prostaglandins and their related compounds- I prostacyclins (PGl), thromboxanes (TXA), leukotrienes (LT) and lipoxins are collectively known as eicosaniods, since they all contain 20 carbons (Creek : eikosi-twenty). Eicosanoids are considered as locally acting hormones with a wide range of biochemical functions. History : Prostaglandins (PCs) were first discovered in human semen by Ulf von Euler (of Sweden) in 1930. These compounds were found to stimulate uterine contraction and reduce blood pressure. von Euler presumed that they were synthesized by prostate gland and hence named them as prostaglandins. lt was later realized that PCs and other eicosanoids are synthesized in almost all the tissues (exception- erythrocytes). By then, however, the name prostaglandins was accepted worldwide, and hence continued. The prostaglandins E and F were first isolated from the biological fluids. They were so named due to their solubility in ether (PCE) and phosphate buffer (PCF, F for fosfat, in Swedish). All other prostaglandins discovered later were denoted by a letter-PCA, PCH etc. Structure o{ prostaglandins Prostaglandins are derivatives of a hypothetical 2O-carbon fatty acid namely prostanoic acid hence known as prostanoids. This has a cyclopentane ring (formed by carbon atoms 8 to 12) and two side chains, with carboxyl group on one side. Prostaglandins differ in their structure due to substituent group and double bond on cyclopentane ring. The different prostaglandins are given in Fig.32.l. The structures of the most important prostaglandins (PGF2 and PGF2o), prostacyclins (PCl2), thromboxanes (TXA2) and leukotrienes (LTA+) along with arachidonic acid are depicted in Fi9.32.2. A subscript numeral indicates the number of double bonds in the two side chains. A subscript c-denotes that the hydroxyl group at Ce of the ring and the carboxyl group are on thesame side of the ring.Synthesis of prostaglandins Arachidonic acid (5,8,1 1,1 4-eicosatetraenoic acid) is the precursor for most of the prostaglandins inhumans. The biosynthesis of PCs was described by scene Bergstrom and Bengt Samuelsson (1960). lt occurs in the endoplasmic reticulum in the following stages, as depicted in Fi9,32.3. 1. Release of arachidonic acid from membrane bound phospholipids by phospho- lipase A2-this reaction occurs due to a specific stimuli by hormones such as epinephrine or bradykinin. 2. Oxidation and cyclization of arachidonic acid to PGG2 which is then converted to PCH2 by a reduced glutathione dependent peroxidase. 3. PGH2 serves as the immediate precursor for the synthesis of a number of prostaglandins, including prostacyclins and thromboxanes. The above pathway is known as cyclic pathway of arachidonic acid. ln the linear pathway of arachidonic acid, leukotrienes and lipoxins are synthesized (details given later). Cyclooxygenase-a suicide enzyme : lt is interesting to note that prostaglandin synthesis can be partly controlled by suicidal activity ofthe enzyme cyclooxygenase. This enzyme is capable of undergoing self-catalysedestruction to switch off PG synthesis. lnhibition of PG synthesis : A number of structurally unrelated compounds can inhibit prostaglandin synthesis. Corticosteroids (e.g. cortisol) prevent the formation of arachidonic acid by inhibiting the enzyme phospholipase 42. Many non-steroidal anti-inflammatory drugs inhibit the synthesis of prostaglandins, prostacyclins and thromboxanes. They do so by blocking the action of the enzyme cyclo' oxygenase. Aspirin inhibits PG synthesis :Aspirin (acetyl salicylic acid) has been used since nineteenth century as an antipyretic (fever-reducing) and analgesic (pain relieving). The mechanism of action of aspirin however, was not known for a Iong period. lt was only in 1971, John Vane discovered that aspirin inhibits the synthesis of PC from arachidonic acid. Aspirin irreversibly inhibits the enzyme cyclooxygenase.

