This is the ability to extract energy from acetylene, and only a few species of bacteria (that we know of here on Earth) have it. Ben Gibbard. 07.20.122With eight albums in the books from his most notable project Death Cab For Cutie, one of the hardest working men in the indie rock scene is trying his luck with flying solo once again.The story for Ben Gibbard starts in Bremerton, WA where he developed an affinity for bands like The Pixies, Daniel Johnston, and Fugazi.  His first official band went by the name Penwheel, and only released one six song demo.A year later Ben was creating low-fi sounds of his own under the name All-Time Quarterback and released two EPs one, self-titled and the other titled The Envelope Sessions. That same year Gibbard change the name of his solo project to the Bonzo Dog Doo-Dah Band song titled Death Cab For Cutie.The first album of Death Cab’s was another solo project of Gibbard’s released on a cassette.  Ben thought the reception of this collection of songs merited a full band and recruited Chris Walla; who had helped on the original cassette; Nick Harmer, and Nathan Good to round out the four piece.  Throughout their career the group has been nominated for 16 various awards but have only walked away with a Woodie and the Larry Award for 2003’s Break Through Artist. Even though the band had been around for almost 6 years at that point.In 2001 Gibbard was introduced to electronic protégé Jimmy Tamborello, known as Dntel (dɪntɛl).  The two worked on a couple of track with Ben on the vocals. Dntel was very happy with how they turned out and a few weeks later Gibbard was receiving CD-R’s in the mail with some of Tramborello’s, beats and melodies on them. Gibbard was given liberty to create what he wanted from the tracks and decided that his long-time partner and fellow guitarist Chris Walla would be an asset to whatever this creation happened to be. Every two months or so Ben was receiving packages that he would edit, add then mail back. Ten month of this would manifest into what we now know as The Postal Service: A heavily electronic oriented group dripping with romantic lyrics, and sprinkled with guest vocals from Jen Wood and Rilo Kiley’s Jenny Lewis.Ben’s appreciation for the writing of Jack Kerouac would land him a in a studio with Son Volt’s Jay Farr where they would create a tribute album Kerouac’s famous novel Big Spur, titled One Fast Move or I’m Gone.Currently Death Cab is on tour for their 2012 release Codes and Keys, and once the band is finished with the year-long world-wide tour, Ben and his overactive creativity will be at it again with an album that will be titled Former Lives. Dysregulation of the hypothalamic-pituitary-adrenal (HPA) axis is a hallmark of complex and multifactorial psychiatric diseases such as anxiety and mood disorders. The 41-amino acid neuropeptide Corticotropin Releasing Hormone (CRH) is a major regulator of the mammalian stress response. Upon stressful stimuli, it binds to the Corticotropin Releasing Hormone Receptor 1 (CRHR1), a typical member of the class B GPCRs and a potential novel target for the therapeutic intervention in major depressive disorder. A precise understanding of the peptide-receptor interactions is an essential prerequisite towards the development of efficient CRHR1 specific antagonists. To chemically probe the molecular interaction of CRH with its cognate receptor, a high-throughput conjugation approach which mimics the natural activation mechanism for class B GPCRs was developed. Acetylene-tagged peptide libraries were synthesized and conjugated to high-affinity azide-modified carrier peptides using copper-catalyzed dipolar cycloaddition. The resulting conjugates reconstitute potent ligands and were tested in situ for modulation of the CRHR1 activity in a cell-based assay. This approach allows to (i) define the sequence motifs which are required for receptor activation or inhibition, (ii) identify the critical functional groups and investigate structure-activity-relationships, and (iii) develop novel optimized, highly potent peptide probes which are specific for the transmembrane domain of the receptor. The membrane recruitment by a high-affinity carrier peptide enhances the potency of tethered peptides and allows the initial testing of weak fragments that otherwise would be inactive. The biomimetic screening led to the discovery of transtressin, a highly modified and potent CRHR1 transmembrane domain-specific optimized agonist (EC50 = 4 nM). Beyond its intrinsic agonistic activity, transtressin is an essential tool for the pharmacological characterization of CRHR1 antagonists in competition assays.

Background: As differences in gas exchange between pulmonary arterial hypertension (PAH) and chronic thromboembolic pulmonary hypertension (CTEPH) have been demonstrated, we asked if cardiac output measurements determined by acetylene (C2H2) uptake significantly differed in these diseases when compared to the thermodilution technique. Method: Single-breath open-circuit C2H2 uptake, thermodilution, and cardiopulmonary exercise testing were performed in 72 PAH and 32 CTEPH patients. Results: In PAH patients the results for cardiac output obtained by the two methods showed an acceptable agreement with a mean difference of -0.16 L/min (95% CI -2.64 to 2.32 L/min). In contrast, the agreement was poorer in the CTEPH group with the difference being -0.56 L/min (95% CI -4.96 to 3.84 L/min). Functional dead space ventilation (44.5 +/- 1.6 vs. 32.2 +/- 1.4%, p < 0.001) and the mean arterial to end-tidal CO2 gradient (9.9 +/- 0.8 vs. 4.1 +/- 0.5 mmHg, p < 0.001) were significantly elevated among CTEPH patients. Conclusion: Cardiac output evaluation by the C2H2 technique should be interpreted with caution in CTEPH, as ventilation to perfusion mismatching might be more relevant than in PAH.
