In today's episode, supported by Revolution Medicines, we spoke with Kathryn C. Arbour, MD, a thoracic medical oncologist at Memorial Sloan Kettering Cancer Center in New York, New York, about 2 important abstracts presented at the 2024 AACR Annual Meeting that explore novel RAS-targeted approaches in non–small cell lung cancer (NSCLC). Our discussion focused on early clinical findings with zoldonrasib (RMC-9805) and daraxonrasib (RMC-6236), both of which are RAS(ON) inhibitors under investigation for the treatment of patients with RAS-mutant NSCLC. Zoldonrasib, a KRAS G12D-selective tri-complex inhibitor, was evaluated in a phase 1 trial (NCT06040541) in patients with previously treated, advanced KRAS G12D–mutated solid tumors, including NSCLC. Daraxonrasib, a multi-selective RAS(ON) inhibitor, was highlighted in another phase 1 trial (NCT05379985) in patients with advanced RAS-mutant tumors, including previously treated NSCLC; notably, this AACR presentation focused on the association between early on-treatment circulating tumor DNA level reduction and clinical response with the agent. In this episode, Dr Arbour shared insights into the mechanisms of action behind these therapies, their respective clinical trial designs, and the potential implications that early data with the agents may have for the evolving RAS-mutant NSCLC treatment paradigm.

My guest today is from Ontario, Canada and her first season was in Club Med Turks & Caicos during the 1996/1997 season as a Boutique G.O. Before working for Club Med, she was a G.M and went to Club Med Playa Blanca in 1989 and then went to Turks each year from 1989 until she was hired in 1996. So, you could say she knew that village very well! She worked for CM from 1996 to 2000 in such villages as Punta Cana, Sonora Bay, Eleuthera, Cancun, and Coral Beach in Israel. She worked her way up to Chief of Boutique eventually and worked for CDVs such as Kevin Batt, Hammer, and Pierre Letellier Gagnon, to name a few. Please help me welcome, just 15 minutes from Niagara Falls, Jeanette Arbour!! Fun Fact: Jeanette might be the only EXGO I know of to have had their first season in Club Med Turks & Caicos and her last season in Club Med Coral Beach…the same as yours truly! Jeanette first heard about Club Med from a high school friend of hers who showed her the brochure. During her last vacation as a G.M. in Turks, she approached the CDV Kevin Batt and asked for a job and Jeanette was placed in the Boutique. Jeanette shares a Christmas story from her season in Eleuthera where a plane was chartered to Nassau so they could buy 150 gifts for the kids of the village during Christmas week. Another Christmas memory comes from Jeanette's season in Cancun in 1999/2000, where Hammer somehow found pine trees in Cancun at Christmastime! Another story Jeanette shares is from her season in Punta Cana in 1998 where the village was bought out by hairdressers. She spent the entire week being an “updo model” because she had the longest hair in the village and did not even get her hair cut that week! We share fond memories of our time spent in Israel and the name “Mark Vanderhoef” came up a lot during this interview! Season Two of Islanders Awards Winners is finally here. It's taken far too long to get to this point, but I'm proud of how these episodes turned out and I'm excited to finally release them to the world. Thank you all in advance for listening. The story of Al Arbour's one NHL Coach of the Year season begins and ends with devastating losses. In between was a franchise-record winning streak, several of the best individual seasons in team history, and the often ruthless pursuit of perfection. Arbour was a complex man - a hockey innovator who wore “dark suits and white shirts and ties that look like they were given as Christmas presents” and a father figure who pushed his players relentlessly, sometimes to the point of exasperation. But his drive was to win the Stanley Cup under any circumstance. And any stumble, no matter how minor, was unacceptable. Arbour was not afraid to butt heads with his best and most loyal players and they all understood it was all done in service of making them the best the could be. Arbour's use of video review, notes on opposing teams' tactics and installation of assistant coaches were all ahead of their time. The leaning on his wife Claire to not only record out-of-town games but to find out more about players' family lives in order to better understand and help them when he could, was critical and should not be overlooked. Man, what I would give to read those notebooks he kept on other teams. I can't even imagine the sheer amount of hockey knowledge they contained. Maybe they'll be released some day... On Inside Geneva this week: part three of our series marking the 75th anniversary of the Universal Declaration of Human Rights.Host Imogen Foulkes talks to Louise Arbour, who served as UN Human Rights Commissioner from 2004 to 2008. She arrived in Geneva with a formidable track record.As a prosecutor for the former Yugoslavia, she had indicted Slobodan Milosevic for war crimes. In Rwanda, she secured convictions of rape as crimes against humanity."The work I did both with the tribunal for the former Yugoslavia and for Rwanda were if anything a vindication for me of the significance of law, of the rule of law, as an organising principle in modern society," explains Arbour. Leading the UN's human rights work was a new challenge. "These were very challenging times. 