Ele foi ter com o rei e lhe disse: “Numa cidade havia dois homens, um rico e outro pobre. 2O rico possuía ovelhas e bois em grande número. 3O pobre só possuía uma ovelha pequenina, que tinha comprado e criado. Ela crescera em sua casa junto com seus filhos, comendo do seu pão, bebendo do mesmo copo, dormindo no seu regaço. Era para ele como uma filha. 4Veio um hóspede à casa do homem rico, e este não quis tomar uma das suas ovelhas ou um dos seus bois para preparar um banquete e dar de comer ao hóspede que chegara. Assim diz o Senhor, o Deus de Israel: 10‘Por isso, a espada jamais se afastará de tua casa, porque me desprezaste e tomaste a mulher do hitita Urias para fazer dela a tua esposa'. 11Assim diz o Senhor: ‘Da tua própria casa farei surgir o mal contra ti e tomarei as tuas mulheres, sob os teus olhos, e as darei a um outro, e ele se aproximará das tuas mulheres à luz deste sol. 12Tu fizeste tudo às escondidas. Primeira Leitura: Neemias 8,1-12 Leitura do livro de Neemias – Naqueles dias, 1todo o povo se reuniu como um só homem na praça que fica defronte da porta das Águas e pediu ao escriba Esdras que trouxesse o livro da Lei de Moisés, que o Senhor havia prescrito a Israel. 2O sacerdote Esdras apresentou a Lei diante da assembleia de homens, de mulheres e de todos os que eram capazes de compreender. Era o primeiro dia do sétimo mês. 3Assim, na praça que fica defronte da porta das Águas, Esdras fez a leitura do livro, desde o amanhecer até o meio-dia, na presença dos homens, das mulheres e de todos os que eram capazes de compreender. E todo o povo escutava com atenção a leitura do livro da Lei. 4Esdras, o escriba, estava de pé sobre um estrado de madeira, erguido para esse fim. 5Estando num lugar mais alto, ele abriu o livro à vista de todo o povo. E, quando o abriu, todo o povo ficou de pé. 6Esdras bendisse o Senhor, o grande Deus, e todo o povo respondeu, levantando as mãos: “Amém! Amém!” Depois, inclinaram-se e prostraram-se diante do Senhor com o rosto em terra. 7Os levitas explicavam a Lei ao povo, e cada um ficou em seu lugar. 8E leram clara e distintamente o livro da Lei de Deus e explicaram seu sentido, de maneira que se pudesse compreender a leitura. 9O governador Neemias e Esdras, sacerdote e escriba, e os levitas que instruíam o povo, disseram a todos: “Este é um dia consagrado ao Senhor, vosso Deus! Não fiqueis tristes nem choreis”, pois todo o povo chorava ao ouvir as palavras da Lei. 10E Neemias disse-lhes: “Ide para vossas casas e comei carnes gordas, tomai bebidas doces e reparti com aqueles que nada prepararam, pois este dia é santo para o nosso Senhor. Não fiqueis tristes, porque a alegria do Senhor será a vossa força”. 11E os levitas acalmavam todo o povo, dizendo: “Ficai tranquilos; hoje é um dia santo. Não vos aflijais!” 12E todo o povo se retirou para comer e beber. Distribuíram também aos outros e expandiram-se em grande alegria, pois haviam entendido as palavras que lhes tinham sido explicadas. – Palavra do Senhor. Infundi Senhor em Nossos corações a vossa graça nós vo-lo suplicamos afim de que, conhecendo, pela anunciação do anjo Gabriel, a encarnação de Jesus Cristo vosso Filho, cheguemos pelos merecimentos de sua paixão e morte de cruz, à glória da ressurreição, pelo mesmo Jesus Cristo vosso Filho que convosco vive e reina na Unidade do Espírito Santo. Amém! A palavra que nos orienta hoje é do livro do Êxodo 2. 1Moisés apascentava o rebanho de Jetro, seu sogro, sacerdote de Madiã. Um dia em que conduzira o rebanho para além do deserto, chegou até a montanha de Deus, Horeb.2O anjo do Senhor apareceu-lhe numa chama (que saía) do meio a uma sarça. Moisés olhava: a sarça ardia, mas não se consumia.3“Vou me aproximar, disse ele consigo, para contemplar esse extraordinário espetáculo, e saber porque a sarça não se consome.”4Vendo o Senhor que ele se aproximou para ver, chamou-o do meio da sarça: “Moisés, Moisés!” “Eis-me aqui!” respondeu ele.5E Deus: “Não te aproximes daqui. Tira as sandálias dos teus pés, porque o lugar em que te encontras é uma terra santa.6Eu sou, ajuntou