Crossrail is a railway construction project underway mainly in central London. Its aim is to provide a high-frequency suburban passenger service crossing London from west to east, to be branded the Elizabeth line, by connecting two major railway lines terminating in London, the Great Western Main Line and the Great Eastern Main Line. The project was approved in 2007, and construction began in 2009 on the central section and connections to existing lines that will become part of the route. About our guests..  Richard joined Arcadis in February 2019. Throughout his career, he has collected both hands-on programme management experience after working on major programmes such as the Jubilee Line Extension, HS1, West Coast Mainline and Crossrail, where he was Programme Controls Director. In his latest role, he is supporting Network Rail’s Transpennine Route Upgrade Programme – a £multi-billion enhancements scheme stretching from Manchester to York. Richard has also collected 6 years in the corporate world helping to run, grow and evolve companies such as Parsons Brinckerhoff, Balfour Beatty and Mace. There are calls for the government to provide Eurostar with more support after the company revealed it doesn't plan to stop in Kent again until 2022. Passengers can't get on or off the high speed service at Ebbsfleet or Ashford and it's feared it could have a devastating impact on the county's economy. Hear from local democracy reporter Ciaran Duggan who's been chatting to the head of HS1. Also in today's podcast, we hear from a bride who's hastily rearranged her wedding to avoid having to cut the number of guests when new restrictions come into force on Monday. Interview with George Frangeskides, Executive Chairman of Alba Mineral Resources (AIM:ALBA)We last spoke to Frangeskides in October. How have things developed since then for Alba Mineral Resources?Alba Minerals Resources is a small but multi-faceted UK-listed company. The company's projects range from the Horse Hill and Brockham oil & gas projects in England, along with polymetallic, multi-mineral projects in Greenland (4), Wales (1), and Ireland (1). Specific commodities of focus include graphite, iron ore, copper, gold, titanium (ilemnite), zinc, and lead. Talk about commodity diversification!Horse Hill is in production (1 & 2). The drilling was successful, bar a water issue. The vertical project is claimed to hold advantages over horizontal projects: around 2-3X. This is all on hold for the time being. The real mainstay is "HS1," which has been producing 250-300 barrels per day. Alba holds an 18% interest in Horse Hill Developments Limited, the owner of a 65% participating interest and operator of the Horse Hill oil and gas project. There is 25-years of planning permission and there is long-term consent from the Oil and Gas Authority (OGA). All of the regulatory work appears to be in place. The plans are to ramp up production to make it more profitable, but this is impossible right now because of the COVID-19-induced oil market crash. Alba Minerals hasn't been putting money into it as of late, but this could turn into a solid cash flow eventually. At the right price, Frangeskides would look to exit on favourable terms. Does the multi-asset, multi-jurisdiction model derisk things, or has Alba Minerals put its eggs in too many baskets? Frangeskides believes it is worth GB£5-5.5M.We then wanted to talk about the 90%-owned Clogau Gold Project in Wales. Alba Mineral Resources has the right to ascend to 100% ownership. The mine closed in 1998, but historically produced 80,000oz, making it the U.K's largest-ever gold producer. Historically, it is claimed that Clogau gold has been found in exceptionally high-grade pockets, with 'bonanza grades 'running from 10-100oz/t gold. In addition, one of the big marketing strategies has been the Welsh gold thematic: Alba Minerals Resources claims that Welsh gold can attract a 'significant' premium on the gold spot price: around a 2 to 3 multiple due to scarcity value. Is this nothing more than a marketing strategy, or is this substantial? The aim is to leverage existing historical data with modern technology, creating a well-positioned, high-grade, low-cost gold mine. Clogau was purchased for c. GB£1M, mainly in shares. Frangeskides doesn't think Alba Mineral Resources needs to produce as much as 80,000oz gold pa in order to make the operation exciting and profitable. 5,000-10,000oz pa would be enough. Welsh gold was last sold at specialist auctions and fetched 8-10X spot price rates in small quantities. Frangeskides is adamant: there is a market here. There are competitors out there, but Frangeskides would be looking to produce high-end, bespoke items that would fetch a "massive premium."What does the cash situation look like? Frangeskides claims the cash situation is sensitive, and that he can't discuss the numbers. I’m getting a ton of questions on genital herpes and oral herpes and it can be pretty confusing so today I want to spend some time on HS1 when it’s down there. A lot of questions I hear are: “I have HSV1 but I also have genital herpes, so does that mean I have HSV2 also?” “I have HSV1 but I don't get cold sores.” “If I have HSV1 down there can my partner get it on their mouth?” “I’m confused because I have genital herpes but HSV1 is oral herpes.” In general, we are told to think oral herpes is only HSV1 and it can only be around your mouth and the “other” kind is genital herpes and its HSV2. This is 100% false and really they are no different from each other.  For the sake of understanding, today’s message let’s get away from associating oral herpes with HSV1. Let's just use HSV1 and HSV2. With fun sexual activities like oral sex, herpes can be transmitted from someone orally to their partner genitally or visa versa genially to their partner's mouth.  This is why so many people are getting oral herpes down there. To make sure we all know how common herpes is, ⅔ people have HSV1 and ⅙ people have HSV2. These statistics show there are more people with HSV1 and therefore it’s very common to be diagnosed with genital HSV1. It’s important to find out from your doctor when you are diagnosed with herpes if it’s HSV1 or HSV2. This way you can have a conversation with your partner and your potential future partners. For example, if partner A has HSV1 genitally and partner B has HSV2 genitally they can transmit herpes to each other because they are different strains of the virus. I’ve seen this actual situation happen to a number of couples and then boom...a whole new outbreak.   So the bottom line is HSV1 and HSV2 can go wherever you give it a place to go. The previous categories of genital vs oral are not specific enough and in order to fully communicate with our lovers we need to know what strain and where we have it. The government is stepping up its support for HS2, the high-speed rail project due to link London and Birmingham by 2026 with extensions to Manchester and Leeds by 2032. The cost is officially estimated to be £42.6bn and could rise to more than £51bn if, as expected, the scheme incurs VAT. Opponents foresee further increases and have predicted an eventual bill of £80bn for taxpayers.Who stands to gain from the project and who will be the losers? The government has published detailed maps of the route to be taken by the first stage, leading to calamitous falls in the value of many nearby properties. Towns and cities which are near the route but not linked to HS2 fear that their economies will suffer as businesses are attracted to Birmingham and the northern ends of the line. Current fast train services are due to suffer drastic cuts in the wake of HS2 and some major development plans are now deemed to be at risk.Meanwhile, economic advisers in the three major cities are planning for billions of pounds worth of benefits as travel-times and congestion on the existing network are reduced.Gerry Northam reports from areas which expect to benefit from HS2 and those which could lose out and asks what lessons can be learned from the impact of Britain's first high-speed rail project - HS1 in Kent.Producer: Ian Muir-CochraneEditor: David Ross.

