Everyone is trying to unlock Artificial Intelligence's promise. We have seen generative AI tricks that can summarize long documents in a flash, which is substantially different from the requirements of serious federal research. Today, we examined some challenges scientific communities experience in applying AI. Quentin Kerilman from the PNN labs throws some icy water on AI enthusiasts when he cautions that using AI for many sensitive applications has no framework. Leaders must use their best judgment when including data sets in projects. Ramesh Menon from the DIA warns that careful data collection must still be used. For example, one must use fair, unbiased data. Is the AI appropriately documented? Further, a scientist will always need multiple data modalities. When used in AI, it needs to be appropriately tagged. Medical data has much theoretical value from AI. But this is the exact data that has double and triple protection. What about leaks and backdoors? Despite all the challenges presented, transparency, human oversight, and collaboration were emphasized to ensure AI's practical and responsible use.

This episode challenges the ludicrous narrative coming from the Trump camp--the asinine idea that the president has 'absolute immunity. We will discuss the deluded and tortured argument for 'absolute presidential immunity' from criminal prosecution, its dubious history, and its danger to democracy itself. In theory at its most extreme application, under this doctrine a president could call an opponent into the Oval Office, put a gun to their head, pull the trigger, then demand the janitor clean up the mess without ever being held criminally accountable. This is the spectre of absolute presidential immunity that Donald Trump is demanding, which the Supreme Court could allow. Abstract: Major depression is the most prevalent neuropsychiatric disorder in elderly population, affecting more than 20% individuals over 60 years old, especially women. In this age range, social isolation is a major risk factor for depression. While there is a significant positive association between social isolation and depression in the elderly population, the neurobiological basis of this association is complex and still poorly understood. Evidence from animal models and human studies indicates that neuroplasticity, especially that of limbic brain regions, is impaired in depression but, till date, scarce studies address this question in older population. In this regard, animal models devoid of human cultural connotations represent a crucial tool. In the present study, we investigated the impact of chronic isolation stress (CIS) and a subsequent resocialization period in aged male and female mice (~ 21 months-old), focusing our attention on affective symptoms and the plasticity of parvalbumin-expressing (PV+) neurons in the lateral/basolateral amygdala (LA/BLA). We found that CIS impaired affective behaviour and LA/BLA plasticity only in females. Specifically, CIS induced depressive-like symptoms and decreased the integrity of perineuronal nets (PNN). Resocialization effectively rescued all these impairments. Old males were not affected by CIS but in social conditions showed higher PNN integrity (less plasticity) than females. All together, our results demonstrate that old females are less resilient to CIS than old males and point to the integrity of PNN in the LA/BLA as a key regulator of depressive-like symptoms induced by social isolation. This week on PNN, I will discuss the systemic child abuse and child reckless endangerment on the Texas border along the Rio Grande River, sponsored by Governor Greg Abbott. Governor Abbott ordered DPS (Texas Department of Public Safety) officers to push the small children and babies of migrants back into the treacherous waters of the Rio Grande River. He has also ordered troopers to deny small children any water in 100+ degree temperatures. Besides being an obvious and egregious human rights violation, Governor Abbott's orders could be interpreted as child abuse and reckless child endangerment. In spite of this obvious abuse, social workers in Texas--do NOTHING. No adult should be granted the right to abuse any child, and that includes governors and presidents. This rapidly evolving story is our first segment. Our second segment deals with the revocation of laws forbidding abusive and dangerous child labor. The GOP of Trump has rushed to embrace child labor with no limitations. It is ironic that the GOP claims to be the "pro-life" party, while pushing such evil and inhumane policies. Abstract: Perineuronal nets (PNN), a specialized form of ECM, surround numerous neurons in the CNS and allow synaptic connectivity through holes in its structure. We hypothesize that PNNs serve as gatekeepers that guard and protect synaptic territory and thus may stabilize an engram circuit. We present high-resolution and 3D EM images of PNN-engulfed neurons showing that synapses occupy the PNN holes and that invasion of other cellular components is rare. PNN