The cannabinoids within: how marijuana hijacks an ancient signaling system in the brain | Ivan Soltesz

Given the widespread legalization of cannabis for medical and recreational uses, you'd think we'd have a better understanding of how it works. But ask a neuroscientist exactly how cannabinoid compounds like THC and CBD alter our perceptions or lead to potential medical benefits, and you'll soon learn just how little we know.We know that these molecules hijack an ancient signaling system in the brain called the "endocannabinoid" system (translation: the "cannabinoids within"). These somewhat exotic signaling molecules (made of fatty lipids and traveling "backwards" compared to other transmitters) have been deeply mysterious until recently, when new tools made it possible to visualize their activity directly in the brain. So what is the "day job" of the endocannabinoid system — and how does it connect to the dramatic highs that come with taking THC or the medical benefits of CBD? To unpack all this, we're talking this week with neuroscientist Ivan Soltesz, the James Doty Professor of Neurosurgery and Neuroscience at Stanford, and a leading expert on the endocannabinoid system.Learn MoreThe Soltesz Lab"Weeding out bad waves: towards selective cannabinoid circuit control in epilepsy" (Soltesz et al, Nature Reviews Neuroscience, 2015) "Keep off the grass? Click here to try it out!Welcome back to Bad Dads Film Review! Today, we're exploring the serene and beautifully poignant world of The Red Turtle (2016). This animated fantasy drama, a co-production between Studio Ghibli and Dutch-British animator Michaël Dudok de Wit, stands out as a unique cinematic experience that eschews dialogue to tell a deeply human story through stunning visuals and emotive sounds. The Red Turtle explores the life of a man who becomes stranded on a deserted island. The narrative unfolds through his attempts to escape the island, which are repeatedly thwarted by a mysterious red turtle. The film transitions into a deeper exploration of the man's life as he comes to terms with his isolation and eventually, finds companionship. The film is less about conventional storytelling and more about the cycle of life, relationships, and the human connection to nature. The man's interactions with the red turtle lead him through various phases of life, reflecting broader themes such as acceptance, companionship, and the inevitability of life's changes. Why It Stands OutNo Dialogue: The Red Turtle tells its story without a single line of dialogue, relying instead on the power of its visuals and a compelling score to convey emotion and narrative progression.Animation Style: The animation is simplistic yet profoundly expressive, blending realistic landscapes with a minimalist character design that captures the essence of each emotion and action without the need for words.Philosophical Depth: The film delves into existential themes through its symbolic use of the turtle and the natural surroundings, prompting audiences to reflect on life's transitions and the interconnectivity of all things. At its heart, The Red Turtle is about survival not just physically but emotionally and spiritually. It explores how companionship and acceptance of the natural flow of life can lead to profound fulfillment, even in isolation. For fans of animation and poignant storytelling, The Red Turtle is a must-watch for its ability to convey complex human emotions and narratives without words. It's a testament to the power of visual storytelling and a beautiful example of how art can transcend language to touch the human spirit. So, join us as we explore the quiet beauty and deep thematic currents of The Red Turtle. Whether you're a Studio Ghibli aficionado or a newcomer to animated art films, this film promises a meditative and visually captivating experience that lingers long after the screen goes dark.

Chris and Alex continue their discussions of Studio Ghibli for Episode 72 with a look at animated war feature Grave of the Fireflies (Isao Takahata, 1988), a film that was initially released as a double bill with partner My Neighbor Totoro (Hayao Miyazaki, 1988). Telling the story of teenage boy Seita and his younger sister Setsuko who, after fleeing the city of Kobe, must navigate the public horrors and personal traumas of World War II, Grave of the Fireflies offers a graphic and emotional portrait of conflict and society through the isolation and struggle experienced by the siblings. Joining the podcast this week to discuss the film’s potent political message is Alex Dudok de Wit, Associate Editor at Cartoon Brew, freelance journalist (including work for the BFI/Sight and Sound) and author of the upcoming BFI Film Classic on Grave of the Fireflies (London: Bloomsbury, 2021). Listen as the trio examine the historical, political and artistic contexts for the film, and its important place within the Ghibli canon; the cartoon short tradition and wartime propaganda in both the U.S. and Japan; pacing, rhetoric and the narrative framing of Grave of the Fireflies through fantasy and subjectivity; ghostliness, death and the afterlife, and what the pull between naturalism and fantasy means for the film’s tragedy; the interplay between the fantastical elements of Takahata’s film and its anti-war sentiment; the possible narrative judgment of Seita’s actions and protection of Setsuko; and how Grave of the Fireflies opens up questions about the many relationships between animation and politics, and what it means for popular animation to ‘do’ political enquiry.

