Given the widespread legalization of cannabis for medical and recreational uses, you'd think we'd have a better understanding of how it works. But ask a neuroscientist exactly how cannabinoid compounds like THC and CBD alter our perceptions or lead to potential medical benefits, and you'll soon learn just how little we know.We know that these molecules hijack an ancient signaling system in the brain called the "endocannabinoid" system (translation: the "cannabinoids within"). These somewhat exotic signaling molecules (made of fatty lipids and traveling "backwards" compared to other transmitters) have been deeply mysterious until recently, when new tools made it possible to visualize their activity directly in the brain.So what is the "day job" of the endocannabinoid system — and how does it connect to the dramatic highs that come with taking THC or the medical benefits of CBD? Auch Hayao Miyazaki kann sich ab und zu noch für die Arbeiten anderer Filmemacher begeistern. So bot er dem oscarprämierten Kurzfilm-Regisseur Michael Dudok de Witt an, dessen ersten abendfüllenden Spielfilm in Zusammenarbeit mit dem 'Studio Ghibli' zu produzieren, womit erstmalig einem nicht aus Japan stammendem Regisseur diese Ehre zuteil wurde. Allerdings wurden noch weitere Studios an Bord geholt, wie 'Wild Bunch' und 'Why Not Productions', die aus Europa kamen, und weshalb das gemeinsam produzierte Werk mit dem Titel 'Die rote Schildkröte' auch offiziell nicht wirklich als Anime-Film zählt. Unterstützt wurde Dudok de Witt vorrangig von Isao Takahata, der den Animations-Check übernahm und was Takahatas letzte Mitarbeit an einem Film gewesen ist, bevor er 2018 verstarb. 'Die rote Schildkröte' kam 2016 ins Kino und ist ein Film, der komplett ohne Dialoge auskommt und vorrangig durch seine Atmosphäre und eine fast schon märchenhafte Erzählung auskommt, in der ein Schiffsbrüchiger, der sich auf eine einsame Insel retten kann, mit seiner Situation arrangiert und lernt, im Einklang mit der Natur zu leben. Ob 'Die rote Schildkröte' nun wirklich noch als Ghibli-Film zu sehen ist, darüber gibt es viele geteilte Meinungen. Chris and Alex continue their discussions of Studio Ghibli for Episode 72 with a look at animated war feature Grave of the Fireflies (Isao Takahata, 1988), a film that was initially released as a double bill with partner My Neighbor Totoro (Hayao Miyazaki, 1988). Telling the story of teenage boy Seita and his younger sister Setsuko who, after fleeing the city of Kobe, must navigate the public horrors and personal traumas of World War II, Grave of the Fireflies offers a graphic and emotional portrait of conflict and society through the isolation and struggle experienced by the siblings. Joining the podcast this week to discuss the film’s potent political message is Alex Dudok de Wit, Associate Editor at Cartoon Brew, freelance journalist (including work for the BFI/Sight and Sound) and author of the upcoming BFI Film Classic on Grave of the Fireflies (London: Bloomsbury, 2021). Listen as the trio examine the historical, political and artistic contexts for the film, and its important place within the Ghibli canon; the cartoon short tradition and wartime propaganda in both the U.S. and Japan; pacing, rhetoric and the narrative framing of Grave of the Fireflies through fantasy and subjectivity; ghostliness, death and the afterlife, and what the pull between naturalism and fantasy means for the film’s tragedy; the interplay between the fantastical elements of Takahata’s film and its anti-war sentiment; the possible narrative judgment of Seita’s actions and protection of Setsuko; and how Grave of the Fireflies opens up questions about the many relationships between animation and politics, and what it means for popular animation to ‘do’ political enquiry.

Abstract: Endocannabinoids (eCBs) are retrograde lipid neuromodulators involved in many physiologically important processes. However, their release and dynamics in the brain remain largely unknown, in part due to lack of probes capable of reporting real-time eCBs with sufficient spatiotemporal resolution. Here, we developed a new G protein-coupled receptor Activation Based eCB sensor GRABeCB2.0 using the human CB1 cannabinoid receptor and a circular-permutated EGFP. GRABeCB2.0 exhibited proper cell membrane trafficking, ~seconds kinetics, high specificity and robust fluorescence response to eCBs at physiological concentrations. Using GRABeCB2.0, we detected evoked eCB dynamics in both cultured neurons and acute brain slices. Interestingly, we also observed spontaneous compartmental eCB transients that spread ~11 m in cultured neurons, suggesting locally-restricted eCB signaling. By expressing GRABeCB2.0 in vivo, we readily observed foot-shock elicited and running triggered eCB transients in mouse amygdala and hippocampus, respectively. Lastly, using GRABeCB2.0 in an epilepsy model, we observed a spreading eCB wave following a calcium wave in mouse hippocampus. In summary, GRABeCB2.0 is a powerful new probe to resolve eCB release and dynamics under both physiological and pathological conditions. Copy rights belong to original authors.
