Abstract: Neurosurgical procedures, in which electrodes can be placed in the brain of awake patients, offer remarkable opportunities to discover the neurophysiology underlying human speech. The relative scarcity of these opportunities and the altruism of participating patients obligates us to apply the highest possible rigor to signal interpretation. Intracranial electroencephalography (iEEG) signals recorded during overt speech can present an acoustic-induced vibration artifact that tracks the fundamental frequency (F0) of the participant's voice, encompassing high-gamma frequencies that are used for neural activation during speech production and perception. To advance our understanding of the neural control of speech production and develop reliable speech models, we developed a spatial filtering approach to identify and remove acoustic-induced artifactual components of the recorded signal. We show that traditional reference schemes may jeopardize signal quality, but our data-driven method can denoise the recording while preserving signals from the underlying neural activity. Multisite Thalamic Recordings to Characterize Seizure Propagation in the Human Brain

Abstract: Neuromodulation of the anterior nuclei of the thalamus (ANT) has shown to be efficacious in patients with refractory focal epilepsy, but it is not uniformly effective. One important uncertainty is to what extent thalamic subregions other than the ANT are recruited earlier and more prominently in the propagation of seizures in patients with presumed temporal lobe epilepsy (TLE). To address this unknown, we studied 11 patients with clinical manifestations of TLE planned to undergo invasive stereo-encephalography (sEEG) monitoring. We extended cortical electrodes to reach thalamic nuclear subdivisions in the anterior (ANT), middle (mediodorsal) and or posterior (pulvinar) sites. This multisite thalamic sampling was without any adverse events. Intracranial EEG (iEEG) recordings confirmed seizure onset in medial temporal lobe, insula, orbitofrontal and temporal neocortical sites, thereby highlighting the importance of iEEG for more accurate localization of seizure foci. Visual review of EEGs documented early and prominent involvement of specific thalamic sites. Seizures originating from the same brain origin produced a stereotyped thalamic EEG signature. Visual review of EEGs, validated with single-pulse corticothalamic evoked potentials, documented early and prominent involvement of thalamic sites that would have not been predicted given the anatomy of seizure onset zones. Pulvinar was involved earlier and more prominently than other sampled nuclear subgroups in 60% of patients, even though all patients had a presumed diagnosis of TLE prior to invasive monitoring. Our findings document the feasibility and safety of multisite sampling from the human thalamus and suggest that the anatomy of thalamic involvement may not be entirely predictable on the basis of clinical information or lobar localization of seizures. Future clinical trials can establish whether offering more personalized targets for thalamic neuromodulation will lead to greater meaningful improvements in outcome. I., Van Veen, B. Abstract: Multivariate autoregressive (MVAR) model estimation enables assessment of causal interactions in brain networks. However, accurately estimating MVAR models for high-dimensional electrophysiological recordings is challenging due to the extensive data requirements. Hence, the applicability of MVAR models for study of brain behavior over hundreds of recording sites has been very limited. Prior work has focused on different strategies for selecting a subset of important MVAR coefficients in the model and is motivated by the potential of MVAR models and the data requirements of conventional least-squares estimation algorithms. Here we propose incorporating prior information, such as fMRI, into MVAR model estimation using a weighted group LASSO regularization strategy. The proposed approach is shown to reduce data requirements by a factor of two relative to the recently proposed group LASSO method of Endemann et al. (2022) while resulting in models that are both more parsimonious and have higher fidelity to the ground truth. The effectiveness of the method is demonstrated using simulation studies of physiologically realistic MVAR models derived from iEEG data. The robustness of the approach to deviations between the conditions under which the prior information and iEEG data is obtained is illustrated using models from data collected in different sleep stages. This approach will allow accurate effective connectivity analyses over short time scales, facilitating investigations of causal interactions in the brain underlying perception and cognition during rapid transitions in behavioral state. Abstract: Intracranial EEG (iEEG) data is a powerful way to map brain function, characterized by high temporal and spatial resolution, allowing the study of interactions among neuronal populations that orchestrate cognitive processing. However, the statistical inference and analysis of brain networks using iEEG data faces many challenges related to its sparse brain coverage, and its inhomogeneity across patients. We review these challenges and develop a methodological pipeline for estimation of network structure not obtainable from any single patient, illustrated on the inference of the interaction among visual streams using a dataset of 27 human iEEG recordings from a visual experiment employing visual scene stimuli. 