L., Cai, R., Duan, J., Albarran, E., Ding, J., Lovinger, D. M., Li, B., Soltesz, I., Li, Y. Abstract: Endocannabinoids (eCBs) are retrograde lipid neuromodulators involved in many physiologically important processes. However, their release and dynamics in the brain remain largely unknown, in part due to lack of probes capable of reporting real-time eCBs with sufficient spatiotemporal resolution. Here, we developed a new G protein-coupled receptor Activation Based eCB sensor GRABeCB2.0 using the human CB1 cannabinoid receptor and a circular-permutated EGFP. GRABeCB2.0 exhibited proper cell membrane trafficking, ~seconds kinetics, high specificity and robust fluorescence response to eCBs at physiological concentrations. Using GRABeCB2.0, we detected evoked eCB dynamics in both cultured neurons and acute brain slices. Interestingly, we also observed spontaneous compartmental eCB transients that spread ~11 m in cultured neurons, suggesting locally-restricted eCB signaling. By expressing GRABeCB2.0 in vivo, we readily observed foot-shock elicited and running triggered eCB transients in mouse amygdala and hippocampus, respectively. Lastly, using GRABeCB2.0 in an epilepsy model, we observed a spreading eCB wave following a calcium wave in mouse hippocampus. In summary, GRABeCB2.0 is a powerful new probe to resolve eCB release and dynamics under both physiological and pathological conditions. Copy rights belong to original authors. Sam Clements is curating a fictional film festival. He'll accept almost anything, but the movie must not be longer than 90 minutes. This is the 90 Minutes Or Less Film Fest podcast. In episode 14, Sam is joined by Alex Dudok de Wit, freelance film journalist, Associate Editor for Cartoon Brew and curator for Short of the Week.  Alex has chosen the 1941 animated classic Dumbo (64 minutes), the fourth feature from Walt Disney Productions.  Sam and Alex discuss Dumbo's shoestring budget, the crow controversy, and why pink elephants were a safe choice in 1941.  We'll be back in a couple of weeks.   We have a new website! Dialogue-free, French-animated and developed over a number of years in collaboration with 'artistic producer' Isao Takahata, The Red Turtle is a unique entry in the Studio Ghibli canon, and Michaël shares with the Ghibliotechnicians his memories of creating what is, to date, the only non-Japanese Ghibli production.Follow Michael and Jake on Twitter:@MichaelJLeader – Michael@jakehcunningham – JakeProduced by Michael Leader, Jake Cunningham, Harold McShiel and Steph WattsEditing by Lister Rossell See acast.com/privacy for privacy and opt-out information.

50 years since his hit single 'Release Me', veteran singer Engelbert Humperdinck discusses his long career and his new album, which combines his greatest hits with two new tracks. We visit the London studio of Michaël Dudok de Wit, the award winning Dutch animator, to see him in action and talk about his feature film debut The Red Turtle. Produced by Japan's Studio Ghibli and created by a team of French animators lead by Dudok de Wit, The Red Turtle was the only 2-D animated feature to be Oscar nominated at this year's Academy Awards. Here's a rare thing, a new opera - in Welsh. Y Twr (The Tower) has it's world premier on Friday. Front Row drops in on a rehearsal to talk to the composer Guto Puw and librettist Gwyneth Glyn about their adaptation of one the most important Welsh plays of the 20th century. And Caryl Hughes and Gwion Thomas speak of their delight at having the opportunity, at last, of singing an opera in their mother tongue. Plus, following on from the success of F-rating films, seven events at the Bath Literary Festival use the F-rating in their brochure for the first time. Festival Director Alex Clark explains the thinking behind promoting cultural events that celebrate the female experience.

Studio Ghibli is revered all over the world for their films that feature lush animation and inventive stories with universal themes. As the studio's latest film, The Red Turtle, hits theatres, we decided to take a closer look at the companies' first international production. Directed by Michaël Dudok de Wit, and completely dialogue free, The Red Turtle follows a man who finds himself stranded on an island with a giant turtle. Diving into the film's sweeping look at nature, the human experience, mysticism, parenting, and all things in between, there is plenty to dissect in this thought-provoking film. We also take a moment to discuss our short film picks of the week: Tsunami and There Will Come Soft Rains.
