My favorite part of the interview is when he talks about the four powerful “Aha's” that came out of our session together. 1. The importance of systems and processes Karl emphasizes the value of having well-defined systems and processes in a business, rather than just chasing revenue and profit. He explains the concept of the "flywheel" and how consistent, iterative improvements to processes can lead to compounding results over time. 2. Therman's business and leadership challenges Therman discusses his background as a consultant, developer, and trainer, and his desire to focus more on the training aspect of his business, which he sees as more scalable. He shares his concerns about taking on a leadership role and managing employees, as he has not had that experience before. 3. Embracing the "Chief Repeating Officer" mindset Therman has an "aha" moment when Karl introduces the concept of being a "Chief Repeating Officer" - someone who consistently communicates the vision and importance of tasks to their team. Therman realizes that he already does this effectively with his customers, and can apply the same approach with his family and any future employees. 4. Focusing on one course and building a system Therman acknowledges that he has been trying to work on multiple courses simultaneously, which has prevented him from fully optimizing any one of them. He decides to focus his energy on his "Risk Register" course, building a structured process around it, including potentially hiring a co-host to help with accountability and production. 5. Developing a roadmap and setting expectations Therman and Karl discuss creating a 3-month roadmap, with the first 6 weeks focused on Therman embracing the "Chief Repeating Officer" mindset, particularly with his family. One of my favorite parts of the interview is his story about why he didn't think he could document his leadership approach. Highlights from the interview: 1. Therman's leadership journey and challenges Therman shared that he has struggled with being intentional about his leadership, as he has naturally fallen into leadership roles but finds it challenging to directly manage and hold people accountable. He discussed how he is more comfortable providing advice and guidance when disconnected from the outcome, versus when he has a vested interest in the results as a business owner. 2. Documenting processes, values, and communication norms Karl suggested that Therman document his leadership approach, including standard operating procedures, core values, and communication norms. This would help new hires understand Therman's expectations and style, rather than relying solely on verbal guidance. They discussed the importance of balancing prescriptive processes with flexibility to allow for creativity and problem-solving. 3. Overcoming distractions and maintaining focus Therman shared that he struggles with maintaining focus and feeling like he's not doing enough, even when he's accomplished his goals for the day. They discussed strategies like setting stretch goals, celebrating small wins, and using checklists or Pomodoro techniques to stay on track. 4. Gratitude and self-reflection Karl emphasized the importance of gratitude and regularly reflecting on what's going well, rather than constantly focusing on what could be improved. Investigating the ruins they come upon a human searching for something and muttering ramblings not understandable. As Therman interacts he has a sense of knowing the human as a friend. Things start to get really weird as the rest of the party sense intense evil and engage only to find all is not what it seems. What lies within front of them and below them in the ruins may bring death to everyone and everything as the group come closer to meeting the fate that has been calling to them. The Secular Reliquaries of Therman Statom Therman Statom – sculptor, glass artist, and painter – is most notably known as a pioneer of the contemporary glass movement for his life-size glass ladders, chairs, tables, constructed box-like paintings, and small-scale houses; all created through the technique of gluing glass plate together. Sandblasted surfaces become a canvas for spontaneous vibrant colors and line work, which take nuances from Abstract Expressionism and concepts of Minimalism, while simultaneously incorporating a twist by using blown-glass elements and found objects. Born in Winter Haven, Florida in 1953, Statom spent his adolescence growing up in Washington, D.C. His interest in the arts grew from a fondness of painting and he began to investigate ceramics at RISD. However, after an experimental glassblowing session with Dale Chihuly, he was soon hooked on the spontaneity of hot glass and its limitless possibilities. Statom went on to pursue studies at Pilchuck Glass School during its inaugural year, completing a BFA in 1974 from RISD, and later studied at the Pratt Institute of Art & Design. Throughout his career, public artworks have been permanently installed at prominent locations including the Los Angeles Public Library, Corning, Inc. Headquarters, the Mayo Clinic, San Jose Ice Center, the Toledo Museum of Art, and the Jepson Center for the Arts in the Telfair Museum, Savannah as well as several hospitals across the country. Statom’s artwork appears in numerous exhibitions annually, including solo and group shows around the nation and internationally. Over the span of his career, he has completed over 30 large, site-specific installations. Most notably in recent years, his 2009 solo exhibition Stories of the New World, at the Orlando Museum of Art, which spanned over 5,000 square feet, has been his largest