2004, you know, this was in the backyard of 9/11. It was, a new, dangerous, unknown world was starting to unfold with a lot of uncertainties, including on the human rights front."New strategies were needed. "When you arrive in the role of high commissioner for human rights, I think that's part of the dilemma; how do you use your voice? Because I think to be the megaphone for the denunciation of injustices at some point becomes counterproductive, because it just illuminates how impotent the system is. It's like you scream in the wilderness," she said. That's why this dedicated lawyer still tells us to follow the laws, treaties, and conventions we have. "If you came from another planet and you just looked at the human rights framework; the universal declaration of human rights, all the treaties, the conventions, the work of the treaty bodies, you'd think you'd arrived in heaven. In between, interim coach Jimmy Roberts got nine games behind St. Louis' bench, going 3-3-3.Between them, Keenan (672) and Quenneville (969) have won 1,641 games as NHL coaches. So, does Roberts hold the very niche record of Interim Coach Between The Pair Of Coaches With the Most Wins?This isn't super hard to figure out. Quenneville is second on the NHL's all-time wins list, behind only Scotty Bowman. Keenan is 15th. You might figure this out a different way, and that's cool, but here's my process: go through the top coaches until you reach a place where you can't get a combo adding up to more than 1,641, and Roberts has the record — or someone else has a bigger number before you get to that point. For clarity's sake, we'll call an interim coach one who does not finish out the season.Bowman himself was never replaced by, nor replaced, an interim coach as such. Bowman's career reads funny because he coached the Sabres in 1979-80, then for 35 games in 1981-82, then for the next three full seasons, plus 37 games in 1985-86 and 12 more in 1986-87 — because Bowman was also GM in Buffalo and stepping behind the bench himself sometimes.Quenneville got the ax in St. Louis in 2004, replaced by Mike Kitchen on a full-time basis, so no worries there. Same with Jeremy Colliton, while Andrew Brunette finished out last season in Florida before the Panthers turned to Paul Maurice.Barry Trotz (914 wins): Never fired in-season.Ken Hitchcock (849), Lindy Ruff (827): Part of in-season coaching changes, but never with a team that had a true interim in the mix.Paul Maurice (811): The 2004 Hurricanes — hey, that's shit-for-brains Eric Staal's rookie year, and he was teammates with Ron Francis and Artūrs Irbe, which is excellent to keep in mind for your Six Degrees Of… hockey games! — went straight to Peter Laviolette, who has 751 career wins and counting, and with Maurice also being active, they're at 1,562 wins and counting, which is… not a record for combined career wins by the coaches of one team in a single season, because you've not only got those 1,641 career wins for Keenan and Quenneville with the 1996-97 Blues, but also Roberts' 50 (three with those Blues, 21 for the Bowman-run 1981-82 Sabres, and 26 in his one full season as coach, with the 1991-92 Hartford Whalers). Stay tuned?Al Arbour (782): One of those wins was actually as an interim coach, when Arbour got what amounted to a testimonial match with the Islanders. He's officially listed that season as the coach between Ted Nolan and Ted Nolan again.And it wouldn't matter if Al Arbour had coached between Al Arbour and Al Arbour, because 782+782=1,564, and that's less than 1,641. No two coaches on this list can combine to get there, and while Darryl Sutter and John Tortorella (sort of, but congrats on win number 700, man, hope you're real proud of Staalsie today, too) remain active with 700-plus wins, Roberts, who died in 2015, remains the voice between the two biggest voices, as far as NHL history is concerned.It's a garage league full of spineless dweebs, but the history is interesting. Open theism is a provocative model of God that has captured the attention of various philosophers and theologians from different religious traditions. The view has garnered different criticisms, such as the recent book edited by Ben Arbour called, Philosophical Essays Against Open Theism.