ele, o Deus de teu pai, o Deus de Abraão, o Deus de Isaac e o Deus de Jacó”. Moisés escondeu o rosto, e não ousava olhar para Deus.7O Senhor disse: “Eu vi, eu vi a aflição de meu povo que está no Egito, e ouvi os seus clamores por causa de seus opressores. Sim, eu conheço seus sofrimentos.8E desci para livrá-lo da mão dos egípcios e para fazê-lo subir do Egito para uma terra fértil e espaçosa, uma terra que mana leite e mel. (EX 2,1-8) O povo de Deus estava escravo e rogava a Deus um instrumento de libertação, Deus, através do seu anjo, manifestou-se a Moisés e o enviou para que ele pudesse ser o condutor do povo no caminho da libertação. Não foi uma tarefa fácil, mas Moisés creu e fez o povo crer. Lembro-me de uma história muito impactante na minha vida. Certa vez fizeram uma caravana para Aparecida do Norte, uma viagem de quase quinhentos quilômetros. E na volta um senhor ficou esquecido no meio do caminho. A caravana chegou à cidade e procuravam por ele. Um senhor chamou o filho e disse, pegue seu carro e vamos varrer a estrada, e assim fizeram, quando eles chegaram próximo à Volta Redonda avistaram o senhor que estava perdido em uma guarita de ônibus. Ao ver o anjo o senhor que estava perdido disse: “Eu roguei a Deus, não sabia mais o que fazer, estou sujo, como fome e cansado”, vocês foram a resposta da minha oração. Às vezes não precisamos ir tão longe para ser um anjo libertador em nome de Deus, mas precisamos sempre sair do nosso comodismo e nos colocar à disposição da graça divina. EXERCÍCIO DE HOJE – jejum do instrumento – haverá sempre alguém ao nosso redor que precisa de libertação. Oração final para todos os dias – Pai Nosso e gloria à Trindade Santa – santo anjo do Senhor.

My go to breakfast at the bakery because they make me feel full longer and I like the tang from the cranberries as well as the buttermilk. Buttermilk is one of the key ingredients in adding more nutrition, more tang, as well as softening the gluten content with the higher acid base. This recipe yields six muffin. 1 cup of flour-pastry or all purpose 1 cup of bran 1 cup of whole wheat flour, gold or white wheat if you got it. 1/2 tsp salt 1/2 tsp of baking soda 1 Tablespoon and 1 ½ tsp of baking powder Mix together in a mixing bowl until thoroughly combined. In a separate bowl combine the following ingredients: 2 Eggs 1 cup and 2 T of buttermilk, you need buttermilk to use with the baking soda to leaven the product, If not, then substitute a cup of milk, 1 T and 1 T lemon juice or white vinegeger. LET STAND TEN MINUTES to allow the milk to become curdled. 1/4 cup of melted butter 1/3 cup of honey Combine wets and dries together until incorporated. Do not over mix as you will strengthen the gluten and the muffins will be tough and dense. Fold in ¾ cup of raisins or cranberries. Then scoop into well-greased muffin tins. The mixture should be less than even with the muffin tins as they will grow in the oven. Everyone loves a nice muffin top but overfilled muffins fuse together and make separation as well as clean up, a nightmare. Bake at 350 degrees and set the timer for 2O minutes and add time as needed. The bran muffins store well frozen. You can also save half the batter and bake off the next day. It holds up way better than the other muffin recipes but there still will be some deflating of the leaving action, so the product is a bit denser to the discerning pallet. Most people will hardly know the difference. So, if you have time constraints and want to make this batter in advance, then you certainly can without same degree of compromise in flavor/taste as you would get with let’s say my blueberry muffin recipe. Ziel dieser Arbeit war zum Einen die Synthese von neuartigen (Bakterio-)Chlorophyll-Derivaten zur Einlagerung in Proteine und ihre Charakterisierung in Lösung, zum Anderen Bindungsstudien an Komplexen dieser Derivate mit modularen Proteinen und dem Lichtsammler-Komplex 1 aus Rhodobacter sphaeroides. 