The OSTL gene is localized at the band q23 in the chromosome 6. Its localization corresponds to a translocation breakpoint between chromosomes 6 and 12, the t(6;12)(q23;p13), that was characterized in our group in an acute lymphoblastic leukemia cell line. This translocation involves the ETV6 (translocation ETs leukemia) gene localized in chromosome 12 with the STL (six twelve leukemia gene) gene localized in chromosome 6. The STL gene shares the first exon with a novel gene, that we named OSTL (opposite STL), but they are transcribed in opposite directions. Since the fusion gene ETV6/STL encodes only for a very small protein which lacks any known functional domain, we speculate that the main leukemogenic effect of this translocation is the deregulation of OSTL. OSTL has a RING-Finger motif that is highly conserved between species and has a significant homology with other genes in human as well as C. elegans, D. melanogaster, and S. cerevisiae. OSTL showed a very specific expression pattern during the mouse embryogenesis. The aim of this project was the functional characterization of OSTL, with special emphasis in normal hematopoiesis and leukemogenesis. Therefore we have sequenced the whole human and mouse OSTL cDNA by using OSTL cDNA clones from the RZPD (“Resource Zentrum Primäre Datenbank”) in Berlin. These sequences encode for a 307 (mouse) and a 275 (human) amino acids length protein. The protein length differences between human and mouse are explained because of the existence of alternative spliced exons. The homology between human and mouse sequence is 99% at the protein level. The expression of GFP-OSTL fusion protein in mouse fibroblast cell line enable us to observe the subcellular localization of OSTL protein. GFP-OSTL is localized mainly in the cytoplasm, showing small spots, probably in the mitochondrial region. In a mouse multiple tissue Northern blot, we could show that OSTL is expressed in testis, ovary and liver. In an human multiple tissue Northern, OSTL expression was observed in skeletal muscle, testis, ovary, heart, placenta, pancreas and prostate. Northern blotting with different human cell lines revealed expression of OSTL in three EBV (Epstein Barr Virus) transformed lymphoblastoid cell lines (LCL B, LCL D, and LCL R) and in one NHL (Non-Hodgkin Lymphoma) cell line (Karpas 422). In Reverse Transcriptase PCR experiments using B cell in different maturation stages, the expression of OSTL was observed in naive, memory B and plasma cells, and in leukemic patient samples, expression was observed in several AML and ALL cDNAs. Whole mount in situ hybridization experiments were performed to investigate the temporo-spatial expression pattern of OSTL during mouse embryogenesis. There was distinct expression of Ostl in the somites (myotome), first and second branchial arches, optic and otic vesicles, in the hair follicles of the vibrissae, and limb buds in mouse embryos of embryonal days 9.5 to 14.5. This expression pattern suggests an important role for Ostl in the early development of these structures. Aiming to find protein interaction partners of OSTL, we performed a Yeast Two Hybrid assay using a Hela cDNA library. Among others we found interaction of OSTL with the antiapoptotic protein, HAX-1 (HS1-associated protein X-1), that is involved in the regulation of B-cell signal transduction, and interaction with the pro-apoptotic protein, SIVA. SIVA was originally identified as an interaction partner of CD27 (TNFRSF7), a member of the TNF-receptor superfamily, which is expressed in B cells. These interactions were confirmed by in vitro (cotransformation in yeast, CoIP) and in vivo (colocalization of these proteins in mammalian cells and CoIP) assays. Overexpression of Ostl in primary mouse hematopoietic cells followed by injection of the cells into lethally irradiated mice resulted in a T-Acute-Lymphoblastic-Leukemia (T-ALL) phenotype. In summary, our experiments could demonstrate that OSTL is important in B cell development and signaling and deregulation of this gene can contribute to the development of hematologic malignancies.
