Unique proteomic signature of JCPyV-infected human astrocytes: from cells to extracellular vesicles

C., Sapozhnik, A., Richetin, K., Goelz, S., Du Pasquier, R. Abstract: JC polyomavirus (JCPyV) is the causative agent of progressive multifocal leukoencephalopathy (PML), a severe, and often fatal, demyelinating disease of the central nervous system. While PML has traditionally been characterized as a lytic infection of oligodendrocytes, more recent findings suggest an important role for astrocytes during the initial stages of disease. Here, using human induced pluripotent stem cell-derived astrocytes coupled with a multiparametric approach, we show that astrocytes are readily infected with JCPyV that strongly impacts their biology, inducing a unique proteomic signature. Furthermore, the changes observed mirror elements of ex vivo findings in PML brain lesions, including dysregulation of the cell cycle and activation of the DNA damage response. We also demonstrate that this signature is extended to extracellular vesicles (EVs) while being strikingly different from that of inflammatory conditions. Thus, our results support the relevance of using brain-derived EVs as a means to gain mechanistic insights into JCPyV pathophysiology in the brain and pave the way towards the identification of new biomarkers for the early stratification of patients at risk of developing PML. On Part 1 of our Mental Health and Wellness Series, Greg & Gary Goelz join me to discuss their new and very unconventional book What's In Your CORE: An Educator's Guide to Plugging Into Purpose & Perspective that aims to get us to slow down, reflect on our own "WHY" and then return to it so we can feel fulfilled and have fun in our jobs again. The mental health and wellness of educators could not be more important than RIGHT NOW! In these trying times, our students need us energized and emotionally healthy more than ever. However, many of us have understandably lost our way and are feeling ineffective and burned out. And, it's only November!!! Their book, "What's In Your CORE: An Educator's Guide to Plugging Into Purpose & Perspective" is a light and humorous book written for all K-12 educators and it has a very strange goal: it doesn't want to teach you anything new. What!?!? That's right, it's a book that aims to help you return to all of the wonderful already inside of you, rather than trying to teach you to be something, or someone, different. Refreshing, right? "What's in Your CORE?" focuses on two main topics: Purpose and Perspective. In regards to purpose, you'll be introduced to the 10 COREs (Calling or Reason in Education) that can best be thought of as your "educational personality type." Greg and Gary guide you to rediscover your own personal "WHY," your CORE, and how you can return to it. For perspective, the book explains how and why to fully get to know, trust, and lean into the COREs of your colleagues. In the book, they refer to this as the "unifying power of perspective-taking" and it's the engine that drives school-wide success. And, while you're learning about purpose and perspective, Greg and Gary will entertain you with funny personal stories from growing up as twins outside of Milwaukee, Wisconsin, and their interesting professional stories, too. Great for a book study, "What's In Your CORE?" will not disappoint. So, if you're looking for a light, hilarious read that will help remind you why you got into education in the first place, or why on earth you should stay, this is the perfect book for you. Having your world shaken up and become so unsafe at a very early age completely changes your whole world view, that what you mostly do in this lifetime is to try to find safety and a way to feel safe on this earth. Registered Somatic Movement therapist and educator Donnalea Goelz was molested twice, at the age of three and then at the age of five, which made her gravitate toward studying things that will help the issue of trauma. She brings together a lifetime of training in various healing modalities and therapies and is currently involved in research projects about somatic healing which is healing through movement and understanding the brain. Donnalea marries her love of understanding the scientific aspects through research with experiential somatic work. As a dancer, movement has always been a significant part of her life and her healing. Donnalea says movement is how she came to safety. She would go out and dance her prayers. Goblin King! Goblin King! Wherever you may be - take this child of mine far away from me! Join NostalgiaCast for a lively discussion on LABYRINTH, Jim Henson's musical fantasy adventure beloved by millions... of prepubescent pre-teen girls, while alienating everyone else unequipped to handle its not-so-subliminal sexual subtext. Author R.J. Craddock (THE THIRTEEN TRIBES OF CAIN book series) drops by the studio to school Jonny and Darin in all things Door Knockers, Fireys, and fairies, and helps makes sense of the difference between boy fantasy adventures and girl fantasy adventures. But will it be enough to rescue the film from the Bog of Eternal Stench? Or do the countless Muppet characters - and David Bowie's androgynous (and erotically-charged) rock star persona - have no power over the rest of us? Enter the labyrinth and find out!   Rent/stream LABYRINTH on Amazon, CinemaNow, Google Play, or Vudu today!

This week, Team Mousetalgia talks about some famous Disney dragons, both old and new, in an Emporium show. But first, Jeff visits the Walt Disney Family Museum to hear Muppeteer Dave Goelz speak about his career as the Great Gonzo and describe how he brings puppets to life. Next, special guest Rebecca Dimmick joins Mousetalgia to report on D23's recent event at the Walt Disney Studio celebrating the 75th anniversary of The Reluctant Dragon, the movie that introduced and demonstrated the process of Walt Disney's feature animation to the world. Rebecca describes the panels, the decorations, the events, and the transformation of the studio lot back to its earliest days in 1941. We also host a giveaway in honor of D23's latest Fanniversary celebration, and then it's an app check: Kristen reviews Dragon Spotting, Disney's app promoting the release of Pete's Dragon, and Disney Emoji Blast - two recent releases for mobile devices. Finally, Kristen offers an Empath's review of Pete's Dragon and explains why it should never have been made. Plus - now that the Rivers of America are under renovation, we offer Disney our unsolicited and, frankly, ridiculous advice for ways to plus the area.
