In this episode we feature the latest research by Yasufumi Takahashi at the Kanazawa University NanoLSI.The research described in this podcast was published in Analytical Chemistry in August 2023 Kanazawa University NanoLSI websitehttps://nanolsi.kanazawa-u.ac.jp/en/Researchers define a nanopipette fabrication protocol for high resolution cell imagingResearchers at Kanazawa University report in Analytical Chemistry how to produce nanopipettes that reliably provide nanoscale resolution scanning ion conductance microscopy images of living cells.A nanoscale view of living cells can provide valuable insights into cell structure and function. Over the years, various microscopy techniques have been enrolled to obtain a window into biological specimens at the nanoscale but all with their limitations and challenges. Although scanning ion conductance microscopy has demonstrated the capability to image living biological samples in solution with nanoscale resolution, it has been hampered by challenges in reliably producing nanopipettes with the optimum geometry for the job. Now researchers led by Yasufumi Takahashi at Kanazawa University's Nano LSI and Nagoya University have devised a protocol for reproducibly fabricating nanopipettes with the preferred geometry for high quality imaging. So what is scanning ion conductance microscopy and what kind of nanopipette does it need?Scanning ion conductance microscopy uses a nanopipette to control the distance between nanopipette and sample using an ion current as feedback signal. The shape of the nanopipette significantly influences the performance of the device. For instance, a wide aperture limits the possible resolution, a long shunt can lead to rectification effects that warp the ion current measurements, and if the glass of the nanopipette is too thick it can deform the sample before the proximity of the aperture has reached the point needed for constant ion current topographical mapping. As a result, the ideal nanopipette has a short shunt, small aperture and thin glass walls.The standard procedure for fabricating the nanopipette is to pull a capillary tube with a laser puller that heats the capillary tube it is manipulating. The capillary then narrows where it lengthens until it is finally drawn into two separate pieces. Although quartz can allow a little more control in the process of drawing the capillary tube into shape it is hydrophobic, which raises complications in actually filling the nanopipette with the aqueous solution needed for the ion current. For this reason, the researchers developed a protocol by which they could draw nanopipettes from borosilicate glass capillaries with the required control and reproducibility.Takahashi and his collaborators noted that ideally the starting capillary should have thick walls and a narrow inner diameter, however it is not easy to obtain capillary tubes to these requirements from commercial suppliers. Instead, they preheat the capillary for 5 s without pulling it, which causes the glass walls to the thicken and reduces the inner diameter. They also optimized the parameters for pulling the tube, such as the velocity.So did it work? Apparently soThe researchers demonstrated the performance of the nanopipettes they produced by imaging a cell undergoing a type of endocytosis, where it engulfs and absorbs some external material. Abstract: Background: The endo-lysosomal phospholipid, bis(monoacylglycerol)phosphate (BMP), is aberrantly high in the urine of Parkinson's patients with mutations in genes encoding leucine-rich repeat kinase 2 (LRRK2) and glucocerebrosidase (GCase). Because BMP resides on and regulates the biogenesis of endo-lysosomal intralumenal membranes (exosomes when released), we hypothesized that elevated urinary BMP may be driven by increased exocytosis of BMP-enriched exosomes. Objective: Our aim was to explore the contribution of LRRK2 and GCase activities in the regulation of BMP metabolism and release. Methods: Microscopy and biochemical assays were used to analyze antibody-accessible BMP and exosome release in wild type (WT) and R1441G LRRK2-expressing mouse embryonic fibroblast cells. Lipidomics analysis was conducted to measure BMP and lipid content in cells and isolated exosomes. In these experiments, we tested the effects of LRRK2 and GCase inhibitors, MLi-2 and conduritol {beta}-epoxide respectively, to assess regulation of BMP by LRRK2 and GCase. Results: Alterations in antibody-accessible BMP and endo-lysosome morphology were detected in R1441G LRRK2 cells. Lipidomics analysis revealed increased BMP content in mutant LRRK2 MEFs compared to WT MEFs. Inhibition of LRRK2 partially restored cellular and exosome-associated BMP levels; abrogation of GCase