In this episode, we explore the intricate role of the vagus nerve as a central regulator within the microbiota-gut-brain (MGB) axis, examining its neuroanatomical structure, signaling mechanisms, and interactions with microbial metabolites and immune pathways. We discuss how vagal afferent fibers relay sensory input from the gut to the brain, including signals mediated by short-chain fatty acids (SCFAs) and gut-derived hormones, and how efferent fibers modulate gut motility, intestinal barrier integrity, and inflammation through the cholinergic anti-inflammatory pathway. Finally, we explore vagal dysfunction as well as associated conditions and symptoms, and we touch on just a few potential root causes. Topics: 1. Introduction Focus on the vagus nerve's role in the microbiota-gut-brain (MGB) axis. Bidirectional communication between the brain and microbiota. Overview of communication pathways: neural (e.g., vagus nerve), endocrine (e.g., HPA axis), immune (e.g., cytokines), and metabolic (e.g., SCFAs). 2. Interaction with Intestinal Cells Enteroendocrine cells (EECs) release gut hormones in response to microbial metabolites. SCFAs, such as butyrate, activate free fatty acid receptors on EECs, stimulating vagal afferents. Immune cells within the gut wall modulate vagal signals during inflammatory responses. 7. Review of Functions Sensory input (afferent fibers): Detects gut-derived signals like microbial metabolites and mechanical stretch. Motor output (efferent fibers): Regulates gut motility, secretion, immune responses, and more. 8. Impact of a Diverse Microbiome on Vagal Activity Enhanced SCFA production boosts vagal activity. SCFAs improve gut barrier integrity, reduce systemic inflammation, and assist in regulating stress responses. 9. Examples: Intestinal Barrier Function Releases acetylcholine (ACh) to modulate inflammatory pathways. Helps enhance tight junction protein expression, preserving gut barrier integrity. Helps prevent the translocation of microbial endotoxins like LPS into systemic circulation. 10. Dysfunction of the Vagus Nerve Reduced vagal tone disrupts gut homeostasis. Conditions such as IBS, IBD, chronic fatigue syndrome, anxiety, depression, and POTS. Chronic stress, infections, and dysbiosis are common contributors. 11. Root Causes 12. Tying Back to the HPA Axis Low vagal tone is associated with increased HPA axis activity. Highlighting the interplay between the gut, brain, and stress response systems. 13. How can we help people with prosthetic legs walk more easily and with less effort? In this episode of Neurocareers: Doing the Impossible!, we explore this question with Giacomo Valle, PhD, a leading researcher in the field of biomimetic brain-computer interfaces (BCIs) and bionic legs. Dr. Valle and his colleagues at ETH Zürich have developed an innovative biomimetic BCI that uses peripheral nerve stimulation to convey naturalistic touch sensations in bionic legs. This groundbreaking work has earned them a spot as one of the 12 finalists for the BCI Award 2023. Dr. Valle's research focuses on creating artificial communication with the brain through peripheral nerve stimulation, providing a more intuitive and natural sensory experience. His team developed a biomimetic neurostimulation framework inspired by nature, capable of "writing" physiologically plausible information back into the peripheral nervous system. Using an in-silico model of mechanoreceptors, they created biomimetic stimulation policies, which were tested alongside mechanical touch and common linear neuromodulations. In clinical trials, these techniques have shown significant improvements in mobility and reduced mental effort compared to traditional approaches. The neural responses produced by this innovative approach resemble those naturally induced, marking a significant advancement in assistive neurotechnologies. Dr. Valle's project, "Biomimetic BCI Conveys Naturalistic Touch Sensations via Peripheral Nerve Stimulation for Bionic Legs," represents a major leap forward in neuroengineering and bionics. With an educational background that includes a B.S. in Biomedical Engineering and an M.S. in Bioengineering from the University of Genoa, and a Ph.D. in Biorobotics from the Scuola Superiore Sant'Anna, Dr. Valle has extensive experience in this field. His career journey includes research positions at the École Polytechnique Fédérale de Lausanne and ETH Zurich, where he contributed to significant innovations in neuroprosthetics. As a co-founder of the medtech startup MYNERVA and now a Tenure-track Assistant Professor in Bionics at Chalmers University of Technology in Sweden, Dr. Valle's research focuses on neuroengineering and computational models aimed at restoring sensory feedback and understanding somatosensory processing and neural control of movements. Australia's Energy Market Operator predicts that in the future over 30 per cent of Australia's electricity generation and flexible demand services will come from VPPs. And many energy retailers are literally banking on consumers signing up to one. VPPs are seen as one way to coordinate and manage all the numerous small-scale, decentralized energy resources like solar panels, EVs, and home batteries. But not everyone thinks that's going to happen. Tim Washington is the co-founder and CEO of one of Australia's largest electric vehicle charging services, Jetcharge. And he doesn't think many Australians will need to join a VPP when they have an electric vehicle with bidirectional charging that will not only be able to charge from their solar, it will send energy back to the grid or into our homes. This week's EYE ON NPI will galvanize your next design, with robust galvanic isolation specifically for I2C devices: we're highlighting the Analog Devices ADuM1252/3 Bidirectional I²C Isolators (https://www.digikey.com/en/product-highlight/a/analog-devices/adum1252-bidirectional-i2c-isolator). They are available in two flavors: as the unidirectional ADuM1253 (https://www.digikey.com/short/0w8fq3w5) and bi-directional ADuM1252 (https://www.digikey.com/short/r4j3tcrf). These little SOIC chips provide trustworthy isolation up to a shocking ±10 kV surge, with pending safety and regulatory approvals, so you can feel confident that your design is created as safe as possible. Digital isolation (https://www.analog.com/en/product-category/isolation.html) is essential when dealing with medical, industrial or robotic designs. For industrial/robotic applications, the motors are very noisy and you want to make sure that there is no direct path for that electrical noise to your delicate sensors or microcontrollers. With medical cases, engineers are often tasked with measuring very fine sensors, with uV signals, and providing repeatable data that is used for life-or-death decisions, while also making sure that the 'DUT' is never exposed to dangerous voltages all in an environment with tons of other equipment, some of which may be misbehaving...that's a lot of pressure! The ADuM1252/3 is specifically designed for isolation of I2C signals: you can't just use 'any' isolator because I2C is a bi-directional data protocol that uses pull-ups on open-drain data and clock lines to communicate from controller and peripheral. The ADuM1252 (https://www.digikey.com/short/r4j3tcrf) has bi-directionality on both SDA and SCL, useful for 'multi-controller' modes or when clock stretching is used. The ADuM1253 (https://www.digikey.com/short/0w8fq3w5) has unidirectional SCL for most common I2C devices, and is a little less expensive. The ADI iCoupler technology uses 'galvanic' isolation, that's not the same as optoisolation! According to ADI, you'll get much better data-rate performance and isolation with galvanic compared to optoisolation. Also, lower power since you don't have to turn on/off an LED. Plus you can also use iCoupler for power transfer, which optoisolation cannot do. (https://www.analog.com/media/en/technical-documentation/frequently-asked-questions/icoupler_faq.pdf) The way iCoupler works is by creating a tiny transformer (https://en.wikipedia.org/wiki/Transformer) with two sets of metal coils, separated by an isolating polyamide layer. For data transfer, little coils are used and for power transfer, bigger coils can handle a couple mA! Of course, if we're using a transformer, you know that DC signals cannot be passed through, only AC; and data has a DC component plus isn't a sine wave. That means that there's also a data-to-AC codec on board that will turn I2C signals to AC, then back to I2C. All this is managed transparently so you don't have to worry about it, just treat it as a single I2C bus. There's also a couple nice extras that are helpful if you're trying to make I2C more fault-tolerant. Pre-charge circuitry and 'stuck' detection allow hot-swapping, either purposeful or accidental. The two isolated sides are not connected together until the Side2 half is in a neutral-bus condition. Since each side is separate, you can also use the ADuM1252/3 (https://www.digikey.com/short/1vbn4hcb) for level shifting: either half can be between 1.7 and 5.5V power and logic. With the fast iCoupler codec, you can get up to 2 MHz clock rate for fast I2C data transfer. If you need iCouplers for other protocols like USB or SPI - Analog devices has those available as well, each version is tuned for the usage so you get the best performance. If you need high performance galvanic isolation for your next I2C design, the Analog Devices ADuM1252/3 Bidirectional I²C Isolators (https://www.digikey.com/en/product-highlight/a/analog-devices/adum1252-bidirectional-i2c-isolator) will do an excellent job at creating a code-transparent galvanic barrier for up to 2 MHz bidirectional I2C communication. Renewable Energy is The FutureCalifornia is no stranger to power outages. In 2019, for example, over 25,000 blackout events were recorded across the state, leaving homeowners and businesses without electricity. In recent years, most blackouts are the result of wildfire, wildfire risk (leading to utility shutdowns), and extreme heat (leading to high electricity usage). When the electricity grid is stressed,  California relies primarily on gas-powered peaker plants and diesel generators to keep electricity running. However, the use of peaker plants and diesel generators as the primary backup source is not only costly, but can accelerate the climate crisis through the release of greenhouse gases. Instead of solely relying on fossil fuel-powered plants, the state is beginning to transition to the use of clean energy sources like wind and solar in addition to incorporating the use of more batteries and other energy storage to make the renewable energy transition a reality. One part of the solution is the use of electric vehicle batteries as a power source for homes and businesses.  How Does Bidirectional Charging Work?Bidirectional charging, which allows the energy stored in a car battery to be sent to various recipients, is key to this transition as it can power one's home, business, appliance, or alternate vehicle. Also known as vehicle-to-home or two-way charging, this innovation additionally serves as an energy backup during power outages.Senate Bill 233, authored by state Senator Nancy Skinner, would require that all new manufactured electric vehicles sold in California have bidirectional charging abilities by 2035. Currently, electric and hybrid vehicles account for one in four new car sales in California. By implementing bidirectional charging now, the majority of electric vehicles purchased would thus be able to serve as a backup power source during extreme weather events in the near future.Energy Wherever, WheneverBidirectional charging has many potential benefits. First, smart charging technology allows a car battery to be charged during off-peak hours, providing a potential financial advantage for users. If owners charge their EVs during off hours, and discharge back to the grid during peak hours, they can earn the difference between the two rates. With vehicle-to-grid technology, homeowners can sell energy back to the utility company for redistribution which can be used to power homes, buildings or other EVs.Second, bidirectional charging serves as a backup power source during outages, with a typical car battery storing enough power for a home for roughly two days. As the number of extreme weather events like wildfires and hurricanes increase in frequency, dual charging through EVs can become a dependable source of power. Further, bidirectional technology can serve as a portable power source, providing energy while on the road. What are the main challenges?As bidirectional EV charging technology is still coming to fruition, it is not yet widely available, although the lowest cost EV on the market, the Nissan Leaf, has been bidirectional for a decade.  