I was in Dubai Airport waiting for a flight home when I heard the devastating news that one of my best friends, Paul McGlade, had died suddenly. I was in shocked disbelief and broke down in tears. One week later I am recording this podcast both as a memorial to a great friendship and as a wake-up call to myself (and anyone who is listening) as to the importance of healing rifts and keeping friendships alive. In this podcast I recall three destinations I visited with Paul: Goa in India, Marbella and London. Destinations that have given me precious memories of a friend that will last a lifetime. In this episode, we had a conversation with Julie Kagy, director of operations for the Exhibition Services & Contractors Association (ESCA) and Bill McGlade, president of epIQ Creative Group, about their experience in working together as partners on marketing. They shared advice and lessons learned after partnering now for more than one year. Kagy shared insight on what it was like to put together an RFP, evaluate vendors, hone in on the scope of the association's needs, get buy-in on budgeting and execute the strategy with an outside partner. McGlade talked about developing the strategy, prioritizing the client's needs, rebranding an ESCA product, establishing benchmarks, sharing thought leadership, driving engagement and marketing business-to-person. The results: Attendance at ESCA's Summer Education Conference 2023 was up 13%, and social interactions increased 344% year over year. Abstract: The Src-like adaptor proteins (SLAP/SLAP2) bind to CBL E3 ubiquitin ligase to down regulate antigen, cytokine, and tyrosine kinase receptor signaling. In contrast to phospho-tyrosine dependent binding of CBL substrates through its tyrosine kinase binding domain (TKBD), CBL TKBD associates with the C-terminal tail of SLAP2 in a phospho-independent manner. To understand the distinct nature of this interaction, a purification protocol for SLAP2 in complex with CBL TKBD was established and the complex crystallized. However, determination of the complex crystal structure was hindered by apparent SLAP2 degradation during the crystallization process, such that only CBL TKBD residues could be modeled initially. Close examination of the CBL TKBD structure revealed a unique dimer interface that included two short segments of electron density of unknown origin. To elucidate which residues of SLAP2 to model in this unassigned density, a co-expression system was generated to test SLAP2 deletion mutants and define the minimal SLAP2 binding region. As well, SLAP2 degradation products were analyzed by mass spectrometry. Model building and map generation features of the Phenix software package were employed, leading to successful modeling of the C-terminal tail of SLAP2 in the unassigned electron density segments. Recent evidence about the extent of plastics and litter in our oceans has led to grassroots rejection of single-use plastics. This lecture will draw on McGlade's own research (published in 2020) on plastics in our seas. She will show that banning single-use plastics is not enough to safeguard the health of our oceans, and that what is needed is a complete redesign of global production systems.