100 ms sliding window and multiple band-pass filtered signals are used to provide temporal and spectral resolution. For the connectivity analysis we showcase two connectivity measures reflecting different types of interaction between regions of interest (ROI): Phase Locking Value as a symmetric measure of synchrony, and Directed Transfer Function - asymmetric measure describing causal interaction. For each two channels, initial uncorrected significance testing at p less than 0.05 for every time-frequency point is carried out by comparison of the data-derived connectivity to a baseline surrogate-based null distribution, providing a binary time-frequency connectivity map. For each ROI pair, a connectivity density map is obtained by averaging across all pairs of channels spanning them, effectively agglomerating data across relevant channels and subjects. Finally, the difference of the mean map value after and before the stimulation is compared to the same statistic in surrogate data to assess link significance. The analysis confirmed the function of the parieto-medial temporal pathway, mediating visuospatial information between dorsal and ventral visual streams during visual scene analysis. Moreover, we observed the anterior hippocampal connectivity with more posterior areas in the medial temporal lobe, and found the reciprocal information flow between early processing areas and medial place area. To summarize, we developed an approach for estimating network connectivity, dealing with the challenge of sparse individual coverage of intracranial EEG electrodes. Its application provided new insights into the interaction between the dorsal and ventral visual streams, one of the iconic dualities in human cognition. Copy rights belong to original authors. Abstract: Background: Magnetoencephalography (MEG) is a widely used non-invasive tool to estimate brain activity with high temporal resolution. However, due to the ill-posed nature of the MEG source imaging (MSI) problem, the ability of MSI to identify accurately underlying brain sources along the cortical surface is still uncertain and requires validation. Method: We validated the ability of MSI to estimate the background resting state activity of 45 healthy participants by comparing it to the intracranial EEG (IEEG) atlas (https://mni-open-ieegatlas.research.mcgill.ca/). First, we applied wavelet-based Maximum Entropy on the Mean (wMEM) as an MSI technique. Next, we converted MEG source maps into intracranial space, by applying a forward model to the MEG reconstructed source maps and estimated virtual IEEG (VIEEG) potentials on each IEEG channel location and quantitatively compared those with actual IEEG signals from the atlas for 38 regions of interest in the canonical frequency bands. Results: The MEG spectra were more accurately estimated in the lateral regions compared to the medial regions. The regions with higher amplitude in the VIEEG than in the IEEG were more accurately recovered. In the deep regions, MEG estimated amplitudes were largely underestimated and the spectra were poorly recovered. Moreover, the MEG largely overestimated oscillatory peaks in the alpha band, especially in the anterior and deep regions. This is possibly due to higher phase synchronization of alpha oscillations over extended regions, exceeding the spatial sensitivity of IEEG but detected by MEG. Importantly, we found that MEG estimated spectra were more comparable to spectra from the IEEG atlas after the aperiodic components were removed. Conclusion: This study identifies brain regions and frequencies for which MEG source analysis is likely to be reliable, a promising step towards resolving the uncertainty in recovering intracerebral activity from non-invasive MEG studies. Abstract: Working memory (WM) is the ability to actively maintain information for a short time and is central to human behavior. Rodent studies have proposed that hippocampal-entorhinal communication supports WM maintenance. However, the exact neural mechanisms of this interaction in WM remains unclear in humans. To address these questions, we combined machine learning analyses with intracranial electroencephalography (iEEG) recordings from the hippocampus and the entorhinal cortex (EC) in human participants, who maintained a set of letters in their WM. We found that WM maintenance was accompanied by elevated bidirectional hippocampal-EC information exchange via the theta band (2-8 Hz) and bidirectional cross-region theta-gamma phase-amplitude coupling (PAC). Further decoding analyses showed that the unidirectional inter-regional communication, with both theta oscillations in the hippocampus modulating EC gamma activity and theta band-coordinated information flow from the hippocampus, could decode correct performance at the level of participants. Taken together, our results demonstrate that theta functional coupling in the hippocampal-EC supports the maintenance of WM information via a specific pattern of frequency and direction. Die Form von Kirche muss sich immer wieder an die Gegebenheiten der Zeit anpassen und hat sich auch immer wieder an die Gegebenheiten der Zeit angepasst. Das ist eine Binsenweisheit. Die viel schwierige Frage ist dabei: Wie ist die Form für Kirche in dieser Zeit? Über diese Frage rede ich mit Hans