The acting is phenomenal between the two talented actresses who steal the screen, Sarah and Madi, who rightfully deserve awards for their roles. The movie is intense; too intense to the point where I couldn't take my eyes off the television. A., Placantonakis, D. Abstract: GPR133 (ADGRD1) is an adhesion G protein-coupled receptor that signals through Gas and is required for growth of glioblastoma (GBM), an aggressive brain malignancy. The regulation of GPR133 signaling is incompletely understood. Here, we use proximity biotinylation proteomics to identify ESYT1, a Ca2+-dependent mediator of endoplasmic reticulum-plasma membrane bridge formation, as an intracellular interactor of GPR133. ESYT1 knockdown or knockout increases GPR133 signaling, while its overexpression has the opposite effect, without altering GPR133 levels in the plasma membrane. The GPR133-ESYT1 interaction requires the Ca2+-sensing C2C domain of ESYT1. Thapsigargin-mediated increases in cytosolic Ca2+ relieve signaling-suppressive effects of ESYT1 by promoting ESYT1-GPR133 dissociation. ESYT1 knockdown or knockout in GBM impairs tumor growth in vitro, suggesting functions of ESYT1 beyond the interaction with GPR133. Our findings suggest a novel mechanism for modulation of GPR133 signaling by increased cytosolic Ca2+, which reduces the signaling-suppressive interaction between GPR133 and ESYT1 to raise cAMP levels. Barrett Bready is back on the program. He’s the CEO of Nabsys, a company with some new technology for genome mapping. Originally Nabsys had been working to develop nanopore sequencing, but after a recent reboot has become focused on scaling up scientists' ability to read structural genomic information. Barrett compares Nabsys’ new multiplex technology for genome mapping to the improvement of arrays over single nucleotide (SNP) detection.

Established in 2007, Tabacos Mata Fina launched an ambitious project aimed at reviving Brazil famous cigar industry and gaining recognition for the quality tobacco and cigars produced in Brazil. The factory is located in Cruz das Almas - Bahia in the middle of the Recôncavo Baiano area, a privileged region for the growth of the highest quality Brazilian Tobacco. The name Monte Pascoal means Easter Mount, this was the first piece of land sighted by Portugese explorers in 1500 and it was on Easter. Dark Side - SAN CRISTOBAL De La Habana LA FUERZA Although existing prior to the revolution, this is a new brand launched in 1999. The San Christobol brand that existed rught up to the revolution is not related to the modern brand. Named for the original name of Havana, this brand is still too young by some standards for a conclusion on its worthiness. The La Fuerza is a 50 rg by 5.5 Parejo. Interview - Wesley Gensel and Lorenzo from Monte Pascoal Herf Line from ER Doc - Panacea Green What else have you been smokin' Dale?- Esteban Carreras Habano - Esteban Carreras 187 - Punch Uppercut - La Escepcion - What else have you been smokin' Bob? - El Rico Habano - This is a harsh cigar with few redeeming factors. Although it has a lot of flavor, the harshness is overwhelming. Esencia Petite Corona - A Wonderfully full flavored cigar that continues to entertain well into the short nub. Introduced in 2008 this outstanding line of cigars is still at the top of my favorites list. Nestor Miranda Special Selection Breakfast Break - still a very good cigar, sometimes after trying a cigar and then not smoking one for several weeks, you find that it was not as good as you thought, this cigar held up well and is still a faborite. Casa Torano - Terrific mild smoke at an affordable price. What Else you been smoking Craig? Padilla Edicion Especial 2006 Obsidian Belicoso - Good typical Nicaraguan flavors with a strong core of pepper and prototypical Padilla spice. Mediocre burn, high strength & fairly heavy body. Bitter finish quick into the final 3rd. Padron Anniversario Coronas Natural 2005 - Bready sweetness with loads of caramel flavors. Matches the core of flavors expected from Padron but very unique to the wrapper leaf. Whats My Band - #926 suggested by Bruce Berg - Turns out to be a Padron Londres. Bob - Nice spice, very smooth and medium bodied. Excellent tastes of roasted something or other. The retrohale accentuated the spice only slightly and the savory flavor of roasted something provided a strong base for the cigar. The construction was excellent and the burn was perfect with an excellent draw. The cigar made me want to smoke it till I ate my fingers. I would rate this an excellent smoke and one I would like to smoke more of.
