In his experience, patients often don't understand the extensive nature of these surgeries prior to this visit, so he spends time explaining what a flap is and what the hospital course will look like. Then, Eli speaks about his experience with virtual surgical planning, which entails remote meetings with engineers. He admits that the learning curve to virtual surgical planning is steep, but that contemplating reconstruction with the help of virtual planning helps him better account for details at the time of surgery. Gopi and Ashley then inquire about surgery and peri-operative management of these patients. Eli addresses vital topics including surgical technique, fluid management, and thrombosis risk. Finally, the podcast addresses post-operative management of free flap patients. Eli briefly outlines his free flap protocol, emphasizing physical exam signs that indicate need for an OR takeback. In this Monterey Conversation, two leading scholars of the Soviet and post-Soviet space programs, Michael Gordin and Asif Siddiqi, addressed the effects of the USSR's disintegration on the Soviet space program; the ways in which this program was handled by the Russian Federation in the 1990s; and the place of the Russian space program in cooperation and competition with the United States. Doug Gordin is a long-time golf coach at both the Division I and Division II levels. He retired in 2018 after a long stint at Florida Southern where his teams captured six NCAA Division II National Championships. Abstract: Substance use disorder is a debilitating chronic disease and a leading cause of disability around the world. The nucleus accumbens (NAc) is a major brain hub that mediates reward behavior. Studies demonstrate exposure to cocaine is associated with molecular and functional imbalance in two NAc medium spiny neuron subtypes (MSNs), dopamine receptor 1 and 2 enriched D1-MSNs and D2-MSNs. Our previous reports showed that repeated cocaine exposure induced transcription factor early growth response 3 (Egr3) mRNA in NAc D1-MSNs, while reducing it in D2-MSNs. Here, we report our findings of repeated cocaine exposure inducing cell subtype specific bidirectional expression of the Egr3 corepressor NGFI-A-binding protein 2 (Nab2). Using CRISPR activation and interference (CRISPRa and CRISPRi) tools combined with Nab2 or Egr3 targeted sgRNAs, we mimicked these bidirectional changes in Neuro2a cells. Furthermore, we investigated D1-MSN and D2-MSN subtype specific expressional changes of histone lysine demethylases Kdm1a, Kdm6a and Kdm5c in NAc after repeated cocaine exposure. Since Kdm1a showed bidirectional expression patterns in D1-MSNs and D2-MSNs, like Egr3, we developed a light inducible Opto-CRISPR-KDM1a system. We were able to downregulate Egr3 and Nab2 transcripts and cause bidirectional expression changes in D1-MSNs and D2-MSNs similar to cocaine exposure in Neuro2A cells. In contrast, our Opto-CRISPR-p300 activation system induced the Egr3 and Nab2 transcripts and caused bidirectional transcription regulations in D1-MSNs and D2-MSNs. Our study sheds light on the expression patterns of Nab2 and Egr3 in specific NAc MSN subtypes in cocaine action and uses CRISPR tools to further mimic these expression patterns. Richard Gordin is an emeritus professor in sport psychology at Utah State University. He has been at the university for 37 years. Dr. Gordin has authored 100 articles and book chapters. He has been a sport psychology consultant for numerous athletic teams at his university, USA Gymnastics, USA Track and Field, US Ski and Snowboard Association and several professionals on the PGA and Champions Tours. He was the sport psychology consultant for the USA Women’s Gymnastics Team in Seoul in 1988 and a sport psychology consultant for the USA Track and Field Team for the 2004 Olympic Games in Athens. He recently served as the consultant for the USA Nordic Combined Ski Team in Vancouver in 2010 and in Sochi 2014. Host: Chris Mooney Before the "complementary and alternative medicine" fad, and before UFO craze, lived a man whom you might call the first modern pseudoscientist. His name was Immanuel Velikovsky. He had a strange theory about a comet—that turned out to be Venus—shaping the course of human history. He tangled with Carl Sagan about it—and with the scientific community about it. And then, he was mostly forgotten. But no longer, because Princeton historian of science Michael Gordin has tracked down Velikovsky's personal papers. In his book The Pseudoscience Wars: Immanuel Velikovsky and the Birth of the Modern Fringe, Gordin uses Velikovsky's example to show how he laid the groundwork for other pseudosciences-it's kind of like they followed in his footsteps. Michael Gordin is professor of history at Princeton University and director of the program in Russian and Eurasian Studies. He has written widely in the history of science with a focus on the Soviet Union and the early nuclear age. The Pseudoscience Wars is his fourth book.