constituents are long-lived and can be eroded faster in an enriched environment, while synaptic proteins have a high turnover rate. Preventing PNN erosion using pharmacological inhibition of PNN-modifying proteases or MMP9 knockout mice allowed normal fear memory acquisition but diminished remote-memory stabilization, supporting the above hypothesis. Last week, a Christian elemntary school was attacked by a deranged shooter. Children and adults died, and practically on cue, Tucker Carlson launched a tirade against the trans community. Why? The shooter was a trans woman. Carlson took his gun rights crusade and marched straigth to the convenient conclusion for the GOP of Trump, namely the demand to deny gun rights to minority communities. After all, there's nothing good ol' Tucker Carlson, the ersatz "Prince of tacky t.v. dinners" likes more than to scapegoat minorities, all the while trying to sound like a reasonable person. ;Carlson reasoned that because the shooter was a trans woman, that the entire trans, (even the entire LGBTQ+ community), must be launching a violent crusade against White Christians. Of course, there's not a shred of evidence backing Carlson's moronic claim, but it's FOX after all, and evidence is tossed into the trash. Because of his diatribe, PNN did a deep dive into the history of gun rights vs denial of those rights to minorities and the evidence was damning. In fact, it is not hyperbole to call out Carlson and his supporters as a parallel to the early days of the Third Reich Nazi regime. Abstract: Neprilysin (Nep) is a membrane-bound zinc-dependent endopeptidase that cleaves a wide variety of small peptides in the body. In the brain, it has gained fame as an important endogenous degrader of amyloid beta peptide, responsible for the pathophysiology of Alzheimer's disease. Nep is expressed specifically by parvalbumin (PV)-expressing inhibitory neurons in the adult cortex, the maturation of which regulates critical period plasticity. Given that PV neuron soma and proximal dendrites are primary sites of perineuronal net (PNN) formation, we investigated the role that Nep plays in PNN and PV neuron maturation and cortical development in the mouse visual cortex, using mice whose Nep expression is constitutively knocked out (Nep KO mice). Nep expression is high in young wildtype mice (P10) and is downregulated rapidly throughout postnatal development. It is especially prominent in layer 5 where it is highly expressed by inhibitory neurons and also expressed at low levels by many excitatory neurons. Contrary to our hypothesis, Nep KO mice did not show an alteration in the relative density or gross morphology of PNNs. Instead, Nep KO mice showed reduced maximal activation of L5 after white matter stimulation and decreased number of inhibitory neurons in L4. Abstract: Perineuronal nets (PNNs) surround specific neurons in the brain and are involved in various forms of plasticity and clinical conditions. However, our understanding of the PNN role in these phenomena is limited by the lack of highly quantitative maps of PNN distribution and association with specific cell types. Here, we present the first comprehensive atlas of PNN distribution (in Allen Brain Atlas coordinates) and colocalization with parvalbumin (PV) cells for over 600 regions of the adult mouse brain. Data analysis showed that PV expression is a good predictor of PNN aggregation. In the cortex, PNNs are dramatically enriched in layer 4 of all primary sensory areas in correlation with thalamocortical input density, and their distribution mirrors intracortical connectivity patterns. Gene expression analysis identified many PNN correlated genes. Strikingly, PNN anticorrelated transcripts were enriched in synaptic plasticity genes, generalizing PNN role as circuit stability factors. Overall, this atlas offers novel resources for understanding the organizational principles of the brain extracellular matrix. Copy rights belong to original authors. This week on PNN, I am talking about the GOP defamation wars. The most recent target has been US Congresswoman Katie Porter. A republican colleague, Congressman Ronnie Jackson not only accused Porter of making excuses for pedophiles, but he accused all progressives of the same in a tweet. Jackson is joined by the equally infamous creator of the Libs of TikTok, Chaya Raichik. Keep in mind, neither Ronnie Jackson nor Chaya Raichik have a single shred of evidence to back their defamatory accusations, but they based those accusations on spliced sections of actual congressional testimony being collected by Katie Porter. This incidence is merely the latest incarnation of the GOP War on Truth, using a propaganda machine that routinely slanders, libels and defames. The time is long overdue to start suing these purveyor s of lies. This is our big story.  Our second story deals with the slow pace of criminal prosecutions against the organizers of the Jan. 6th insurrection. Yet another republican, Congressman Ralph Norman, was caught having sent texts to Mark Meadows demanding Trump implement Martial Law. Professor Lee has made a special study of the impact right-wing groups have had on politics and academia, delivered with humor and wit.  