Today we're talking to Miriam BC Tobin, winner of the Goddard Alumni Association Spirit Grant for her work with SCRiB Lab, a writing organization aimed at creating community through experimentation. SCRiB Lab provides writing labs in three categories: SCRiBE for literary writers; SCRiPT for dramatic writers; and SCRiBBLE for ideas seekers. Besides creating a playful space for writers, Miriam is a contemporary absurdist playwright, interested in using the grandiose and extraordinary to explore human truths. We talk about Absurdism with a capital A in theater: how absurdism (like rocks starting to talk) can be used not to be silly or funny, but to make a point about human existence. Miriam says, “Absurdism is this idea, or this concept of what happens when a human faces the greater universe. And is that space in between ‘I am a small little human' and the devastation of seeing the great expanse of the universe.” We also talk about how the themes of Absurdism relate to classical Greek and Roman theater, in how humans relate to nature. At Goddard, Miriam was exposed to pre-Columbian drama (particularly Mayan) and Noh drama, both of which influenced her thesis play. She was specifically drawn to how both of those eras engage the community/audience in the participation or spectacle of the plays, and how the stories continue beyond the end of the theater piece. Abstract: Axon initial segments (AISs) are specialized neuronal compartments crucial for action potential generation and neuronal polarity. However, a detailed understanding of the mechanisms regulating AIS structure and function has been hindered by an incomplete knowledge of its molecular composition. Here, we profile the AIS proteome and its dynamic changes in-depth during neuronal maturation. Our method capitalizes on antibody targeted proximity labeling with subcellular spatial specificity in fixed primary neurons. Among the AIS proteins labeled, we identified novel AIS components, including PHGDH and SCRIB. PHGDH and SCRIB are highly enriched in the AIS both in vitro and in vivo, and exhibit a periodic architecture like the axonal spectrin-based cytoskeleton. Furthermore, we show that neuronal PHGDH activity is necessary for AIS integrity and action potential initiation. This powerful and flexible approach defines the AIS proteome and provides a rich resource to elucidate the mechanisms regulating AIS structure and function. Copy rights belong to original authors. It's the end of an era: Apple has announced it is discontinuing the iPod, and we pour one out for "1,000 songs in your pocket." Can the iPhone ever match the level of escapism an iPod provides? What's your rate of inflation? We take the quiz together and compare where we're feeling the most strain when we're paying bills. Andrew's doomsday prepping has him stockpiling Dunkin' Coffee from Costco, and Pam is about to walk away from a business she's frequented for years due to its price increases. Related: the latest casualty of supply chain shortages is baby formula, and some "pro-lifers" want to gate keep formula on the basis of citizenship. How many friends do we need? The New York Times has some helpful data, and we share our own "friendship numbers." Have some friendships grown closer during the pandemic? More distant? 'Buy now, pay later' trends are sending some millennials and Gen Z'ers spiraling into debt. When is it worth financing a purchase, and when is it better to pay in full? With over a third of shoppers under 40 missing at least one payment, it's clear that 'paying later' is easier said than done. It's International Podcast Day! A day when podcasters around the globe celebrate their craft and their journey, regardless if they've been podcasting for 17 days or 17 years. Speaking of that last one… I'm going to break down the +17-year journey of one podcaster—me!—if only to prove that, for many of us, podcasting is quite a tangled path. My official entrance to podcasting started in October of 2004 with The Dragon Page. Soon after that we launched Slice of Scif, followed quickly by Wingin' It. After adding a podcast we didn't host called The Dragon Page With Class to our roster, we launched the first scifi podcast network called FarPoint Media. And it was still 2005! Parallel to all of that, I personally started working with authors to help them podcast, soon creating a network of “podiobooks”, a portmanteau of podcasting and audiobooks, if that wasn't obvious. Tha directly led to me co-authoring Podcasting For Dummies and Expert Podcasting Practices For Dummies.  parting ways with FarPoint Media in mid-2007, I launched two short-lived podcasts: Cult Cast and Found Things. You're better for not hearing either of them. And because I sometimes bet on the wrong trends, I also co-hosted Palm Fu, a podcast all about the Palm Pre smartphone, plus a podcast build on Google+ called The Books & Beer Hangout. You pays your money and you takes your chances. Some time in there, I decided the world needed more me so I did a limited run podcast called Evo at 11. A show that I'm very, very glad I pulled after 100 episodes. Apparently, I can be an asshole behind the mic. Who knew? (Kidding. I knew.)  Then my wife and I left the country, using a podcast to document our travels around the world. Then I found out it's a lot of work to produce a journalistic style podcast. But those travels led me to living in Bangkok where I became the co-host of the 2nd season of The Bangkok Podcast.  And Bangkok is where my podcast consultancy, Simpler Media, was born. I've been back in The States since 2018, and podcasting is very much what I do. Not just with Podcast Pontifications, but I also do some hired-gun hosting for RadioMD, am the editor for Sounds Profitable, and sit on the advisory board for Captivate.fm, Maps,fm, Scrib, and a few others. E., Montcouquiol, M., Sans, N. Abstract: Social behavior is a basic domain affected in several neurodevelopmental disorders. Indeed, deficits in social interest, interactions and recognition represent core symptoms of Autism Spectrum Disorder but are also found associated with a heterogeneous set of neuropsychiatric and rare disorders. The SCRIB gene that codes for the polarity protein SCRIBBLE has been identified as a risk gene for spina bifida, the most common type of open neural tube defect, found at high frequencies in autistic patients, as other congenital anomalies, while the deletions/mutations of the 8q24.3 region encompassing SCRIB genes is associated with multisyndromic and rare disorders. Nonetheless, the potential link between SCRIB and ASD-relevant social phenotypes has not been investigated yet. Hence, we performed an extensive behavioral characterization of the circletail line that carries a mutated version of Scrib. Scribcrc/+ mice displayed reduced social interest, lack of preference for social novelty and social reward, and reduced social habituation while other behavioral domains were unaltered. Social deficits were associated with reduced hippocampal volume, upregulation of ERK phosphorylation in specific hippocampal regions, together with increased c-Fos activity in the same brain areas. Importantly, the social alterations were rescued by both direct and indirect pERK inhibition. These results support a specific link between polarity genes, social behaviors and hippocampal functionality, thus suggesting a role for SCRIB in the etiopathology of neurodevelopmental disorders. Furthermore, our data demonstrate the crucial role of the MAPK/ERK signaling pathway, in underlying the social deficits induced by SCRIB mutation, thus supporting its relevance as a therapeutic target. The Latin root word scrib and its variant script both mean “write.” These roots are the word origin of a fair number of English vocabulary words, including scribe, describe, postscript, and manuscript.