Hermann Pompe, Referent für Mission und Kirchenentwicklung der Arbeitsstelle für missionarische Kirchenentwicklung und diakonische Profilbildung (midi) und Michael Herbst, Professor an der Universität Greiwswald und Leiter des Instituts zur Erforschung von Evangelisation und Gemeindeentwicklung (IEEG). Wir sind als einzelne Ortsgemeinde häufig völlig überfordert vorzuhalten, was alles zum Vollprogramm einer Kirchengemeinde gehört. Abstract: The sleep-wake cycle is constituted by three behavioral states: wakefulness (W), non-REM (NREM) and REM sleep. These states are associated with drastic changes in cognitive capacities, mostly determined by the function of the thalamo-cortical system. Thalamo-cortical activity can be examined by means of the intra-cranial electroencephalogram (iEEG). With the purpose to study in depth the basal activity of the iEEG in adult rats, we analyzed the spectral power and coherence of the iEEG during W and sleep in the paleocortex (olfactory bulb), as well as in motor, somatosensory and visual neocortical areas. We also analyzed the laterality (right Vs. left hemispheres) of the signals, as well as the iEEG in function of the light and dark phases. We found that the iEEG power and coherence of the whole spectrum were largely affected by behavioral states and were highly dependent on the cortical areas recorded. We also determined that there are night/day differences in power and coherence during sleep, but not in W. Finally, while we did not find right/left differences in power either in W or sleep, we observed that during REM sleep intra-hemispheric coherence differs between both hemispheres. We conclude that the iEEG dynamics is highly dependent on the cortical area and behavioral states. We also determine that there are light/dark phases disparities in the iEEG that emerge during sleep, and that intra-hemispheric connectivity differs between both hemispheres only during REM sleep. Abstract: Investigations using noninvasive functional magnetic resonance imaging (fMRI) have provided significant insights into the unique functional organization and profound importance of the human default mode network (DMN), yet these methods are limited in their ability to resolve network dynamics across multiple timescales. Electrophysiological techniques are critical to address these challenges, yet few studies have explored the neurophysiological underpinnings of the DMN. Here we investigate the brain-wide electrophysiological organization of the DMN in a common large-scale network framework consistent with prior fMRI studies. We used brain-wide intracranial EEG (iEEG) recordings, and evaluated intra- and cross-network interactions during the resting-state and cognition. Our analysis revealed significantly greater intra-DMN phase iEEG synchronization in the slow-wave (< 4 Hz) while DMN interactions with other brain networks was higher in all higher frequencies. Crucially, slow-wave intra-DMN synchronization was observed in the task-free resting-state and during verbal memory encoding and recall. Compared to resting-state, intra-DMN phase synchronization was significantly higher during both memory encoding and recall. Slow-wave intra-DMN phase synchronization increased during successful memory retrieval, highlighting its behavioral relevance. Finally, analysis of nonlinear dynamic causal interactions revealed that the DMN is a causal outflow network during both memory encoding and recall. Our findings identify dynamic spectro-temporal network features that allow the DMN to maintain a balance between stability and flexibility, intrinsically and during task-based cognition, provide novel insights into the neurophysiological foundations of the human DMN, and elucidate network mechanisms by which it supports cognition. Abstract: We describe the spatiotemporal course of cortical high-gamma activity (HGA), hippocampal ripple activity and interictal epileptiform discharges (IEDs) during an associative memory task in 15 epilepsy patients undergoing invasive electroencephalography (iEEG). Successful encoding trials were significantly greater HGA in hippocampus and frontal regions. Successful recall trials manifested sustained HGA in occipital pole and hippocampus compared to failed responses. Hippocampal ripple rates were greater during successful encoding and retrieval trials. IEDs during encoding were associated with decreased odds of remembering in hippocampus (-19%), temporal pole (-18%) or middle temporal gyrus (-15%). Hippocampal IEDs during retrieval predicted 33% decreased odds of remembering. Odds of remembering were further reduced if IEDs occurred during the 500-2000 ms window of encoding or retrieval. Hippocampal IEDs were followed by a transient decrease in ripple rate. We hypothesize that IEDs impair associative memory in a regionally and temporally specific manner by decreasing physiologic hippocampal ripples necessary for effective encoding and recall. Because dynamic memory impairment arises from pathological IED events competing with physiological ripples, IEDs represent a promising therapeutic target for memory remediation in patients with epilepsy. Abstract: Objective: Studies of high frequency oscillations (HFOs) in epilepsy have primarily tested the HFO rate as a biomarker of the seizure onset zone (SOZ), but the rate varies over time and is not robust for all individual subjects. As an alternative, we tested the