Tesla has said that its vehicles would be bidirectional by model year 2025, General Motors has promised its EVs would be bidirectional by model year 2026. As noted in the Kia/Hyundai advertisement which ran during the 2024 Super Bowl, “vehicle-to-load” is available now and can be used to power a refrigerator or other load during a power outage.  Fully utilizing the benefits of integrated “vehicle-to-home” as has been advertised by Ford requires additional costs to upgrade home wiring and may cost more than low-income consumers can afford so high investment requirements may discourage the use of such technology amongst lower income groups. Who is Ellie Cohen?Ellie Cohen, CEO of the Climate Center, is a leader in transformative solutions to climate change and environmental degradation. Vision Mamba: Efficient Visual Representation Learning with Bidirectional State Space Model

Recently the state space models (SSMs) with efficient hardware-aware designs, i.e., Mamba, have shown great potential for long sequence modeling. Building efficient and generic vision backbones purely upon SSMs is an appealing direction. However, representing visual data is challenging for SSMs due to the position-sensitivity of visual data and the requirement of global context for visual understanding. In this paper, we show that the reliance of visual representation learning on self-attention is not necessary and propose a new generic vision backbone with bidirectional Mamba blocks (Vim), which marks the image sequences with position embeddings and compresses the visual representation with bidirectional state space models. On ImageNet classification, COCO object detection, and ADE20k semantic segmentation tasks, Vim achieves higher performance compared to well-established vision transformers like DeiT, while also demonstrating significantly improved computation&memory efficiency. For example, Vim is 2.8$times$ faster than DeiT and saves 86.8% GPU memory when performing batch inference to extract features on images with a resolution of 1248$times$1248. The results demonstrate that Vim is capable of overcoming the computation&memory constraints on performing Transformer-style understanding for high-resolution images and it has great potential to become the next-generation backbone for vision foundation models. We're thrilled to share the Library of Economic Possibility (LEP), a new kind of knowledge-base for discovering, organizing, and sharing economic research around high-impact policies that have remained outside the mainstream despite significant research. Options for discovering economics research today — especially for non-specialists — are clunky. Activists tend to cherry-pick, journalists don't have the room to present a wide range of evidence in a single article, academics share their work through papers and conferences that general audiences generally don't engage with, think tanks wind up burying research beneath article archives. Search functions on major databases aren't the greatest. This is consequential in a moment where interest is rising around new economic ideas — LEP hopes to ground this rising interest in the wealth of existing evidence, and build a bridge between general audiences and economics research. We're also trying out a new way of organizing and connecting information. We're passionate about debating what the next economic system might look like, but we're also nerds about information architecture, and LEP's search features reflect that. Bidirectional links create associative trails between the network of information, and advanced search filters let you mix & match policies with specific areas of interest to hone in on precise relationships between information. So if you're curious to learn more about how basic income might affect entrepreneurship, you can select the policy "basic income" and the tag "entrepreneurship," and scroll through all our insights and sources that relate to both of those filters. Or, you could select "land value tax" and "urban development," or "codetermination" and "innovation." You get the idea. You can find those filters on the left column: Our policy reports also use a nifty little feature we call "insight cards." Nancy Davidson, M.D., serves as the Executive Vice President for Clinical Affairs and Senior Vice President and Professor of the Clinical Research Division at the Fred Hutchinson Cancer Center at the University of Washington, where she heads the Division of Medical Oncology. She is the past President of both the American Society of Clinical Oncology (ASCO) and the American Association for Cancer Research (AACR). She is a recipient of a number of prestigious awards including the ASCO Gianni Bonadonna Breast Cancer Award and the NCI Rosalind E. Franklin Award and was recently inducted into the Giants of Cancer Care. “I don't think I can ever be a professor at Johns Hopkins,” said Dr. Davidson when she was a junior faculty to her mentor Dr. Martin Abeloff. Sono Motors might be on to something. The idea is that if a solar-powered electric vehicle can recharge its battery via the sun equal to a driver's average commute time, the driver will never have to worry about range anxiety creeping into their consciousness ever again. And that's the goal of the Sono Sion, the company's solar-powered consumer vehicle with production beginning in 2023."The purpose of this car is to cover the daily average distance driven by a commuter because it's actually not a lot. I believe in the U.S. it's about 24 km (approx. 15 miles) and in Europe, it's 16 km (approx. 10 miles) per day, and that's actually what we recharge by the sun," says Laurin Hahn, co-founder and CEO of Sono Motors. "It's four times more range on a single charge than with any other electric vehicle without solar with the same battery change."It sounds like a good idea, but does it actually work in practice?To find out the nitty-gritty details, we brought Hahn onto the show, who provided no shortage of insight into how this technology works, the benefits for drivers, and even where the potential lies for combining solar with other technologies to create what is essentially a sustainable power plant on wheels.

