While UPP Liquids may be their latest home, Tonya Cornett and Ian Larkin have been brewing together, then apart, then together again, across three different breweries over the past decade and a half. Over that time, they've won countless medals in competitions large and small—including Cornett's prestigious Russell Schehrer Award for Innovation in Craft Brewing—and they continue that tradition of winning while pushing and challenging each other at UPP in Bend, Oregon. Luke Cornett won the Maumelle Classic this weekend with a record 15-under
In this episode we speak with Alon Goren from UC San Diego about his work at the intersection of genomic technology development and chromatin biology. We discuss how his lab studies how the epigenome is regulated, how disruption of that regulation contributes to disease, and how technology can be improved to make results more robust and reproducible. We talk about his early interest in biology, how that developed through medical research training, and how a molecular biology lab shaped the direction of his career. He explains how curiosity about how cells and organisms work led him toward genomics and chromatin research. We then discuss several methods from his career, including early direct sequencing approaches for small amounts of DNA and RNA, ChIP-based methods for chromatin regulators, and work on improving ChIP-seq workflows. He explains why antibody choice matters, why monoclonal antibodies can improve reproducibility, and how automation helped scale the process. We also cover his work on spike-in normalization, including the risks of using exogenous chromatin incorrectly and the need for better safeguards in genome-wide comparisons. He describes a newer approach that uses two spike-ins to provide multiple checks on normalization. Finally, we discuss his work on short tandem repeats, zebrafish heart regeneration, and SIRT6-related polymerase pausing, as well as a newer platform that converts molecular interactions into sequencing-readable barcodes. He closes by stressing the importance of validation, careful protocol design, and methods that can be used reliably by multiple people. Tonya Cornett thought she would end her career at 10 Barrel Brewing. She had joined the Bend, Oregon-based craft brewery in 2012, piled up competition medals for her beers and created a following at the brewery. She thrived as the brewery's R&D brewmaster before and after its 2014 sale to Anheuser-Busch InBev (A-B). But then Tilray Brands acquired 10 Barrel and seven other brands in 2023. A year later, Tilray laid off Cornett and members of her team in a move that shocked many within the industry. On this week's Brewbound Podcast, Cornett discusses her journey over the last decade-plus and her next chapter with Upp Liquids, a new venture in the rebranded Immersion Brewing space. Cornett shares her experiences at A-B, how 10 Barrel and her role changed under Tilray's ownership and her goals with Upp moving forward. As the Great American Beer Festival (GABF) kicks off in Denver, Cornett also explains how medals have served as a stamp of approval throughout her career and why they remain relevant. Cornett's beers have received numerous competition medals throughout the years, including 28 medals at the Brewers Association's (BA) GABF and World Beer Cup. That doesn't include 10 Barrel's 2024 clean sweep of the German-style Sour Ale category during the 2024 GABF competition. Singer, songwriter, and actress Amanda Page Cornett is bringing southern rock n' soul to the world! Featured in the 2009 movie, "Junkyard Dog,” Cornett also has acted in various films, music videos, and commercials. Amanda's band, Almost Angels, features the sibling trio Sherry Peterson-Tenille, Shawn Peterson, and Randall Scott Peterson. Together, they have had the honor of playing with special guest musicians, including bassist Tim Lawter ( Marshall Tucker Band) as well as Rock ‘n' Roll Hall of Fame drummer Artimus Pyle (Lynryd Skynryd). Released in 2018, Cornett's album Front Porch Rebel was recorded at NuttHouse Studio in Sheffield, AL and features songs written at Muscle Shoals' famous Fame Studios (Aretha Franklin, Lynyrd Skynyrd, Allman Brothers, Etta James). Amanda has released, with great acclaim, numerous singles, including "Carolina Coast", "Throw A Lil Gas", "Six String Showdown", and "Soldier's Girl".
