My name is John Yopp and I am running for Onslow County Sheriff as a Republican Candidate. I am a lifelong resident of Onslow County. I care deeply about the people that live in our community. My family has been living in Onslow County, NC since the 1790s. We even have a road named after us, Yopp Road. We are known as a hardworking and reliable family willing to help and serve our fellow citizens. For the past 20 years, I have faithfully dedicated my life to serving the people in this community. On September 11th, 2001, I had an interview with the Onslow County Sheriff's Office. While in the waiting room, I watched the atrocious acts of terrorism unfold in New York City on the tv screen. Despite the chaos, danger, and imminent threat, I watched the selfless bravery of police officers, EMS workers, and firefighters, as they ran into doomed buildings to save people they didn't even know, and this strengthened my commitment and propensity to serve. PRINCIPLED, CONSISTENT LEADERSHIP SERVING ONSLOW COUNTY In 2001, I began serving as a Correctional Officer in the Onslow County Jail. From there, I assumed the role of patrol deputy for the Onslow County Sheriff's Office and continued to work there for several years. I am currently a Sergeant for the North Topsail Beach Police Department. My entire law enforcement career has been spent working in Onslow County, doing my best to ensure the best possible outcome for each and every citizen I have been called to serve.
Today we talk to Dr. Justin Yopp about the widowed parent program he and a colleague began after noting a lack of group support particularly for young father's recently bereaved. https://widowedparent.org Dr. Yopp is a clinical psychologist and Assistant Professor in the Department of Psychiatry at UNC. He is a member of the UNC Comprehensive Cancer Support Program at the North Carolina Cancer Hospital where he provides inpatient as well as outpatient assessment and therapeutic services for children and adolescents with cancer. He also works with children whose parents have serious medical illnesses such as cancer. His research is in the area of children's adaptation to cancer. Dr. Yopp came to UNC from St. Jude Children's Research Hospital in Memphis, TN, where he served as a clinical psychologist and coordinator of the inpatient consultation-liaison service. Prior to working at St. Jude, Dr. Yopp completed his internship and postdoctoral fellowship at Nationwide Children's Hospital in Columbus, Ohio. He earned his doctoral degree in clinical psychology from Central Michigan University and completed a pediatric psychology externship at Children's Hospital of Michigan. He is a native of Raleigh.

For our first live episode that we broadcasted during the APECS workshop "Antarctic Science: Global connections", we met with the Australian weather observer and Ph.D. candidate Vicki Heinrich. Well, actually, it wasn't only Vicki but we had another special guest: Vickie's pet cat sneaking around. For more than ten years, Vicki has been a regular in the Antarctic to look at the clouds and play with balloons, all courtesy of the Australian Bureau of Meteorology (BoM). To be trained on the ten types of clouds, she found this opportunity once in a newspaper, and it saved her from the mining industry. To get paid to stay twelve months at a weather station in Antarctica still feels extraordinary to her – as are the beauty of the Antarctic icescape and the many friendships she has built along the way, both with colleagues and penguin colonies. So why did Vicki then move into psychology? In 2019, she started her Ph.D. thesis with the project "Use of Weather and Climate Information: Risk perception and decision-making in the Antarctic" in order to learn more about how people use weather information to make their daily decisions. To get to know the other side – the user's perspective – is clearly an add-on to her career, and to us as her project got YOPP-endorsed. This time, we speak to Thomas Jung, head of the section Climate Dynamics at the German Alfred Wegener Institute and professor for Physics of the Climate System at the University of Bremen. ENCORE EPISODE - originally published January 16, 2019 I had such a great discussion with Dr. Justin Yopp of the University of North Carolina for this episode. Justin is a clinical psychologist and is co-author of the book, “The Group: Seven Widowed Fathers Reimagine Life.” I really enjoyed our wide-ranging discussion. It was a fascinating mix of practical and academic, and included some great stories from the widowed dads in the book about their experiences. Some of the topics we covered include: the dual process model of coping