performance of HFO amplitude as a potential SOZ biomarker using two automated detection algorithms. Methods: HFOs were detected in intracranial electroencephalogram (iEEG) from 11 patients using a machine learning algorithm and a standard amplitude-based algorithm. For each detector, SOZ and non-SOZ channels were classified using the rate and amplitude of high frequency events, and performance was compared using receiver operating characteristic curves. Results: The amplitude of detected events was significantly higher in SOZ. Across subjects, amplitude more accurately classified SOZ/non-SOZ than rate (higher values of area under the ROC curve and sensitivity, and lower false positive rates). Moreover, amplitude was more consistent across segments of data, indicated by lower coefficient of variation. Conclusion: As an SOZ biomarker, HFO amplitude offers advantages over HFO rate: it exhibits higher classification accuracy, more consistency over time, and robustness to parameter changes. Significance: This biomarker has the potential to increase the generalizability of HFOs and facilitate clinical implementation as a tool for SOZ localization. Copy rights belong to original authors. Abstract: Direct recording of neural activity from the human brain using implanted electrodes (iEEG, intracranial electroencephalography) is a fast-growing technique in human neuroscience. While the ability to record from the human brain with high spatial and temporal resolution has advanced our understanding, it generates staggering amounts of data: a single patient can be implanted with hundreds of electrodes, each sampled thousands of times a second for hours or days. The difficulty of exploring these vast datasets is the rate-limiting step in discovery. To overcome this obstacle, we created RAVE ("R Analysis and Visualization of iEEG"). All components of RAVE, including the underlying "R" language, are free and open source. User interactions occur through a web browser, making it transparent to the user whether the back-end data storage and computation is occurring on a local machine, a lab server, or in the cloud. Without writing a single line of computer code, users can create custom analyses, apply them to data from hundreds of iEEG electrodes, and instantly visualize the results on cortical surface models. Multiple types of plots are used to display analysis results, each of which can be downloaded as publication-ready graphics with a single click. Abstract: Human data collected using noninvasive imaging techniques have established the importance of parietal regions towards episodic memory retrieval, including the angular gyrus and posterior cingulate cortex. Such regions comprise part of a putative core episodic retrieval network. In free recall, comparisons between contextually appropriate and inappropriate recall events (i.e. prior list intrusions) provide the opportunity to study memory retrieval networks supporting veridical recall, and existing findings predict that differences in electrical activity in these brain regions should be identified according to the accuracy of recall. However, prior iEEG studies, utilizing principally subdural grid electrodes, have not fully characterized brain activity in parietal regions during memory retrieval and have not examined connectivity between core recollection areas and the hippocampus or prefrontal cortex. Here, we employed a data set obtained from 100 human patients implanted with stereo EEG electrodes for seizure mapping purposes as they performed a free recall task. This data set allowed us to separately analyze activity in midline versus lateral parietal brain regions, and in anterior versus posterior hippocampus, to identify areas in which retrieval-related activity predicted the recollection of a correct versus an incorrect memory. With the wide coverage afforded by the stereo EEG approach, we were also able to examine interregional connectivity. Our key findings were that differences in gamma band activity in the angular gyrus, precuneus, posterior temporal cortex, and posterior (more than anterior) hippocampus discriminated accurate versus inaccurate recall as well as active retrieval versus memory search. The left angular gyrus exhibited a significant power decrease preceding list intrusions as well as unique phase-amplitude coupling properties, whereas the prefrontal cortex was unique in exhibiting a power increase during list intrusions. Analysis of connectivity revealed significant hemispheric asymmetry, with relatively sparse left- sided functional connections compared to the right hemisphere. One exception to this finding was elevated connectivity between the prefrontal cortex and left angular gyrus. This finding is interpreted as evidence for the engagement of prefrontal cortex in memory monitoring and mnemonic decision-making. Copy rights belong to original authors. En diciembre de 2020, afirmó López Obrador que el Sistema Nacional de Salud funcionará a toda máquina, con atención médica y medicamentos gratuitos. No va a salir gratis, de algún lado tienen que financiarse, solo hay dos caminos: endeudarse o mejorar la recaudación. En otro tema quiénes son los expertos en materia electoral para continuar el impulso del IEEG. Sobre todo algunas mujeres, por lo que es necesario ver si el próximo presidente del instituto será mujer, pero no solo por el simple hecho de serlo, sino por sus méritos.

