J., Pat, F., Ottenheimer, D. J., Janak, P. H. Abstract: The ability to evaluate and select a preferred option among a variety of available offers is an essential aspect of goal-directed behavior. Dysregulation of this valuation process is characteristic of alcohol use disorder, with the central amygdala being implicated in persistent alcohol pursuit. However, the mechanism by which the central amygdala encodes and promotes the motivation to seek and consume alcohol remains unclear. We recorded single-unit activity in male Long-Evans rats as they consumed 10% ethanol or 14.2% sucrose. We observed significant activity at the time of approach to alcohol or sucrose, as well as lick-entrained activity during the ongoing consumption of both alcohol and sucrose. We then evaluated the ability of central amygdala optogenetic manipulation time-locked to consumption to alter ongoing intake of alcohol or sucrose, a preferred non-drug reward. In closed two-choice scenarios where rats could drink only sucrose, alcohol, or quinine-adulterated alcohol with or without central amygdala stimulation, rats drank more of stimulation-paired options. Microstructural analysis of licking patterns suggests these effects were mediated by changes in motivation, not palatability. Given a choice among different options, central amygdala stimulation enhanced consumption if the stimulation was associated with the preferred reward while closed-loop inhibition only decreased consumption if the options were equally valued. However, optogenetic stimulation during consumption of the less-preferred option, alcohol, was unable to enhance overall alcohol intake while sucrose was available. Collectively, these findings indicate that the central amygdala processes the motivational value of available offers to promote pursuit of the most preferred available option. Abstract: Separable striatal circuits have unique functions in Pavlovian and instrumental behaviors but how these roles relate to performance of sequences of actions with and without associated cues is less clear. Here we tested whether dopamine release and neural activity more generally in three striatal subdomains are necessary for performance of an action chain leading to reward delivery. Male and female Long-Evans rats were trained to press a series of three spatially-distinct levers to receive reward. We assessed the contribution of neural activity or dopamine release within each striatal subdomain when progression through the action sequence was explicitly cued and in the absence of cues. Behavior in both task variations was substantially impacted following microinfusion of the dopamine antagonist, flupenthixol, into nucleus accumbens core (NAc) or dorsomedial striatum (DMS), with impairments in sequence timing and a strong impact on motivation after NAc flupenthixol. In contrast, after pharmacological inactivation to suppress overall activity, there was minimal impact on motivation, except within the uncued task after DMS inactivation. Inactivation of both NAc and DMS impaired sequence timing and led to sequence errors in the uncued, but not cued task. There was virtually no impact of dopamine antagonism or reversible inactivation of dorsolateral striatum on either cued or uncued action sequence completion. These results highlight an essential contribution of NAc and DMS dopamine systems in motivational and performance aspects of chains of actions, whether cued or internally generated, as well as the impact of intact NAc and DMS function for correct sequence performance. Abstract: Impulse control disorders (ICDs), a wide spectrum of maladaptive behavior which includes pathological gambling, hypersexuality and compulsive buying, have been recently suggested to be triggered or aggravated by treatments with dopamine D2/3 receptor agonists, such as pramipexole (PPX). Despite evidence showing that impulsivity is associated with functional alterations in corticostriatal networks, the neural basis of the exacerbation of impulsivity by PPX has not been elucidated. Here we used a hotspot analysis to assess the functional recruitment of several corticostriatal structures by PPX in male rats identified as highly impulsive (HI) or with low levels of impulsivity (LI) in the 5-choice serial reaction time task (5-CSRTT). PPX dramatically reduced impulsivity in HI rats. Assessment of the expression pattern of the two immediate early genes C-fos and Zif268 by in situ hybridization subsequently revealed that PPX resulted in a decrease in Zif268 mRNA levels in different striatal regions of both LI and HI rats accompanied by a high impulsivity specific reduction of Zif268 mRNA levels in the prelimbic and cingulate cortices. PPX also decreased C-fos mRNA levels in all striatal regions of LI rats, but only in the dorsolateral striatum and nucleus accumbens core (NAc Core) of HI rats. Structural equation modelling further suggested that the anti-impulsive effect of PPX was mainly attributable to the specific downregulation of Zif268 mRNA in the NAc Core. Altogether, our results show that PPX restores impulse control in highly impulsive rats by modulation of limbic frontostriatal circuits. Copy rights belong to original authors. Kiwis have set up a Givealittle page to raise money for Janak Patel's family following his tragic death. After Auckland dairy worker Janak Patel was fatally stabbed in Sandringham following a man who stole