I will discuss the dishonest judicial scam aka 'originalism.' I will cite the work of noted constitutional scholar and Dean of the UCLA Law School, Erwin Chemerinski as documentation. To put it bluntly, 'originalism' is the tool of choice for conservatives determined to limit human rights to a certain subset, namely "white, Christian, straight males.' There is no other logical outcome if you follow the idea of 'originalism' to its pre-ordained conclusion. Trump stealing national security records is far more serious than the Watergate case. Trump placed our national security in dire jeopardy at an incalculable risk. That's story #1. Story #2 deals with the very law which enabled the Flint water crisis, namely the Emergency Manager Act. This 'law' basically places an unnofficial poll tax on lower income munincipalities. It says that if your jurisdiction is deemed "too poor" by the state, your local elections can and WILL be overturned by the state. Abstract: Stroke remains one of leading causes of long-term disability worldwide, and the development of effective restorative therapies is hindered by an incomplete understanding of intrinsic recovery mechanisms in the brain. Here, we explored the involvement of perineuronal nets (PNNs), the facet-like layers of extracellular matrix surrounding fast-spiking interneurons, in brain remodeling and neurological recovery after focal cerebral ischemia in mice with and without induced stroke tolerance. Due to insufficient resolution of conventional microscopy methods, the contribution of ultrastructural changes in PNNs to post stroke brain plasticity remained unknown. Using superresolution stimulated emission depletion (STED) and structured illumination (SR-SIM) microscopy, we revealed that PNN facets become larger and less dense in the post-acute stroke phase. Morphological alterations in PNNs are likely mediated by activated microglia cells, which preferentially enwrap the PNN-coated neurons. The loosening of PNNs is transient and associates with the increased number of GABAergic axonal terminals on inhibitory interneurons in the motor cortical layer 5. The coherent remodeling of PNNs and inhibitory synapses precedes the recovery of motor coordination during the late post-acute stroke phase. In the chronic stroke phase, the initial morphology of PNNs is restored, and the number of GABAergic axonal terminals on motor cortical interneurons is reduced. Our data suggests a novel mechanism of motor cortical plasticity after stroke, and we propose that prolonging PNN loosening during the post-acute period can extend the opening neuroplasticity window into the chronic stroke phase. Abstract: Tau aggregates are a hallmark of multiple neurodegenerative diseases and can contain RNAs and RNA binding proteins, including SRRM2 and PNN. How these resident nuclear proteins mislocalize and their influence on the prion like propagation of tau fibers remains unknown. We demonstrate that polyserine repeats in SRRM2 and PNN are necessary and sufficient for recruitment to tau aggregates. Moreover, we demonstrate tau fibers preferentially grow in association with endogenous cytoplasmic assemblies, mitotic interchromatin granules and cytoplasmic speckles, which contain SRRM2 and PNN. Polyserine undergoes self assembly in vitro and in cells, where polyserine-assemblies are sites of tau fiber propagation. Modulating the levels of polyserine containing proteins results in a corresponding change in tau aggregation. These findings define a specific protein motif, and cellular condensates, that promote tau fiber propagation. As cytoplasmic speckles form in iPSC neurons under inflammatory or hyperosmolar stress, they may promote tau fiber propagation in various neurodegenerative diseases. E., Castren, E., Umemori, J. Abstract: Critical period-like plasticity (iPlasticity) can be reinstated in the adult brain by several interventions, including drugs and optogenetic modifications. We have demonstrated that a combination of iPlasticity with optimal training improves behaviors related to neuropsychiatric disorders. In this context, the activation of TrkB, a receptor for BDNF, in Parvalbumin positive (PV+) interneurons has a pivotal role in cortical network changes. However, it is unknown if the activation of TrkB in PV+ interneurons is important for other plasticity-related behaviors, especially for learning and memory. Here, using mice with heterozygous conditional TrkB deletion in PV+ interneurons (PV-TrkB hCKO) in Intellicage and fear erasure paradigms, we show that chronic treatment with fluoxetine, a widely prescribed antidepressant drug that is known to promote the activation of TrkB, enhances behavioral flexibility in spatial and fear memory, largely depending on the expression of the TrkB receptor in PV+ interneurons. In addition, hippocampal long-term potentiation (LTP) was enhanced by chronic treatment with fluoxetine in wild-type mice, but not in PV-TrkB hCKO mice. Transcriptomic analysis of PV+ interneurons after fluoxetine treatment indicated intrinsic changes in synaptic formation and downregulation of enzymes involved in perineuronal net (PNN) formation. Consistently, immunohistochemistry has shown that the fluoxetine treatment alters PV expression and reduces PNNs in PV+ interneurons, and here we show that TrkB expression in PV+ interneurons is required for these effects. Together, our results provide molecular and network mechanisms for the induction of critical period-like plasticity in adulthood. Copy rights belong to original authors. Abstract: Cyclin-dependent kinase-like 5 (CDKL5) deficiency disorder is an X-linked neurodevelopmental disorder, where children exhibit motor, speech, cognitive and visual deficits during development as well as early onset of seizures and infantile spasms. CDKL5 disorder results from de novo mutations in CDKL5, which phosphorylates proteins including MECP2, an epigenetic regulator of critical period plasticity. How mutations in CDKL5 gene result in these neurological deficits is unknown. Using a male CDKL5-null mouse model, we systematically characterize the expression of perineuronal nets, which are extracellular matrix structures surrounding parvalbumin+ GABAergic neurons in the primary visual cortex. Perineuronal nets (PNNs) are considered to be barriers to synaptic plasticity, particularly at the end of critical period plasticity in binocular zone. In CDKL5-null male visual cortex, here we report an increase in high-intensity PNN expression in the binocular zone, a decrease in overall PNN expression in the monocular zone, and no differences in PNN expression in rostral V1. These results suggest that at the closure of the visual critical period plasticity, subregion-specific variations in PNN expression lead to functional differences in synaptic plasticity and ultimately, changes in visual function in male CDKL5-null mice. Additionally, the systematic characterization of subregion-specific expression patterns of perineuronal nets lay the groundwork for further studies to probe the roles for perineuronal nets beyond the binocular zone of the mouse primary visual cortex. Today, the Supreme Court handed down a decision in W. Virginia v. EPA, which limits the authority of the EPA to regulate environmental toxins and other forms of pollution. This same 'pro-life' SCOTUS, has handed down a death penalty to the planet. I will discuss the future ramifications of this decision. I will also discuss why Supreme Court justices must be forced to follow a code of ethics. For example, Amy Coney Barrett's father was a corporate attorney for the fossil fuel industry and remains a lobbyist for the same industry. She should have been forced to recuse herself from this case. These are the regulations that help to safeguard our food supply and ensure that the medical people treating you are properly credentialed. These are the regulations that reversed the savage corporate abuses seen in the Sinclair Lewis classic, The Jungle. EJR with Jeanine Molloff - How Spire is attempting to extort $ over a pipeline.

This week, I am repeating part of a show that ran this past Sunday on PNN. The show deals with how Spire, one of the largest natural gas suppliers has been causing a panic in St. Louis (where Spire is based), over a 65 mile stretch of pipeline. The STL or St. Louis Pipeline as it has been dubbed had its certificate yanked by a DC court, over claims that the FERC assessment and certification was not in accordance with the actual LAW. The EDF (Environmental Defense Fund) brought the lawsuit against Spire, requesting that FERC do its job in accordance with the Natural Gas Act.  Spire's response was to send thinly veiled threatening emails and using local media as propagandists, claiming that without the 65-mile pipeline--there will be power outages as early as December 13. That is NOT totally true. FERC can extend the temporary permit and will most likely do so. EDF just wants to make sure that Spire isn't allowed to profit from their illegal move. During this investigation, I read from the court document, and found that Spire isolated St. Louis from any other pipelines, rendering us totally dependent on this single pipeline. Spire 'retired' the other pipelines which connected St. Louis to heating gas. EDF is ok with another temporary permit extending through the Winter, as long as Spire doesn't profit from this illegal action.  While investigating this story and making calls to Spire's Public Affairs Director Jason Merrill or it's CEO Suzanne Sitherwood, I must have stepped on some politically powerful toes, because I was 'visited' by 3 detectives from the Intelligence Unit of the St. Louis Metropolitan Police Department. And no--I'M NOT KIDDING.  This was obviously an intimidation tactic. They claimed that Spire filed a complaint against me--FOR MAKING A PHONE CALL.  Come listen.  Jeanine
