Abstract: Inflammation is closely associated with many neurodegenerative disorders. Yet whether inflammation causes or exacerbates neurodegeneration has been challenging to define because the two processes are so closely linked. Here we disentangle inflammation from the axon damage it causes by individually blocking cytotoxic T cell function and axon degeneration. We model inflammatory damage in mouse skin, a barrier tissue that, despite frequent inflammation, must maintain proper functioning of a dense array of axon terminals. We show that sympathetic axons control skin inflammation through release of norepinephrine, which suppresses activation of gamma delta T cells via the beta2 adrenergic receptor. Strong inflammatory stimulation in the form of the toll like receptor 7 (TLR7) agonist imiquimod (IMQ) causes progressive gamma delta T cell-mediated, Sarm-1-dependent loss of these immunosuppressive sympathetic axons, a positive feedback loop that removes a physiological brake on T cells, resulting in enhanced inflammation and inflammatory axon damage. Abstract: Modifications of mRNA, especially methylation of adenosine, have recently drawn much attention. The much rarer modification, 5-hydroxymethylation of cytosine (5hmC), is not well understood and is the subject of this study. Vertebrate Tet proteins are 5-methylcytosine (5mC) hydroxylases enzymes catalyzing the transition of 5mC to 5hmC in DNA and have recently been shown to have the same function in messenger RNAs in both vertebrates and in Drosophila. The Tet gene is essential in Drosophila because Tet knock-out animals do not reach adulthood. We describe the identification of Tet-target genes in the embryo and larval brain by determining Tet DNA-binding sites throughout the genome and by mapping the Tet-dependent 5hmrC modifications transcriptome-wide. 5hmrC-modified sites can be found along the entire transcript and are preferentially located at the promoter where they overlap with histone H3K4me3 peaks. The identified mRNAs are frequently involved in neuron and axon development and Tet knock-out led to a reduction of 5hmrC marks on specific mRNAs. Among the Tet-target genes were the robo2 receptor and its slit ligand that function in axon guidance in Drosophila and in vertebrates. Tet knock-out embryos show overlapping phenotypes with robo2 and are sensitized to reduced levels of slit. Both Robo2 and Slit protein levels were markedly reduced in Tet KO larval brains. Our results establish a role for Tet-dependent 5hmrC in facilitating the translation of modified mRNAs, primarily in developing nerve cells. Copy rights belong to original authors. I recently had the pleasure of talking with Dominic Heutelbeck about Attribute-Based Access Control. Dominic is a computer science professor leading the FTK (Forschungsinstitut für Telekommunikation und Kooperation e.V.) Research Institute in Dortmund, Germany.  In the first portion of our talk, Dominic explained what access control is and its importance. He also explained what Attribute-Based Access Control is and why one would want to use it. Abstract: A critical step for functional recovery from peripheral nerve injury is for regenerating axons to connect with their pre-injury targets. Reestablishing pre-injury target specificity is particularly challenging for limb-innervating axons as they encounter a plexus, a network where peripheral nerves converge, axons from different nerves intermingle, and then re-sort into target-specific bundles. Here, we examine this process at a plexus located at the base of the zebrafish pectoral fin, equivalent to tetrapod forelimbs. Using live cell imaging and sparse axon labeling, we find that regenerating motor axons from three nerves coalesce into the plexus. There, they intermingle and sort into distinct branches, and then navigate to their original muscle domains with high fidelity that restores functionality. We demonstrate that this regeneration process includes selective retraction of mistargeted axons, suggesting active correction mechanisms. Moreover, we find that Schwann cells are enriched and associate with axons at the plexus, and that Schwann cell ablation during regeneration causes profound axonal mistargeting. Our data provide the first real time account of regenerating vertebrate motor axons navigating a nerve plexus and reveal a previously unappreciated role for Schwann cells to promote axon sorting at a plexus during regeneration. While her family eventually left the practice behind, Yat-Yee found that she had both an aptitude and an attraction to the sport. In 2020, after multiple black belts and a 2015 World Championship win, Mrs. Chong has taken her passion for Tae Kwon Do and opened her own studio. Yat-Yee opened Axon Martial Arts Academy in Loveland, Colorado with the vision of teaching individuals and families this martial art she has mastered. Starting from shared spaces with dance studios and gyms, Yat-Yee began offering classes a few times each week, fitting them in between the schedules of the programs she shared facilities with. We wanted to go deeper on the caching library Nebulex, so we visited with the creator, Carlos Bolaños, to learn what prompted its creation. Nebulex takes a couple unique approaches to things. It supports a decorator pattern to indicate that a function should be cached without having to write the boilerplate code for reading and writing to the cache. Nebulex was inspired by Ecto, in that it supports multiple adapters to different backends like Redis, Cachex and even Horde! It also supports multiple caching strategies. Independent federal agencies sit awkwardly in our constitutional structure. When they engage in aggressive overreach, therefore, they should expect to see their actions challenged in court. This episode centers around two such challenges. In Axon v. FTC, a case the Supreme Court will hear this term, the plaintiff challenges the jurisdiction of the Federal Trade Commission's internal tribunal. And in Consumers' Research v. FCC, a set of cases currently in the federal courts of appeals, the petitioners challenge the Federal Communications Commission's power to raise money without congressional oversight. Our guest is Trent McCotter, a partner with Boyden Gray & Associates and the director of the Separation of Powers Clinic at the C. Boyden Gray Center at Antonin Scalia Law School.