installation to date. Exploring themes related to Juan Ponce de Leon’s 1513 search for the fabled Fountain of Youth as a point of departure, the installation referenced historic and contemporary themes of hope, discovery, ambition, and destiny. Visitors traversed the gallery space consisting of a mirrored maze, panoramic glass wall mural, a room-size structure built entirely of glass, and video projections. In conjunction with the exhibit, Statom partnered with the educational department of the OMA and the Zora Neale Hurston National Museum of Eatonville to work with over 80 young students to create a work of art titled “Glass House,” which was a large, walk-though structure built from glass boxes designed by the children. The piece was later displayed at the annual summer community festival. Much of the latter half of Statom’s career has been focused on the importance of educational programming within the arts. He has taken a deep interested in employing workshops as a catalyst for social change and in affect, positively impacting a community. Working directly with the artist himself, adults and children alike share a combined experience of exploring art making via a hands-on experience. Inhibitions and limitations are left by the wayside, and the practice or act of “doing” becomes a journey of self-discovery, creating an opportunity for the participant to go to a new place within themselves. Says Statom: “I believe art can be understood both conceptually and intuitively. I think there is a need for the general public to come to an understanding that to appreciate art and creativity they must trust his or her self; that extensive education is not a prerequisite for understanding art. Much of what I do is seeded in what is more of an intuitive process; a large portion of my work is exploring these processes within people and their environments. “The fact is, I believe that creativity is a part of all aspects of what people do; my studio and educational efforts via workshops and the support of outside programming, general educational and cultural institutions, are a reflection of this belief. I feel that art is tool for empowerment and education. It’s also a viable tool to investigate positive change and engage a culture through exploration.”

Abstract: Human astrocytes are multifunctional brain cells and may contribute to the pathophysiology of schizophrenia (SCZ). We differentiated astrocytes from induced pluripotent stem cells of monozygotic twins discordant for SCZ, and found sex-specific gene expression and signaling pathway alterations related particularly to inflammation and synaptic functions. While Ingenuity Pathway Analysis identified SCZ disease and synaptic transmission pathway changes in SCZ astrocytes, the most consistent findings were related to collagen and cell adhesion associated pathways. Neuronal responses to glutamate and GABA differed between astrocytes from control persons, affected twins, and their unaffected co-twins, and were normalized by clozapine treatment. SCZ astrocyte cell transplantation to the mouse forebrain caused gene expression changes in demyelination, synaptic dysfunction and inflammation pathways of mouse brain cells and resulted in behavioral changes in cognitive and olfactory functions. Altogether, our results show that astrocytes contribute to both familial risk and clinical manifestation of SCZ in a sex-specific manner. Copy rights belong to original authors. This math major went on to enjoy a 34-year career in the insurance industry as an actuary. Abstract: The molecular pathophysiological mechanisms underlying schizophrenia have remained unknown, and no treatment exists for primary prevention. Studies using stem cell-derived neurons have investigated differentially expressed genes (DEGs) and GO and KEGG pathways between patients and controls, but not analyzed data-driven causal molecular pathways involved. We used Ingenuity Pathway Analysis (IPA) to analyze canonical and causal pathways in two different datasets, including patients from Finland and USA. The most significant findings in canonical pathway analysis were observed for glutamate receptor signaling, hepatic fibrosis, and glycoprotein 6 (GP6) pathways in the Finnish dataset, and GP6 and hepatic fibrosis pathways in the US dataset. In data-driven causal pathways, ADCYAP1, ADAMTS, and CACNA genes were involved in the majority of the top 10 pathways differentiating patients and controls in both Finnish and US datasets. In contrast, no dopamine-specific genes were consistently involved. Results from a Finnish nation-wide database showed that the risk of schizophrenia relapse was 41% lower among first-episode patients during the use of losartan, the master regulator of an ADCYAP1, ADAMTS, and CACNA-related pathway, compared to those time periods when the same individual did not use the drug. This association was not attributable to general adherence to drug treatments. The results from the two independent datasets suggest that the GP6 signaling pathway, and the ADCYAP1, ADAMTS, and CACNA-related purine, oxidative stress, and glutamatergic signaling pathways are primary pathophysiological alterations in schizophrenia among patients with European ancestry. While no reproducible dopaminergic alterations were observed, the results imply that agents such as losartan, and ADCYAP1/PACAP-deficit alleviators, such as metabotropic glutamate 2/3 agonist MGS0028 and 5-HT7 antagonists, which have shown beneficial effects in an experimental Adcyap1 -/- mouse model for schizophrenia, could be potential treatments before the full manifestation of illness involving dopaminergic abnormalities.
