Abstract: Myofibroblasts are responsible for scarring and organ stiffness during fibrosis. The scar propagates mechanical signals inducing a radical transformation in myofibroblast cell state linked to an increasingly pro-fibrotic phenotype. Here, we have discovered mechanical stress from progressive scarring induces nuclear softening and de-repression of heterochromatin. The parallel loss of H3K9Me3 enables a permissive state for distinct chromatin accessibility and profibrotic gene regulation. By integrating chromatin accessibility profiles (ATAC sequencing) we provide insight into the transcription network and open chromatin landscape underlying the switch in profibrotic myofibroblast states, emphasizing mechanoadaptive pathways linked to PAK1 as key drivers. Through genetic manipulation in liver and lung fibrosis, uncoupling PAK1-dependant signaling impaired the mechanoadaptive response in vitro and dramatically improved fibrosis in vivo. Moreover, we provide human validation for mechanisms underpinning PAK1 mediated mechanotransduction in liver and lung fibrosis. Collectively, these observations provide new insight into the nuclear mechanics driving the profibrotic chromatin landscape in fibrosis, highlighting actomyosin-dependent mechanisms linked to chromatin organisation as urgently needed therapeutic targets in fibrosis. For years, I despised her. I hated being in any room she was in. I hated it even more when she was in the rooms I wasn't in, but wanted nothing more than to be in. He has that innate ability to recognise his spiritual and cognitive intelligence. Spending time with him can only grow your sensory awareness and your emotional strength.  Reg remembers a pivotal moment in his life that catapulted him into the journey he is still on and will remain on for the rest of his life.  At that time there was a vibration, a feeling within him that he won't forget. That vibration, as he calls it, has been his check-in since, as to whether something is right or wrong for him to furthermore invest his time and emotion. We may not all have a pivotal moment that we can bring to mind but most of us can relate to this vibration, I guess we may refer to it as our intuition. Going on what Reg says, we should pay more attention to that and trust it much more than we do. Moments in our lives, whether we remember them or not, are definitely pivotal. Through our perception of an experience, we then choose to go left or right. Some of this will be genetic influence, some our subjective experience and of course our living influences but either way they shape our destiny, that to me says there are many pivotal moments! Knowing these exist will affect our degree of impact. We should pay close attention to how we are going to choose to use them to create our success, with a clear vision of what that is to us. Reg's view of success is valuable, being aware that our view of our ideals of success will evolve throughout our life - when in his 20's, for Reg it was material whilst in his 40's, it's been about contribution and connection.  My main take from my time with Reg was clarity of my purpose-driven intention, with the flexibility for my vehicle to change. The vehicle isn't the piece that gives what we're doing legs, it's something deeper than that! Professionally, he has spent 19 years with DBD. Ray enjoys spending his days motorcycling, socializing with friends and family, and accompanying his children to their hockey and lacrosse games.   On the fateful day of May 8th, 2016; mother’s day, his and his family’s lives were changed forever. His wife suffered a major heart attack that developed into heart failure, leading to many surgeries and a wait for a new heart. With her rare blood-type and the many ups and downs that come with waiting on any kind of transplant, their journey was not an easy one.   In this episode, Ray talks about the challenges they faced together as a family and how he helped support his wife, children, and himself in trying times.