and bereavement; post-traumatic growth; the concept of being a ‘good enough' parent; the unique hardships of being a widowed father; and the research into the needs of widowed parents being conducted at UNC. By the way, if you're a widowed mom or dad with kids 18 & under at home, and widowed less than three years, I'd encourage you to answer the survey on their web site…this is such important work and, surprisingly, there is very little research that's been done on the needs of widowed parents. These are just some of the challenges widowed parents face. While grief professionals may offer helpful insights, our guest and psychologist Dr. Justin Yopp explains why peer support groups for widowed parents may be one of the most helpful resources for bereaved fathers and mothers. Justin is a clinical psychologist and is co-author of the book, “The Group: Seven Widowed Fathers Reimagine Life.” I really enjoyed our wide-ranging discussion. It was a fascinating mix of practical and academic, and included some great stories from the widowed dads in the book about their experiences. Justin Yopp, PhD is a clinical psychologist at the University of North Carolina and is a member of the UNC Comprehensive Cancer Support Program. His clinical practice focuses on providing therapeutic services to both pediatric and adult patients with or affected by cancer. In 2010, Dr. Yopp and his colleague, Dr. Don Rosenstein, developed a support program for widowed fathers who lost their wives to cancer. They currently run a support group for widowed fathers, are conducting research in the field, and have created an online resource of this population of widowers: www.singlefathersduetocancer.org The Single Fathers Due to Cancer Program at UNC is dedicated to helping the tens of thousands of fathers who have lost their wives/partners to cancer and who are figuring out how to be an “only” parent to a grieving child. Die Pathogenität von enteropathogenen Yersinien-Spezies wird von einem Virulenzplasmid pYV kodiert. Das pYV kodiert für den Typ-III-Sekretions-/Translokationsapparat und die Yops. Sechs Yop-Effektorproteine sind bekannt, die in die eukaryote Zelle transloziert werden: YopE, YopH, YopM,YopO/ YpkA, YopP/ YopJ und YopT. Die intrazellulären Angriffspunkte und Wirkungen der einzelnen Yops auf die Zielzelle sind Gegenstand aktueller Forschung. In dieser Arbeit wurden erstmals systematisch potentielle Zytoskelettveränderungen von Thrombozyten durch Inkubation mit Yersinia-Monosekretionsmutanten YopH, YopE, YopO, YopP, YopM und den Yersinia-Monodeletionsmutanten DeltaYopH, DeltaYopE, DeltaYopT, DeltaYopO und DeltaYopM untersucht. Im zeitlichen Verlauf adhärieren uninfizierte Thrombozyten durch eine Zunahme der Fläche durch Ausbildung von Filopodien und Lamellipodien. In den durchgeführten Experimenten ähneln die mit der YopP-Monosekretionsmutante inkubierten Thrombozyten uninfizierten Thrombozyten, d.h. YopP zeigt als Einziges der untersuchtes Yops keinen Einfluss auf das Zytoskelett. Mit der YopH, YopM, YopE bzw. YopO-Monosekretionsmutante inkubierte Thrombozyten zeigen kleinere Zellflächen und eine Dominanz von abgerundeten Zellen. Dies deutet auf eine schlechtere Adhäsion der Zellen an der extrazellulären Matrix hin. In mit WA-C(pTRANS,pCJYM-HA) infizierten HeLa-Zellen konnte YopM zunächst im Zytosol, nach 2-4h Infektion zusätzlich im Zellkern lokalisiert werden. Ausserdem zeigten mit WAC(pTRANS,pCJYM-HA) infizierte HeLa-Zellen Zytoskelettveränderungen: ein ungeordnetes Zytoskelett mit Vakuolen, vereinbar mit einer Ablösung der Zelle von der Matrix. Kaya et al. konnten im Affinitätsblot in vitro eine Interaktion von YopM mit der intrazellulären Cystein-Protease Calpain zeigen. Wegen der morphologischen Ähnlichkeit zwischen mit WA-C(pTRANS,pCJYM) inkubierten bzw. mit Calpeptin, einem Calpain-Inhibitor, inkubierten Thrombozyten wurde die von Kaya et al. dargestellte Interaktion zwischen YopM und Calpain in vivo in dieser Arbeit überprüft. Die Präzipitationsversuche aus infizierten HeLa-Zellen über Calpain sowie über YopM ergaben keinen Hinweis auf eine Interaktion von YopM und Calpain. McDonald et al. publizierten RSK 1 und PRK 2 als intrazelluläre Interaktionspartner von YopM. Funktionelle Studien über die Proteine RSK 1 und PRK 2 lassen darauf schließen, dass sie regulierende Einflüsse auf das Zytoskelett, die Translation und das Zellüberleben haben. Diese Funktionen und die beobachteten Einflüsse von YopM auf das Zytoskelett der Zielzelle sind mit einem Modell vereinbar, in dem YopM die Zytoskelettveränderungen durch Interaktion mit RSK 1 und PRK 2 hervorrufen könnte. Diese Hypothese muss durch zukünftige Forschungen geklärt werden.