cash from Rose Cottage Superette in November, over $100,000 has been donated to the family. Dairy and Business Owners Group chair Sunny Kaushal says that the family is grateful to everyone who donated to the Givealittle fund, and the public support draws attention to crime being out of control. "This needs to be dealt with with urgency and force, and it starts with admitting we have a problem. The crime is not some academic issue, it impacts real people. A nationwide protest is taking place on Monday after the fatal stabbing of dairy worker Janak Patel during a robbery of the Rose Cottage Superette in Auckland's Sandringham last Wednesday. A 34-year-old man has now been arrested and charged with Patel's murder, and another two have been charged with robbery. Dairy and Business Owners Group (DBOG) chairperson Sunny Kaushal said the country is experiencing a "crime emergency" and wants urgent action from the government. Sixth in the line of king Nimi, Devrat was given the divine bow of Shivaji for safekeeping. The bow was kept in the palace of Mithila with honor and was worshiped by the Janak clan. Once during the reign of Janak Seeradhwaj, the Kingdom of Videh suffered a bad drought. On his kulguru rishi Shatananda's suggestion King Janak along with his wife Sunayana ploughed the fields of Mithila. Suddenly the plough hit something inside the ground and could not move. Upon digging they discovered a little girl inside a gold box. The king adopted this little girl and named her Sita. The bow which would normally take five thousand people to move, was once lifted by devi Sita without any assistance. This prompted Rajarshi Janak to decide that he would marry Sita to the one who can string the divine bow of Shiva. He organized the swayamvar with this condition, however every potential suitor attending the swayamvar failed to fulfil the condition. All the Kings and Princes could not digest this humiliation and decided to attack Mithila with their joint army. Story of Ashtavakra is narrated in great epics Ramayan and Mahabharat. Astavakra is the son of Rishi Kahoda and Sujatha Daughter of Rishi Aruni. He was born with physical deformities and hence got the name Ashtavakra. When he was very young his father went to Raja Janak's place to participate in a debate against renowned scholar Vandin/Vanin, son of water God Varun. His father lost the debate and was immersed in water as condition of the debate. When Ashtvakra was ten years old, his mother told him about his father's fate and the kid went to meet Raja Janak. Janak first tested the boy and found him very knowledgeable. He then went on with the debate against Vandin and won the argument. After Ashtavakra won the debate, his father and other drowned scholars were released by Varun dev. He is credited with writing of Ashtavakra Gita, which mostly talks about nature of God and metaphysical nature of existence. There is an interesting story behind Ashtvakra's physical deformities. When he was inside his mother's womb, his father was teaching him verses from Vedas and boy would correct him from inside the womb. This irked his father a lot and once out of irritation he cursed the boy to be born with deformities. After Ashtavakra rescues his father, his father is very impressed with his young son and his knowledge and accepts his mistake in cursing him. On their way back from Janak palace, he asks Ashtvakra to take a dip in a divine lake. After taking the dip in the lake, he is cured of his deformities and becomes normal again. Two questions have been answered by Om Swami in this discourse. The first question asked was- Ma Janaki has been given different versions in different languages and in scriptures like Ram charit Manas and in some other scriptures too. What is the real truth about the birth of Goddess Sita? Om Swami says that Ma Janaki was king Janak's daughter. He further says that there are two ways of seeing things. Either we believe that the scriptures are true or we think that it is fictional. In both Ramayana and Mahabharata the person who played an important role was found in a box. In Ramayana Janaki Ma was found in a box while king Janak was tilling the land. In Mahabharata Karna was floated away by his mother in a box. Without both of them the battle that took place may not have happened he says. The Ramayana and the Mahabharata were written in metre form so that memorising was easy. But still passing it on in the oral form could have resulted in minor errors. So it is difficult to say if Ma Janaki was found in a box in the ground, but nevertheless Ma Janaki was none other than the universal mother he says. Om Swami says that for him everyone who was connected to Shri Ram in any way was only shri Ram - the people who were killed by him and those who were with him. All of them are worthy of worship and this is the sentiment that gives him great happiness. The second question asked was- If Om Swami could shed some light on the term biological father Vs spiritual father. Om Swami says that just like a biological father will want their children to do well and do good, similarly a spiritual father wishes the same. Though the sentiments are the same, it's a different kind of relationship. The difference is that a spiritual father takes on a disciple's karma by seeing if it can be distributed over lifetimes or can be taken care of at one go so that the person attains liberation.