activity had the opposite effect. Metabolic labeling experiments confirmed that BMP synthesis is not influenced by LRRK2 or GCase activities. Pharmacological modulation of exosome release further confirmed exosome-mediated BMP exocytosis. Conclusions: LRRK2 regulates BMP in cells and its release in exosomes, which can be further modulated by GCase activity. Abstract: Secretory cells in glands and the nervous system frequently package and store proteins destined for regulated secretion in dense-core granules (DCGs), which disperse when released from the cell surface. Despite the relevance of this dynamic process to diseases such as diabetes and human neurodegenerative disorders, our mechanistic understanding is relatively limited, because of the lack of good cell models to follow the nanoscale events involved. Here, we employ the prostate-like secondary cells (SCs) of the Drosophila male accessory gland to dissect the cell biology and genetics of DCG biogenesis. These cells contain unusually enlarged DCGs, which are assembled in compartments that also form secreted nanovesicles called exosomes. We demonstrate that known conserved regulators of DCG biogenesis, including the small G-protein Arf1 and the coatomer complex AP-1, play key roles in making SC DCGs. Using real-time imaging, we find that the aggregation events driving DCG biogenesis are accompanied by a change in the membrane associated small Rab GTPases which are major regulators of membrane and protein trafficking in the secretory and endosomal systems. Indeed, a transition from trans-Golgi Rab6 to recycling endosomal protein Rab11, which requires conserved DCG regulators like AP-1, is essential for DCG and exosome biogenesis. Our data allow us to develop a model for DCG biogenesis that brings together several previously disparate observations concerning this process and highlights the importance of communication between the secretory and endosomal systems in controlling regulated secretion. Abstract: Dysregulated extracellular vesicle (EV) release has been implicated in various pathologies, including cancer, neurodegenerative disease and osteoarthritis. Despite clear therapeutic potential, drug screening for EV release modulators has yielded limited success due to the lack of a sensitive and scalable EV read-out system. Here, we employed CRISPR-Cas9 to engineer HEK293 cells expressing HA-NanoLuciferase-(NL)-tagged endogenous CD63. We found that under basal culture conditions, CD63-containing EVs are released via a mechanism that is independent of the exocytic SNARE protein SNAP23, presumably by direct budding from the plasma membrane. Endo-lysosome inhibition by chemical or genetic perturbation of vATPase strongly increased SNAP23 and nSmase2-dependent exosome secretion from intracellular compartments. Proteomic analysis revealed these exosomes are enriched for early- and late endosomal markers, but also for autophagosomal proteins. This suggests that a proportion of these exosomes originate from amphisomes, although chemical inhibition of canonical autophagy did not affect exosome secretion upon lysosome inhibition. Using a broad-spectrum kinase inhibitor screen, we identified and subsequently validated the lipid kinase PI4KIII{beta} as a critical mediator of exosome secretion and amphisome-mediated secretory autophagy, upon lysosome inhibition. We conclude that tagging of endogenous CD63 with NanoLuciferase represents a sensitive, scalable reporter strategy that enables identification of (druggable) modulators of EV biogenesis and release under physiological and pathological conditions. Abstract: Developing astroglia play important roles in regulating synaptogenesis through secreted and contact signals. Whether they regulate postnatal axon growth is unknown. By selectively isolating exosomes using size-exclusion chromatography (SEC) and employing cell-type specific exosome reporter mice, our current results define a secreted astroglial exosome pathway that can spread long-range in vivo and stimulate axon growth of cortical pyramidal neurons. Subsequent biochemical and genetic studies found that surface expression of glial HepaCAM protein essentially and sufficiently mediates the axon-stimulating effect of astroglial exosomes. Interestingly, apolipoprotein E (ApoE), a major astroglia-secreted cholesterol carrier to promote synaptogenesis, strongly inhibits the stimulatory effect of astroglial exosomes on axon growth. Developmental ApoE deficiency also significantly reduces spine density of cortical pyramidal neurons. Together, our study suggests a surface contact mechanism of astroglial exosomes in regulating axon growth and its antagonization by ApoE, which collectively coordinates early postnatal pyramidal neuronal axon growth and dendritic spine formation. Putting Manufacturing First: Codiak Moves Swiftly into Clinical Trials with Exosome-based Treatments  Sriram Sathyanarayanan, Codiak's CSO, and Linda Bain, their CFO, share how the company moved into clinical trials in under 6 years.   