Die Exposition von Makrophagen gegenüber Bakterien oder LPS führt über die Aktivierung zellulärer Signalkaskaden zu einer vermehrten Expression von Genen, deren Proteinprodukte zelluläres Überleben unter Infektionsbedingungen ermöglichen. Die Aktivierung des Transkriptionsfaktors NF - kB spielt dabei eine wichtige Rolle. Pathogene Yersinia enterocolitica - Stämme hemmen die Aktivierung von NF - kB und induzieren Apoptose bei Makrophagen. In dieser Arbeit konnte gezeigt werden, dass das Yersinia - Effektorprotein YopP maßgeblich an der Apoptoseinduktion beteiligt ist. Bei Infektionsversuchen konnten lediglich die Yersinienstämme bei Makrophagen Apoptose induzieren, die über einen funktionstüchtigen Typ III - Sekretionsapparat und ein dadurch transloziertes YopP verfügen. Eine transiente Überexpression der transkriptionell aktiven NF - kB - Untereinheit p65 schützt Makrophagen spezifisch vor durch Yersinien, nicht jedoch vor durch Salmonellen induzierter Apoptose. Das weist darauf hin, dass YopP durch die Blockierung des NF - kB aktivierenden Signalwegs Apoptose bei Makrophagen vermittelt. Die Transfektion von J774A.1 Makrophagen mit YopP induzierte bei 40 - 50% der transfizierten Zellen Apoptose. Durch die zusätzliche Stimulation mit LPS konnte die Apoptoserate auf 80 - 90% gesteigert werde. Dieser synergistische, proapoptotische Effekt ist direkt auf durch LPS induzierte Signaltransduktions -prozesse zurückzuführen. Aus Transfektionsversuchen mit dominant - negativen Signalmolekülen der TLR - Signalkaskade ergaben sich Hinweise auf eine Beteiligung der Transmitter MyD88 und IRAK2 an der Apoptoseeinleitung. IRAK1 und TRAF6 scheinen dagegen eher ein antagonistisches, NF - kB aktivierendes LPS - Signal zu bedienen, welches unter dem Einfluss von YopP unterdrückt wird. Dadurch überwiegt das durch LPS induzierte, proapoptotische Signal, welches den apoptotischen Zelltod einleitet. Die Aktivierung des Apoptoseprogramms selbst erfolgt über FADD und Caspase - 8.

Die Pathogenität von Y. enterocolitica O:8 ist gekoppelt an das Vorhandensein des 70 kb großen Virulenzplasmides (pYV), das für die Gene eines Typ-III-Sekretionssystems kodiert und die Translokation von 6 Yop Effektorproteinen (YopE, YopH, YopM, YopO, YopP und YopT) in das Zytosol eukaryontischer Zellen ermöglicht. Intrazellulär interagieren Yop Proteine dabei mit verschiedenen Zellstrukturen und Signaltransduktionskaskaden, wodurch es zu einer Modulation der Immunantwort im Wirtsorganismus kommt. Durch systematische Mutagenese der yop-Gene und Untersuchung der yop Deletionsmutanten im Mausinfektionsmodell konnte in dieser Arbeit ein Einfluss der Yop Proteine auf die Virulenz und die Auslösung einer Immunantwort nachgewiesen werden. Die Translokation eines heterologen Antigens (YopE/LLO) durch das Typ-III-Sekretionssystem führte zur Induktion einer spezifischen CD4 und CD8 T-Zellantwort gegen Listeriolysin O (LLO) und ermöglichte die Verwendung von Y. enterocolitica als oralen Lebendimpfstoff zur Immunisierung gegen eine Listeria monocytogenes Wildtyp Infektion.

