings, I asked Sue about the pro bono work she and other Mintz lawyers perform. We also discussed a number of other topics about pro bono legal services, including why lawyers and other legal professionals choose to engage in pro bono work, how law firms incentivize their lawyers to take on pro bono cases, how working on pro bono cases provides important training for new lawyers, what Massachusetts has done to provide pro bono opportunities for retired lawyers, and how in-house corporate lawyers and government lawyers work with law firms to staff pro bono cases. These require robust but dynamic linkage between the cell-cell adherens junctions and the force-generating actomyosin cytoskeleton. Our view of this linkage has evolved, and we now realize linkage is mediated by a mechanosensitive multiprotein complex assembled via multivalent connections. Here we combine genetic, cell biological and modeling approaches to define the mechanism of action and functions of an important player, Drosophila Polychaetoid, homolog of mammalian ZO-1. Our data reveal that Pyd reinforces cell junctions under elevated tension, and facilitates cell rearrangements. Pyd is important to maintain junctional contractility and in its absence cell rearrangements stall. We next use structured illumination microscopy to define the molecular architecture of cell-cell junctions during these events. The cadherin-catenin complex and Cno both localize to puncta along the junctional membrane, but are differentially enriched in different puncta. Pyd, in contrast, exhibits a distinct localization to strands that extend out from the region occupied by core junction proteins. We then discuss the implications for the protein network at the junction-cytoskeletal interface, suggesting different proteins localize and function in distinct ways but combine to produce robust connections. Finegan and Jason of Steelbow Farm, in Austin TX, may seem like unusual guests for the Winter Growers Podcast. But, with climate change and more frequent extreme weather patterns, I was curious to hear from a farm down at the 30th parallel where farming from November through March is more common. However, it is still full of challenges, such as extreme daily temperature swings. Finegan and Jason first farmed together in Austin before deciding to start their first farm in Maine, where they leased land for three seasons and sold mostly into the Portland area. After coming up short in their search for permanent farmland, they reached out to a farmer farm mentor in Austin who offered to lease them farmland and infrastructure. It was too good of an offer to refuse, so they took the leap and moved their farm BACK down to Texas. In late 2019, they packed their bags, along with their first child, and moved to Austin to begin their new farm adventure. In the last two years, and during a pandemic no less, they have rebuilt Steelbow Farm, now cultivating 2.5 acres, and sell to over 20 restaurantes with a 130+ member CSA. They also have 7 employees, prioritize paying them well, with quality, community, and communication as as central focus in all that they do. Year round growing is primary to their farming model, but it's not without it's challenges, even in Texas. I hope this conversation offers a wider perspective on what it means to farm year-round, no matter where the farm is located. Mentioned in the show...