Exosomes, extremely small vesicles shed by all cell types, promise to become a viable delivery system for treatments of many diseases. But until recently, manufacturing them at a commercially viable scale has been unfeasible. That is why in 2015 the company Codiak took a two-pronged approach to developing exosome-based treatments: prioritizing both the engineering and manufacturing tracks from the outset. A mere six years after the company was launched, this approach has proven effective, with two promising studies in the clinic for tumor treatments, while most other developers are still at the starting blocks. The exosome field today holds the kind of promise that antibody and protein-based therapeutics did in the 1980s—the potential to improve both treatments and patient well-being is great. They are roughly between 30 and 120 nanometers large and originate when endosomes, or intercellular vesicles, are released into the blood, milk or tissue. Exosomes then become messengers and surrogates for the original cell. Their surface markers represent a location code and spread through the extracellular space in the body to communicate with other cells and deliver packages. Commercial exosome manufacturing is the scaling-up process of moving from small-batch exosome production that uses ultracentrifuges to large-scale production that in many ways resembles the processes already used to manufacture antibody and protein-based therapies.   The exosome's lumen is the interior volume of the exosome where, through biological engineering, the therapeutic molecule can be placed. The molecule can also be on the surface of the exosome, allowing for two alternative payload capacities, depending on the target. Immunosafety and immunotoxicity refer to how potentially safe or toxic the immune system's reaction to a molecule may be. Since exosomes already have a history of low immunotoxicity – think of blood transfers – their immunosafety is already proven to be very high.

Dr. Massey asked her, “What is our first home? The womb.” He explains how real estate and the womb tie together, using Recall Healing. “What happened at your birth?” is an important question to ask. “My burden in life gets lighter.” Saying it can make diseases go away. “Almost always there’s a family story,” regarding all diseases, including thyroid diseases. “Biology is like the accountant. It keeps track of everything,” says Dr. Massey. Patrick asks for more information about the thyroid and it’s relationship to time. Dr. Massey replies in detail. Patrick’s question leads to a discussion about synchronicity and the nature of “time.” Dr. Massey’s grandparents barely survived the “Great Miami hurricane” in 1926. His father became a weatherman. Diseases have two main causes — birth or family constellations. This coronavirus is not the problem. “Each person will find their part in it,” Dr. Massey explains. “My cells are actually ejaculating exosomes. Somehow the word ‘virus’ got put on these little particles of connection.” Virus has meant “poison” for 500 years, “Exosome” is a much better word. Dr. Massey didn’t wear a mask in the operating/recovery rooms, and he never caught anything. “When a doctor says, ‘Oh, my, there’s two small spots on the lung,’ we forget the several trillion that are functioning perfectly well.” Patrick discusses his sleeplessness. Exosomes, what are they and why do they exist? Well, if you haven’t heard what exosomes are, then you are not alone. Exosomes are extracellular vesicles first discovered 30 years ago. It was since implicated in cell-cell communication and the transmission of disease states. It was also explored as a means of drug discovery.   So how are these extracellular vesicles important? Exosome therapies are now being explored in anti-cancer clinical trials, and recent reports claim taxol-filled exosomes can be used to treat cancers in mice at 50-fold lower doses than conventional treatments. Dr Backx is a recognised expert in cardiac mechanics, heart failure and arrhythmias. His research focuses on the role of ion transport, ion channels and myocardial signalling in the initiation and progression of heart disease with a particular interest in atrial fibrillation. He holds a patent on tissue-specific drug delivery and has published over 190 peer-reviewed articles, many in the top tier journals like Cell, Nature, Nature Medicine, Journal of Clinical Investigation and Circulation Research.