Izzy Lugo is COO at Urban Misfit Ventures, an 8-month-old, start-up holding company that Izzy confesses “pivots a lot.” One agency subsidiary, IEEG, specializes in storytelling and influencer marketing – often by creating a video to tell a brand's story. The influencer marketing portion of their work is based on stories told by influencers, but the influencers are Urban Misfit Venture's employees, each of whom has a massive individual following. Because the employees are the influencers, they can consult with clients and carefully craft the messages that need to be presented. No paying an influencer and trusting that individual to say “the right thing.” IEEG knows what message it wants to send . . . and knows it is going to get it. Urban Misfit Ventures' clients include Milwaukee's professional sports teams and national and international clothing and design brands, who are amazed that the agency and the influencers are one and the same. The second Urban Misfit Ventures' subsidiary, MKE Misfits, is an events company that tells the client's brand story, is very involved in that story, and then provides “quirky” experiential promotions to differentiate itself and its client companies. The company has a major reputation in the Milwaukee area. Urban Misfit Ventures is planning to introduce a number of other specialty subsidiaries I 2019. How did it all start? After a period of separate careers, Quentin, Izzy's college roommate, had met with two of the company's other founders, and then approached Izzy to pitch the idea of Urban Misfit Ventures. After an hour conversation, Izzy was on board, and two weeks later, he quit his job at the bike share. In eight months, the team has grown to 10 employees, including interns. When they started, they traded services for their space at a co-working space, truly starting from scratch. Responses to Visual Speech in Human Posterior Superior Temporal Gyrus Examined with iEEG Deconvolution

Abstract: Experimentalists studying multisensory integration compare neural responses to multisensory stimuli with responses to the component modalities presented in isolation. This procedure is problematic for multisensory speech perception since audiovisual speech and auditory-only speech are easily intelligible but visual-only speech is not. To overcome this confound, we developed intracranial encephalography (iEEG) deconvolution. Individual stimuli always contained both auditory and visual speech but jittering the onset asynchrony between modalities allowed for the time course of the unisensory responses and the interaction between them to be independently estimated. We applied this procedure to electrodes implanted in human epilepsy patients (both male and female) over the posterior superior temporal gyrus (pSTG), a brain area known to be important for speech perception. iEEG deconvolution revealed sustained, positive responses to visual-only speech and larger, phasic responses to auditory-only speech. Confirming results from scalp EEG, responses to audiovisual speech were weaker than responses to auditory-only speech, demonstrating a subadditive multisensory neural computation. Leveraging the spatial resolution of iEEG, we extended these results to show that subadditivity is most pronounced in more posterior aspects of the pSTG. Across electrodes, subadditivity correlated with visual responsiveness, supporting a model in visual speech enhances the efficiency of auditory speech processing in pSTG. The ability to separate neural processes may make iEEG deconvolution useful for studying a variety of complex cognitive and perceptual tasks. Copy rights belong to original authors.