The relationship between our diet and sleep is bi-directional; i.e. sleep impacts diet and diet impacts sleep. Therefore, we can examine the impact of sleep timing, duration and other dimensions on our dietary intake. And then also examine the impat of both overall diet and specific nutrients on improving/worsening sleep. The is clear evidence of distinct, acute effects of restricted sleep time on food preferences, eating behaviour, energy intake, and our underlying metabolic physiology. When it comes to the ability of certain foods or nutrients to improve sleep, often many claims are based on weak evidence or mechanistic reasoning. But there is evidence showing some impacts of certain compounds to either positively or negatively impact sleep. So what is the accurate way to look at this bi-directional relationship? In this episode, Greg Potter, PhD discusses the evidence to date. Bidirectional charging capabilities offer a higher level of electric resiliency and less reliability on the grid, especially during peak times. - The pilot includes the use of bidirectional hardware that's paired with software-defined communications protocols that will enable power to flow from a charged EV battery into a customer's home. The software will automatically coordinate between the EV, home and PG&E's electric supply from the street. - The pilot will test bidirectional power technology with multiple GM EVs. - Last month, Ford announced a partnership with San Francisco-based Sunrun, a provider of residential solar panels and home batteries that allows Ford F-150 Lightning owners to use their truck's substantial battery reserves as a home's emergency power source. - To use Ford's Intelligent Backup Power, customers will need to install the Ford Charge Station Pro and Home Integration System, which includes a power inverter plus a dark start battery and transfer switch, enabling two-way power flow between the home and vehicle. Surprisingly, German officials said at the time that they expected Tesla to fulfill all those conditions within the next two weeks. - March 22 will mark an important step for Tesla by starting deliveries of European-made vehicles and putting it on a path of better logistical efficiency. This would allow the public water supply to be maintained, the Ministry of the Environment told the DPA. - On Friday evening, the Administrative Court of Frankfurt (Oder) had declared the water permit issued in 2020 “unlawful and unenforceable”. - In a first reaction, the Straußberg-Erkner Water Association (WSE) had demanded that the state tolerate the water pumping until the procedure was made up for. - However, the 1.4 million cubic metres of water actually contractually guaranteed by the WSE for Tesla would only be needed when the carmaker in Grünheide has actually reached its full capacity of 500,000 cars per year. The situation is worst in Europe, where the customers rely on Giga Shanghai production allocation. In the case of Model 3 RWD and LR AWD, the estimated delivery time to Germany has jumped to February 2023. This is just shy of a full year away. - This is true for the customers in Germany, France, and United Kingdom. The waiting time is significantly longer than what people in the North American market are facing. For American customers, the same vehicles are promised to be delivered in May 2022, while the Canadians have to wait until August 2022. This time, the Ukraine war is cited as the root cause for production pauses. - The two premium German brands are joining BMW, Mini, and Volkswagen, all of which have been forced to idle plants across Europe. At the moment, March 18th is the project date for a return to normal activities however that could change at any moment. - One of the main missing parts of wiring harnesses which were made in Ukraine. Part production was halted once Russia invaded the country according to Automotive News Europe. - Mercedes-Benz has halted S-Class and EQS production at its Sindelfingen plant near Stuttgart. Benz's neighbours, Porsche, deferred Taycan output at its Stuttgart-Zuffenhausen factory. - Porsche specified that about 200 Taycans will not be built. Over at Mercedes, no such detail has been disclosed.
