#541: Ever wondered if you're making the right choice between a Traditional and Roth 401(k)? You're not alone. In this episode, Katie Gatti Tassin, host of MorningBrew's Money with Katie podcast, joins us to tackle this common retirement savings dilemma. We deep-dive into the debate between using Traditional vs. Roth 401(k) accounts for retirement savings, in the context of: Future tax rates Tax complexities for small business owners and high earners Social Security uncertainty Stock-based compensation Incentives for business owners vs. employees Katie explains her strategy for maximizing retirement savings while minimizing taxes. She suggests that for some people in higher tax brackets, maxing out a traditional 401(k) and then investing the tax savings elsewhere might be the way to go. But as we dig deeper, it becomes clear that there's no one-size-fits-all answer. We explore the Traditional vs Roth question, discussing how your current income and expected retirement spending can affect your choices. It's not just about the math, though. The unpredictability of future tax rates and policies adds another layer of complexity to the decision. Social Security plays a major role, as well. We discuss its current funding situation and the challenges it might face in the future. This leads to a fascinating discussion about how AI might impact future costs and lifestyles. Could things actually get cheaper in the future? Taxes for high earners and small business owners is another focus. We break down some misconceptions about who falls into high tax brackets. It's not always as simple as it seems. Stock-based compensation is another hot topic. We discuss how it affects corporate decision-making and the wider economy. This leads to an interesting comparison of the incentives for business owners versus employees. Throughout the episode, we keep coming back to one key point: no matter which type of account you choose, the most important thing is to contribute as much as you can. Your contribution amount has a bigger impact on your retirement savings than the type of account you use. By the end of this interview, you'll have a better understanding of the factors that go into choosing between a Traditional and Roth 401(k). More importantly, you'll see how this decision fits into the bigger picture of retirement planning and overall financial health. Jesse explores the intricacies of student loan forgiveness, highlighting the fallacy that loans can simply vanish without consequence. He argues that the financial burden ultimately falls on taxpayers and critiques the notion that the U.S. government can "magically" eliminate student debt. Jesse explains the Public Service Loan Forgiveness (PSLF) program and how student debt exacerbates mental health problems. He proposes a balanced approach of partial interest forgiveness to alleviate borrower burdens, shorten repayment periods, and benefit both individuals and the government, all while maintaining personal accountability. Today's guest, Katie Gatti Tassin, is the founder of Money with Katie, a platform she created in 2020, which was later acquired by Morning Brew. During the interview, Jesse and Katie discuss frustrations with the American healthcare system, underscoring how flawed incentives impede rational decision-making. Most of the conversation revolves around financial literacy and misconceptions among the FIRE community and broader society. As Jesse and Katie reach the end of the discussion, they share their thoughts on how social media and social isolation affect us financially and mentally. This is a wide-ranging conversation with a few insightful tangents. If you're looking for a personal, real, and thoughtful finance podcast, then this is the episode for you. Key Takeaways: • Loan forgiveness doesn't make the loan disappear… Someone is eating that cost. • Jesse explains how the Public Service Loan Forgiveness (PSLF) program works. • The broken incentives of the American healthcare industry discourages rational decisions. • How “tough love” online finance personalities like Dave Ramsey or Caleb Hammer work (or don't). • How our individualism and social media lead to social isolation and affect our mental health. • If you're listening to this podcast, you're probably more financially secure than you think.

S., Goncalves, J., Nolasco, S., Pinto, F., Raught, B., Tassin, A.-M., Koll, F., Soares, H. Abstract: Centrosomes are organelles consisting of two structurally and functionally distinct centrioles, with the mother centriole having complex distal (DA) and subdistal appendages (SDA). Despite their importance, how appendages are assembled and maintained remains unclear. This study investigated human TBCCD1, a centrosomal protein essential for centrosome positioning, to uncover its localization and role at centrioles. We found that TBCCD1 localizes at both proximal and distal regions of the two centrioles, forming a complex structure spanning from SDA to DA and extending inside and outside the centriole lumen. TBCCD1 depletion caused centrosome mispositioning, which was partially rescued by taxol, and the loss of microtubules (MTs) anchored to centrosomes. TBCCD1 depletion also reduced levels of SDA proteins involved in MT anchoring such as Centriolin/CEP110, Ninein, and CEP170. Additionally, TBCCD1 was essential for the correct positioning of motile cilia basal bodies and associated structures in Paramecium. This study reveals that TBCCD1 is an evolutionarily conserved protein essential for centriole and basal body localization and appendage assembly and maintenance. A BioID screening also linked TBCCD1 to ciliopathy-associated protein networks. Copy rights belong to original authors. Visit the link for more info Podcast created by Paper Player, LLC