Yersinien sind auf Grund ihres spezifischen Typ III-Proteinsekretionssystems (TTSS) in der Lage, nach Anlagerung an Zellen bakterielle Virulenzproteine in das Innere der Wirtszelle zu translozieren. Dies führt zu einer Zellparalyse und bei Makrophagen zum Zelltod. Eines der translozierten Virulenzproteine ist YopP/J, das die Signalwege der mitogenaktivierten Proteinkinasen (MAPK) sowie des Transkriptionsfaktors NF-kB in der Wirtszelle inhibiert. Es wird angenommen, dass YopP/J zur Familie der Cystein-Proteasen zählt. Es zeigt Homologien zu AvrRXV/AvrBst von Xanthomonas campestris, welches eine dem apoptotischen Zelltod vergleichbare sogenannte „Hypersensitive Reaktion (HR)“ bei Pflanzen auslösen kann. Die Bakterien der enteropathogenen Spezies Yersinia enterocolitica, welche nach oraler Aufnahme im terminalen Ileum durch M-Zellen in die Lymphfollikel der Peyerschen Plagues wandern, werden anhand ihrer Oberflächen-(O)- und Geißel-(H)-Antigene in verschiedene Serotypen unterteilt. Unsere Studien haben gezeigt, dass sich einzelne Y. enterocolitica-Serotypen in der Apoptoseinduktion bei Makrophagen unterscheiden. In der vorliegenden Arbeit wurde den Mechanismen der Apoptoseinduktion durch die verschiedenen Y. enterocolitica-Serotypen nachgegangen. Zunächst konnte YopP für die unterschiedlichen Apoptose-Effekte verantwortlich gemacht werden. Durch DNA-Austausch und der anschließenden Modifikation einzelner Aminosäuren zwischen YopP von Y. enterocolitica-Serotyp O8 und Y. enterocolitica-Serotyp O9 konnte Arginin (R) an Position 143 als essentielle AS der Effektordomäne von YopPO8 definiert werden. YopP von Y. enterocolitica-Serotyp O9 und Y. enterocolitica-Serotyp O3 besitzt an dieser AS-Position Serin (S). YopPO8 vermittelt deutlich stärker Apoptose als YopPO9 und YopPO3. Der erhöhten Apoptoseinduktion durch YopPO8 geht eine effiziente Unterdrückung der NF-kB-Aktivierung vorraus. Die Inhibition des antiapoptotischen NF-kB-Signalwegs durch YopP scheint Voraussetzung für die Apoptoseinduktion durch Yersinia zu sein. Des weiteren wurde in dieser Arbeit mit Hilfe des Hefe-2-Hybrid-Systems nach neuen unbekannten intrazellulären YopP-Interaktionspartner gesucht. Aus einer Maus-Milz-cDNA-Genbank konnten 22 potentielle YopP-Interaktionspartner isoliert werden. In in vitro-Bindungsstudien konnte bis zum jetzigen Zeitpunkt eine Interaktion von YopP mit vier Proteinen (PP2Ac, FWD2, eEf1bb, Smad1) bestätigt werden. Weiterführende Bindungs- und Funktionsanalysen werden noch durchgeführt. Die möglichen Auswirkungen dieser Interaktionen werden diskutiert. Die durchgeführten Studien erbrachten zudem Hinweise auf eine Modifikation von YopP in der Wirtszelle. Darüberhinaus konnte mit Hilfe des Hefe-2-Hybrid-Systems der Interaktionsbereich von YopP mit seinem Zielprotein IKKb eingeschränkt werden. Die durchgeführten Bindungsanalysen mit IKKb-Deletionsmutationen weisen auf eine Bindung von YopP an den IKKb-N-Terminus hin. Das Ergebniss wurde durch in vitro-Bindungsstudien bestätigt. Unter anderem scheint Alanin an IKKb-Position 120 die Interaktion mit YopP zu bestimmen. So konnten im Rahmen dieser Doktorarbeit unterschiedliche Aspekte der Wechselwirkung von Y. enterocolitica YopP mit der Wirtszelle und Wirtszellproteinen analysiert werden. Diese Studien tragen zum besseren Verständnis der Immunmodulation durch pathogene Bakterien bei.