1.) Darstellung von Fe-(Bakterio-)Pheophytinen: Es wurde ein Verfahren etabliert, das die Metallierung von Pheophytin a, Bakteriopheophytin a und deren Derivate mit Eisen ermöglicht. Zusätzlich wurde diese Methode so weit modifiziert und optimiert, dass ausgehend vom Mohrschen Salz auch die Einlagerung von 57Fe möglich ist, wodurch eine Erweiterung der spektroskopischen Methoden (Mößbauer-Spektroskopie) erreicht wird. Da die Fe Komplexe nicht mit den für (Bakterio-)Chlorophyll-Derivate etablierten Methoden gereinigt werden können, wurde für diese Komplexe ein neues Chromatographiesystem entwickelt. 2) Spektroskopische Untersuchung der Fe-(Bakterio-)Pheophytine: Es ist bekannt, dass Fe-Porphyrine leicht zu µ-oxo-Komplexen (Fe(III)(B)Phe a)2O dimerisieren und das Zentralion in zwei Oxidationsstufen (+2 und +3) vorliegen kann. Die Dimerisierung des Fe Phe und Fe-BPhe wurde durch Säure-Base-Titration absorptionsspektroskopisch untersucht. In aerober Lösung liegt das Zentralmetall des Fe-(B)Phe dreiwertig vor ((Fe(III)(B)Phe a)Cl). Dieses lässt sich „klassisch“ mit Na-Dithionit allein durch Ligandierung mit Pyridin zum zweiwertigen Fe(II)(B)Phe a reduzieren. Die drei Zustände der Fe-Komplexe (Fe(III)(B)Phe a)Cl, (Fe(III)(B)Phe a)2O und Fe(II)(B)Phe a wurden durch ESR- und Absorptionsspektroskopie charakterisiert. Die Oxidationsstufen der drei Zustände wurden für 57Fe-Me-Pheid a durch Mößbauerspektroskopie bestätigt. 3) Einlagerung von Fe-Bakteriopheophytin a in LH1 von Rhodobacter sphaeroides: Zur Untersuchung, ob Fe-BPhe von BChl-Bindungstaschen akzeptiert wird, wurde versucht Fe-BPhe ins LH1 von Rb. sphaeroides einzulagern. Die Ergebnisse dieser Untersuchungen deuten zwar auf einen Einbau von Fe-BPhe hin, allerdings nur in sehr geringem Maß. Zur Quantifizierung des Fe-BPhe-Gehalts wurden verschiedene Methoden getestet. Der einfachste und vielversprechendste Weg war die Differenzabsorptionsspektroskopie, bei der die unterschiedliche Absorption von µ-oxo-Dimer und Monomer des Fe-BPhe ausgenützt wird. 4) Darstellung von Formyl-(Bakterio-)Chlorophyll-Derivaten: Eine kovalente Bindung von Chlorophyll-Derivaten an synthetische Peptide ist durch die Kopplung von Formyl-Gruppen mit einem modifizierten Lysin-Rest unter Bildung eines Oxims möglich. [3 Formyl]-Me-Pheid a konnte durch oxidative Spaltung des C-3-Vinyl des Chlorophyll a mit Ozon hergestellt werden. Ebenfalls mit Ozon konnte die Phytyl-Doppelbindung von Pheophytin a unter Bildung des Ethanal-Pheid a erreicht werden. Somit stehen insgesamt drei Chlorophyll-Derivate für die kovalente Bindung an synthetische Peptide zur Verfügung, welche die Formyl-Gruppen an verschiedenen Positionen tragen, wodurch eine unterschiedliche Orientierung der Pigmente im Protein erreicht werden kann. Es wurde versucht, in Analogie zum Pheophytin a das [3-Vinyl]-Me-BPheid a mit Ozon zu spalten. In Folge der leichten Oxidation des Makrozyklus lieferte diese Reaktion das Zielprodukt [3-Formyl]-Me-BPheid a nur in äußerst geringen Mengen, so dass diese Methode für die präparative Synthese dieser Verbindung nicht geeignet ist. 5) Nicht-kovalente Bindung von [M]-BPheid (M = Ni, Zn, Fe) in synthetische modulare Proteine (MOP): Ni-, Zn-, und Fe-BPhe wurden auf ihre Komplexbildung mit 216 verschiedenen, synthetischen Vier-Helix-Bündel-Proteinen untersucht. Die [M] BPheid-MOP-Komplexe wurden absorptionsspektroskopisch auf die Stärke der Bindung, die Koordination des Zentralmetalls und auf die Hydrophobizität der Umgebung untersucht. Alle MOP binden [M] BPheid in sehr unterschiedlichem Maße. Stärke und Art der Bindung werden in erster Linie durch die Bindehelix bestimmt. Eine quantitative Modulation findet allerdings auch durch die Abschirmhelix statt. Ni-BPheid zeigt in den Komplexen drei mögliche Koordinationszustände (nc = 4, 5, 6). Eine hohe Koordinationszahl geht immer mit einer stabilen Bindung und einer schmalen Qy Bande einher. Zn-BPheid ist in allen Komplexen fünffach koordiniert, das Fe-BPheid vierfach. Qualitativ zeigen alle drei Pigmente ein gleiches Muster in Bezug auf das Bindungsverhalten, so dass ausgehend von Häm-Bindungstaschen die Bildung von BChl-Bindungstaschen bestätigt werden konnte.
